    Development of Sybr Green I-Based Melting Curve Method for HER2I655V Polymorphism Detection in Breast Cancer

    Background: Currently available molecular method to detect HER2I655V polymorphism such as PCR-RFLP is hampered by the costly experimental method, and post-PCR treatment requirement that makes this technique is not meeting for high-throughput analysis purpose. In this study, we developed an accurate, simple, low cost and rapid test to detect polymorphism at HER2 gene using SBR Green I based-melting curve method. Methods: Two forward allele-specific primers and one common reverse primer were used then these primers were tested to discriminate known genotypes of genomic templates (GG type or AA type) and genomic samples retrieved from breast cancer patients. Results: Melting curve analysis derived from SYBR Green I-based allele-specific PCR with defined primers concentration and annealing temperature at 54.3 °C showed good discrimination level of Tm peaks in which GG genotype melted at 89 °C slightly higher than AA genotype which melted at 86 °C, while AG genotype harbored both of homozygous Tm characteristics. Conclusions: This preliminary result will be as basic for further large-scale typing of HER2I655V polymorphism.&nbsp

    The research was conducted due to the fluctuating performance data in PT. TASPEN (Persero) KCU Semarang in the last three years. The occurrence of the up and down performance data the company identified that motivation and work environments that exist in the company during this less noticed by the company. The purpose of this research were to determine the influence of motivation adn work environment agains employee performance and job satisfaction. The Sampling techniques used sampling jenuh means the entirety of the employees be made respondents. The respondents in this research totalled 53 respondents. This research was also using Path analysis techniques supported by SPSS software version 20.3. The validity test, reliability test, coefficient of correlation test, linear and multiple regression, coefficient of determination test, and significance tests (t and F test) with Sobel method beforehand. The results has showed that the independent variable in this study had a variable affect performance motivation through job satisfaction has no relationship mediation, and work environment that affect performance through job satisfaction had partial mediation

    Kualitas Telur Puyuh yang Diberikan Ransum dengan Penambahan Silase Tepung Daun Ubi Kayu

    Telur adalah produk unggas yang mempunyai nilai gizi tinggi dan mudah dicerna.  Penelitian bertujuan mengetahui pengaruh penggunaan silase tepung daun ubi kayu dalam ransum berbentuk mash terhadap kualitas telur puyuh.  Penelitian menggunakan metode Eksperimen dengan Rancangan Acak Lengkap (RAL) terdiri dari: 4 perlakuan dan 4 kali ulangan, dan setiap ulangan terdiri dari 4 ekor burung  puyuh. Perlakuan yang diberikan yaitu T0 (Ransum + silase tepung daun ubi kayu 0%), T1 (Ransum + silase tepung daun ubi kayu 3%), T2 (Ransum + silase tepung daun ubi kayu 6%), T3 (Ransum + silase tepung daun ubi kayu 9%). Parameter yang diamati indeks putih telur, indeks kuning telur, bobot telur, dan nilai haugh unit. Hasil  penelitian ini menunjukkan bahwa penggunaan silase tepung daun ubi kayu dalam formulasi burung puyuh sampai 9% belum mampu meningkatkan indeks putih telur, indeks kuning telur dan  nilai haugh unit tetapi dapat meningkatkan bobot telur puyuh. Perlakuan terbaik pada taraf 3% karena dapat meningkatkan indeks putih telur, indeks kuning telur, dan nilai haugh unit

    Ikan biang (Ilisha elongata) memiliki nilai konsumsi yang tinggi di Provinsi Riau. Di Indonesiaikan biang biasanya diolah menjadi ikan asin agar tahan lama. Metode penggaraman yang dilakukan adalahpenggaraman kering dengan kadar garam 5%, 10% dan 15% (b/b). Tujuan dari penelitian ini adalahmenentukan komposisi proksimat ikan biang asin kering dengan perbedaan konsentrasi garam.Pengujiandilakukan di Politeknik Kelautan dan Perikanan Dumai, Laboratorium karakteristik bahan baku hasilperikanan THP IPB dan Laboratorium Kimia Terpadu IPB. Hasil penelitian menunjukkan kadar air ikan biangasin kering berkisar 14.09% - 18,71%. kadar abu berkisar 22.20% - 24.02%, kadar protein sebesar 46.68% -49.08%, lemak sebesar 4.52% - 5.77%, dan karbohidrat 6.07% - 9.55%.     Genetic continuity across a deeply divergent linguistic contact zone in North Maluku, Indonesia

    <p>Abstract</p> <p>Background</p> <p>The islands of North Maluku, Indonesia occupy a central position in the major prehistoric dispersal streams that shaped the peoples of Island Southeast Asia and the Pacific. Within this region a linguistic contact zone exists where speakers of Papuan and Austronesian languages reside in close proximity. Here we use population genetic data to assess the extent to which North Maluku populations experienced admixture of Asian genetic material, and whether linguistic boundaries reflect genetic differentiation today.</p> <p>Results</p> <p>Autosomal and X-linked markers reveal overall Asian admixture of 67% in North Maluku, demonstrating a substantial contribution of genetic material into the region from Asia. We observe no evidence of population structure associated with ethnicity or language affiliation.</p> <p>Conclusions</p> <p>Our data support a model of widespread Asian admixture in North Maluku, likely mediated by the expansion of Austronesian-speaking peoples into the region during the mid Holocene. In North Maluku there is no genetic differentiation in terms of Austronesian- versus Papuan-speakers, suggesting extensive gene flow across linguistic boundaries. In a regional context, our results illuminate a major genetic divide at the Molucca Sea, between the islands of Sulawesi and North Maluku. West of this divide, populations exhibit predominantly Asian ancestry, with very little contribution of Papuan genetic material. East of the Molucca Sea, populations show diminished rates of Asian admixture and substantial persistence of Papuan genetic diversity.</p

    Bioactive Compounds of Rambutan (Nephelium lappaceum L.)

    Rambutan, a widely popular tropical fruit encompasses rich amount of bioactive compounds. All parts of this plant (leaves, bark, root, fruits, fruit skin, pulp and seeds) finds traditional usage, and are linked with high therapeutic values. Rambutan fruits parts like that of peel, pulp and seeds have been scientifically investigated in-depth and is reported to encompass high amounts of bioactive compounds (such as polyphenol, flavonoid, alkaloid, essential mineral, dietary fiber). These compounds contribute towards antioxidant, antimicrobial, anticancer, antidiabetic and anti-obesity activities. However, literature pertaining towards potential industrial applications (food, cosmetics, pharmaceutical) of rambutan fruits are limited. In the present chapter, it is intended to document some of the interesting research themes published on rambutan fruits, and identify the existing gaps to open up arena for future research work.This chapter theme is based on our ongoing project—VALORTECH, which has received funding from the European Union’s Horizon 2020 research and innovation program under grant agreement No 810630
