    Stem cell labeling using polyethylenimine conjugated (alpha-NaYbF4:Tm3+)/CaF2 upconversion nanoparticles

    We report on a polyethylenimine (PEI) covalently conjugated (alpha-NaYbF4:Tm3+)/CaF2 upconversion nanoparticle (PEI-UCNP) and its use for labeling rat mesenchymal stem cells (rMSCs). The PEI-UCNPs absorb and emit near-infrared light, allowing for improved in vivo imaging depth over conventional probes. We found that such covalent surface conjugation by PEI results in a much more stable PEI-UCNP suspension in PBS compared to conventional electrostatic layer by layer (LbL) self-assembling coating approach. We systematically examined the effects of nanoparticle dose and exposure time on rat mesenchymal stem cell (rMSC) cytotoxicity. The exocytosis of PEI-UCNPs from labeled rMSCs and the impact of PEI-UCNP uptake on rMSC differentiation was also investigated. Our data show that incubation of 100-microg/mL PEI-UCNPs with rMSCs for 4 h led to efficient labeling of the MSCs, and such a level of PEI-UCNP exposure imposed little cytotoxicity to rMSCs (95% viability). However, extended incubation of PEI-UCNPs at the 100 microg/mL dose for 24 hour resulted in some cytotoxicity to rMSCs (60% viability). PEI-UCNP labeled rMSCs also exhibited normal early proliferation, and the internalized PEI-UCNPs did not leak out to cause unintended labeling of adjacent cells during a 14-day transwell culture experiment. Finally, PEI-UCNP labeled rMSCs were able to undergo osteogenic and adipogenic differentiation upon in vitro induction, although the osteogenesis of labeled rMSCs appeared to be less potent than that of the unlabeled rMSCs.     Genetic Variants in WNT2B and BTRC Predict Melanoma Survival

    Cutaneous melanoma (CM) is the most lethal skin cancer. The Wnt pathway has an impact on development, invasion and metastasis of CM, thus likely affecting CM prognosis. Using data from a published genome-wide association study (GWAS) from The University of Texas M.D. Anderson Cancer Center, we assessed the associations of 19,830 common single-nucleotide polymorphisms (SNPs) in 151 Wnt pathway autosomal genes with CM-specific survival (CMSS) and then validated significant SNPs in another GWAS from Harvard University. In the single-locus analysis, 1,855 SNPs were significantly associated with CMSS at P T and BTRC rs61873997 G>A) that showed a predictive role in CMSS, with an effect-allele-attributed hazards ratio [adjHR of 1.99 (95% confidence interval (CI) = 1.41-2.81, P = 8.10E-05) and 0.61 (0.46-0.80, 3.12E-04), respectively]. Collectively, these variants in the Wnt pathway genes may be biomarkers for outcomes of CM patients, if validated by larger studies

    Dose-related liver injury of Geniposide associated with the alteration in bile acid synthesis and transportation.

    Fructus Gardenia (FG), containing the major active constituent Geniposide, is widely used in China for medicinal purposes. Currently, clinical reports of FG toxicity have not been published, however, animal studies have shown FG or Geniposide can cause hepatotoxicity in rats. We investigated Geniposide-induced hepatic injury in male Sprague-Dawley rats after 3-day intragastric administration of 100 mg/kg or 300 mg/kg Geniposide. Changes in hepatic histomorphology, serum liver enzyme, serum and hepatic bile acid profiles, and hepatic bile acid synthesis and transportation gene expression were measured. The 300 mg/kg Geniposide caused liver injury evidenced by pathological changes and increases in serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and γ-glutamytransferase (γ-GT). While liver, but not sera, total bile acids (TBAs) were increased 75% by this dose, dominated by increases in taurine-conjugated bile acids (t-CBAs). The 300 mg/kg Geniposide also down-regulated expression of Farnesoid X receptor (FXR), small heterodimer partner (SHP) and bile salt export pump (BSEP). In conclusion, 300 mg/kg Geniposide can induce liver injury with associated changes in bile acid regulating genes, leading to an accumulation of taurine conjugates in the rat liver. Taurocholic acid (TCA), taurochenodeoxycholic acid (TCDCA) as well as tauro-α-muricholic acid (T-α-MCA) are potential markers for Geniposide-induced hepatic damage

    In this study, a novel 96-well microplate swivel system (M96SS) was built for high-throughput screening of microalgal strains for CO2 fixation. Cell growth under different CO2 supply conditions (0.2, 0.4, 0.8, and 1.2 g L−1 d−1), residual nitrate, and pH value of Chlorella sp. SJTU-3, Chlorella pyrenoidosa SJTU-2, and Scenedesmus obliquus SJTU-3 were examined in the M96SS and traditional flask cultures. The dynamic data showed there was a good agreement between the systems. Two critical problems in miniature culture systems (intra-well mixing and evaporation loss) were improved by sealed vertical mixing of the M96SS. A sample screen of six microalgal species (Chlorella sp. SJTU-3, Chlorella pyrenoidosa SJTU-2, Selenastrum capricornutum, Scenedesmus obliquus SJTU-3, Chlamydomonas sajao, Dunaliella primolecta) was carried out in flasks and the M96SS. Chlamydomonas sajao appeared to be a robust performer (highest cell density: 1.437 g L−1) in anaerobic pond water with 0.8, and 1.2 g L−1 d−1 CO2.     Asymmetric Effect of Media Tone on IPO Underpricing and Volatility

Because of asymmetric information between issuing companies and investors, media coverage plays an important role in conveying information to investors during an initial public offering (IPO). The stock price is affected by a large amount of information released to the public through media coverage. Using a comprehensive sample of 1,075 IPOs on China’s stock market from 2009 to 2016, this paper conducts a textual analysis to determine the tone of media coverage and examines the relationship between media tone and IPO underpricing as well as post-IPO volatility. The empirical results show that media coverage during an IPO is significantly negatively associated with IPO underpricing, which confirms our hypothesis that the tone of media coverage reduces the degree of information asymmetry between investors and issuers, regardless of whether the tone is positive or negative. Consistent with prospect theory, investors are more sensitive to media coverage with a negative tone

    Impact of Climate Change on the Yield and Water Footprint of Winter Wheat in the Haihe River Basin, China

    Climate change can impact the yield and water footprint of crops. Therefore, assessing such impacts carries great significance for regional water and food security. This study validated and verified the variety parameters of winter wheat for the Decision Support System for Agrotechnology Transfer (DSSAT) model, using the long-term (1993–2013) growth and yield data observed from six agricultural experiment stations in the Haihe River Basin (HRB), China. The growth process was simulated under three representative concentration pathways (RCPs), named RCP2.6, RCP4.5, and RCP8.5—climate scenarios driven by the HadGEM2-ES model. The variety parameters of winter wheat showed high accuracy in the simulation of the anthesis and maturity dates, and could be used for long-term prediction of the growth process. The trends of climate change had positive impacts on the water footprint of winter wheat but adverse impacts on the yield. The growing period was shortened by 3.6 days, 4.7 days, and 5.0 days per decade in the RCP2.6, RCP4.5, and RCP8.5 scenarios, respectively, due to the rapid accumulation of heat. The yield would be increased in lower emissions scenarios (17% in RCP2.6), but decreased in high-emissions scenarios due to high temperatures, which may restrict the growth of wheat. The water footprint was decreased by 10%, 11%, and 13% in the RCP2.6, RCP4.5, and RCP8.5 scenarios, respectively, indicating that the water-use efficiency could be improved in the future. The results showed broad application prospects of the DSSAT model in simulating the response of crop growth to climate change

    Conventional geostress measurement methods are limited by deficiencies including the measurable depth, the complexity, and the long duration of operation. To address these problems and achieve the measurement of geostress in deep wells under conditions of complex high pressures and high temperatures, we propose a new measurement method for geostress based on an integrated drilling and optical microscopy system. Its innovative integrated structure eliminates the problems associated with complex procedures and depth limits, and avoids rock creep caused by long delays, significantly improving the accuracy and range of the measurements. It works by using microscopic imaging and direct contact probes to capture the changes of a borehole’s cross-sectional outlines before and after stress relief. The resulting images are analyzed with search circles to obtain the positions of probe apices, which can be fitted into ellipses that describe the outlines, and calculate the state of the stress. The validity and accuracy of the method was verified by in-door tests and field applications in the ZK1 borehole. The results show that: (1) the integrated system can be used to measure micrometer-grade deformations(2) the searchcircle approach can accurately obtain the positions of probe apicesand (3) the stress measurement method based on the system is accurate and feasible.Konvencionalne metode merjenja geostatičnih napetosti so omejene zaradi pomanjkljivosti, vključno z izmerljivo globino, kompleksnostjo in dolgim trajanjem izvedbe. Da bi rešili te probleme in dosegli merjenje geostatičnih napetosti v globokih vodnjakih v pogojih kompleksnih visokih tlakov in visokih temperatur, je predlagana nova merilna metoda za geostatične napetosti na osnovi integriranega sistema vrtanja in optičnega mikroskopa. Njena inovativna integrirana konstrukcija odpravlja probleme, povezane s kompleksnimi postopki in globinskimi omejitvami, ter se izogiba lezenju kamnin zaradi dolgotrajnosti postopka, kar bistveno izboljša natančnost in obseg meritev. Deluje s pomočjo mikroskopskega slikanja in neposrednimi sondami, da zajame spremembe zunanje površine prereza vrtine pred in po zmanjšanju napetosti. Rezultirajoče slike se analizirajo z iskalnimi krogi, da se pridobijo položaji vrhov sond, ki so lahko vstavljeni v elipse, ki opisujejo obrise, in izračunajo stanje napetosti. Veljavnost in natančnost metode sta bili preverjeni z laboratorijskimi preizkusi in aplikacijami na terenu v vrtini ZK1.
