    This study intends to explore the meaning of the Logu Senhor procession for the faith of the people in St. Parish. Ignatius Loyola Sikka. This is based on the results of initial observations which show that there are parishioners in this parish who faithfully follow the Logu Senhor procession but do not understand what the Logu Senhor procession means. This study uses a qualitative method with a descriptive model. Data collection techniques were carried out using interviews and documentation studies. Technical analysis of data includes data collection, data reduction, data presentation, and conclusion drawing. The results of the study explain that the Logu Senhor procession is an annual procession carried out on Good Friday to commemorate the story of the passion of Jesus Christ. With this procession, the people will develop towards maturity in faith, especially in carrying the cross in various forms in their lives in the midst of family, community, church, and nation

    Whole-exome sequencing combined with functional genomics reveals novel candidate driver cancer genes in endometrial cancer

    Endometrial cancer is the most common gynecological malignancy, with more than 280,000 cases occurring annually worldwide. Although previous studies have identified important common somatic mutations in endometrial cancer, they have primarily focused on a small set of known cancer genes and have thus provided a limited view of the molecular basis underlying this disease. Here we have developed an integrated systems-biology approach to identifying novel cancer genes contributing to endometrial tumorigenesis. We first performed whole-exome sequencing on 13 endometrial cancers and matched normal samples, systematically identifying somatic alterations with high precision and sensitivity. We then combined bioinformatics prioritization with high-throughput screening (including both shRNA-mediated knockdown and expression of wild-type and mutant constructs) in a highly sensitive cell viability assay. Our results revealed 12 potential driver cancer genes including 10 tumor-suppressor candidates (ARID1A, INHBA, KMO, TTLL5, GRM8, IGFBP3, AKTIP, PHKA2, TRPS1, and WNT11) and two oncogene candidates (ERBB3 and RPS6KC1). The results in the ''sensor'' cell line were recapitulated by siRNA-mediated knockdown in endometrial cancer cell lines. Focusing on ARID1A, we integrated mutation profiles with functional proteomics in 222 endometrial cancer samples, demonstrating that ARID1A mutations frequently co-occur with mutations in the phosphatidylinositol 3-kinase (PI3K) pathway and are associated with PI3K pathway activation. siRNA knockdown in endometrial cancer cell lines increased AKT phosphorylation supporting ARID1A as a novel regulator of PI3K pathway activity. Our study presents the first unbiased view of somatic coding mutations in endometrial cancer and provides functional evidence for diverse driver genes and mutations in this disease. © 2012, Published by Cold Spring Harbor Laboratory Press.Link_to_subscribed_fulltex

    Impact of opioid-free analgesia on pain severity and patient satisfaction after discharge from surgery: multispecialty, prospective cohort study in 25 countries

    Background: Balancing opioid stewardship and the need for adequate analgesia following discharge after surgery is challenging. This study aimed to compare the outcomes for patients discharged with opioid versus opioid-free analgesia after common surgical procedures.Methods: This international, multicentre, prospective cohort study collected data from patients undergoing common acute and elective general surgical, urological, gynaecological, and orthopaedic procedures. The primary outcomes were patient-reported time in severe pain measured on a numerical analogue scale from 0 to 100% and patient-reported satisfaction with pain relief during the first week following discharge. Data were collected by in-hospital chart review and patient telephone interview 1 week after discharge.Results: The study recruited 4273 patients from 144 centres in 25 countries; 1311 patients (30.7%) were prescribed opioid analgesia at discharge. Patients reported being in severe pain for 10 (i.q.r. 1-30)% of the first week after discharge and rated satisfaction with analgesia as 90 (i.q.r. 80-100) of 100. After adjustment for confounders, opioid analgesia on discharge was independently associated with increased pain severity (risk ratio 1.52, 95% c.i. 1.31 to 1.76; P < 0.001) and re-presentation to healthcare providers owing to side-effects of medication (OR 2.38, 95% c.i. 1.36 to 4.17; P = 0.004), but not with satisfaction with analgesia (beta coefficient 0.92, 95% c.i. -1.52 to 3.36; P = 0.468) compared with opioid-free analgesia. Although opioid prescribing varied greatly between high-income and low- and middle-income countries, patient-reported outcomes did not.Conclusion: Opioid analgesia prescription on surgical discharge is associated with a higher risk of re-presentation owing to side-effects of medication and increased patient-reported pain, but not with changes in patient-reported satisfaction. Opioid-free discharge analgesia should be adopted routinely

    Dynamic subsidence of Eastern Australia during the Cretaceous

    During the Early Cretaceous Australia's eastward passage over sinking subducted slabs induced widespread dynamic subsidence and formation of a large epeiric sea in the eastern interior. Despite evidence for convergence between Australia and the paleo-Pacific, the subduction zone location has been poorly constrained. Using coupled plate tectonic–mantle convection models, we test two end-member scenarios, one with subduction directly east of Australia's reconstructed continental margin, and a second with subduction translated ~ 1000 km east, implying the existence of a back-arc basin. Our models incorporate a rheological model for the mantle and lithosphere, plate motions since 140 Ma and evolving plate boundaries. While mantle rheology affects the magnitude of surface vertical motions, timing of uplift and subsidence depends on plate boundary geometries and kinematics. Computations with a proximal subduction zone result in accelerated basin subsidence occurring 20 Myr too early compared with tectonic subsidence calculated from well data. This timing offset is reconciled when subduction is shifted eastward. Comparisons between seismic tomography and model temperature cross-sections, and an absence of subduction zone volcanism in eastern Australia in the Early Cretaceous provide support for the back-arc basin scenario

    Enantioselective Synthesis of Pyrroloindolines via Noncovalent Stabilization of Indole Radical Cations and Applications to the Synthesis of Alkaloid Natural Products

    While interest in the synthetic chemistry of radical cations continues to grow, controlling enantioselectivity in the reactions of these intermediates remains a challenge. Based on recent insights into the oxidation of tryptophan in enzymatic systems, we report a photocatalytic method for the generation of indole radical cations as hydrogen-bonded adducts with chiral phosphate anions. These noncovalent open-shell complexes can be intercepted by the stable nitroxyl radical TEMPO· to form alkoxyamine-substituted pyrroloindolines with high levels of enantioselectivity. Further elaboration of these optically enriched adducts can be achieved via a catalytic single-electron oxidation/mesolytic cleavage sequence to furnish transient carbocation intermediates that may be intercepted by a wide range of nucleophiles. Taken together, this two-step sequence provides a simple catalytic method to access a wide range of substituted pyrroloindolines in enantioenriched form via a standard experimental protocol from a common synthetic intermediate. The design, development, mechanistic study, and scope of this process are presented, as are applications of this method to the synthesis of several dimeric pyrroloindoline natural products
