    Identification and Characterization of Genes Associated with \u3ci\u3eV-Jun\u3c/i\u3e Induced Cell Transformation

    The v-jun oncogene was initially identified as the causative agent for fibrosarcomas in chickens. Studies show that overexpression of v-Jun proteins transforms chicken embryo fibroblasts (CEF) in vitro, and forms tumors in chickens in vivo. The mechanisms for this are not clearly defined. Conceivably, overexpression of an unregulated transcription factor would cause cell transfonnation by illicit regulation of its target genes. In support of this, we show that in vivo v-Jun complexes exhibit differential binding to in vitro generated AP-1 and \u27AP-1 like\u27 target sequences, suggesting that the pattern of target gene expression is altered during cell transformation. With this in mind, we set out to identify genes associated with v-Jun induced cell transformation. We have isolated several clones by subtractive hybridization, and a modified differential display procedure. One of these is clone 4, showing strong sequence homology, both at nucleotide and amino acid level, to cysteine thiol proteases. Northern blot analysis shows that the steady state levels of clone 4 mRNA are 3 to 7 times higher in v-Jun transformed CEF (VJ-1), when compared to c-Jun overexpressing CEF (CJ-3), or normal CEF infected with vector sequences only (RCAS). Another is clone 15-15, showing strong sequence identity to the chicken Apolipoprotein A1 (ApoA1) gene. Northern blot analysis demonstrates that the steady state levels of ApoA1 mRNA in RCAS is 3 to 10 times higher than in VJ-1 cells, indicating that v-Jun might repress this gene by transcriptional mechanisms. To investigate this possibility, we generated several ApoA1 reporter CAT constructs containing 5\u27 deletions in the promotor, and tested them in VJ-1 and RCAS cells. Our findings suggest that three potential cis-acting sequences could regulate this promotor in normal RCAS CEF. Quite remarkably, none of these constructs were transcriptionally active in VJ-1 cells. DNA binding studies utilizing one of the potential cis-acting regions, suggests that a specific factor is present in normal nuclear extracts, but absent from v-Jun transformed extracts. This observation suggests that this specific factor may be a positive activator protein. In addition, actinomycin D studies demonstrate that the ApoA1 mRNA has a long half-life of up to 20 hours. We therefore propose that ApoA1 is positively regulated by at least three cis-acting sequences, and maintained at high steady state levels in normal CEF. Several possible mechanisms exist to explain ApoA1 repression in normal RCAS and VJ-1 cells. One possibility is the direct repressor mechanism, whereby a silencer region directly inhibits ApoA1 expression in normal cells. In VJ-1 cells however, a squelching mechanism could predominate.     Esta investigación de tipo descriptivo propositivo, se realiza debido al aumento en la prevalencia de trastornos mentales en los jóvenes universitarios en el Perú, siendo una de las poblaciones más vulnerables los estudiantes de Psicología quienes necesitan contar con una óptima salud mental. La presente tiene como objetivo principal elaborar el Programa “AS” sustentado en la Terapia Centrada en Esquemas de Jeffrey Young para disminuir Esquemas Maladaptativos Tempranos en estudiantes de la Escuela Profesional de Psicología de la Universidad Nacional Pedro Ruiz Gallo en Lambayeque. La muestra fue no probabilística intencional y estuvo conformada por 144 estudiantes universitarios de ambos sexos, cuyas edades oscilan entre 18 a 29 años, pertenecientes a cuatro ciclos académicos. La prueba que se utilizó fue el Cuestionario de esquemas de Young- Forma abreviada (YSQ-SF), aplicado de forma grupal y procesado con el programa SPSS 22. Los resultados mostraron que los esquemas más frecuentes en este grupo son: El esquema de Autocontrol Insuficiente con un 44.4%, Fracaso con un 38.2%, Abandono con un 36.8% e Inhibición Emocional con un 35.4%. Con ello se espera contribuir al desarrollo de la Psicología Clínica, brindando alcances teóricos y prácticos; y al desarrollo de nuevas investigaciones y elaboración de distintos programas de intervención que contribuyan al desarrollo de esquemas adaptativos con el fin de mejorar la salud mental en estudiantes universitarios.     Nectin-2 expression in testicular cells is controlled via the functional cooperation between transcription factors of the Sp1, CREB, and AP-1 families

    Nectin-2, a major protein component of the adherens junctions (AJs), is found between Sertoli cells and germ cells in the seminiferous epithelium. Recent studies have shown that the expression of nectin-2 gene in testis is crucial to maintain normal spermatogenesis since male knockout mice lacking nectin-2 gene are sterile and possess morphologically abnormal spermatozoa. However, the molecular mechanisms governing its basal transcription remain poorly understood. By the use of Sertoli and germ cell-lines (TM4 and GC-2spd(ts) cells, respectively) in transient transfection studies, we showed that the minimal mouse nectin-2 promoter was located between nucleotides -316 and -211 (relative to the translation start site). Two putative Sp1 motifs and one each of the CRE, AP1, and AP2 motifs were identified within this region. Mutational studies showed that these two Sp1 motifs cooperated synergistically with the CRE motif, but not the AP1 and AP2 motifs, to regulate nectin-2 gene transcription in both TM4 and GC-2spd(ts) cells. By EMSAs, we found that an AP-1 consensus sequence was able to inhibit DNA-protein complex formation with the CRE motif, suggesting an interaction between the AP-1 transcription factor (c-Jun) and CREB within the CRE motif. Overexpressions of CREB and c-Jun, but not c-Fos, also significantly increased the promoter activity, which suggests that CREB and c-Jun are the crucial transcription factors involved in regulating nectin-2 gene transcription. Chromatin immunoprecipitation assay has shown that, in vivo, CREB, c-Jun, and Sp1 family proteins are bound to the mouse nectin-2 promoter. Analysis of the staged tubules has confirmed that the cyclic expressions of CREB and nectin-2 coincide with the event of adherens junction restructuring between Sertoli cells and germ cells.     O objetivo do presente trabalho foi avaliar o efeito climático sobre respostas fisiológicas de vacas zebuínas em fase de lactação criadas em dois ambientes diferentes. Para a presente pesquisa foram avaliadas 20 vacas da raça Guzerá criadas em ambiente com sombreamento e sem sombreamento. Durante o período experimental, foram realizadas medições das variáveis ambientais diariamente: temperatura do ar, umidade relativa do ar e temperatura de globo negro, para calcular os valores de ITGU. Também foram avaliados parâmetros fisiológicos como a frequência respiratória e temperatura retal; e parâmetro produtivo, como a produção de leite. Para a análise da produção de leite, o mesmo foi pesado semanalmente. Foram observadas alterações fisiológicas nos animais avaliados, com redução na produção leiteira no ambiente sem sombreamento. O ITGU atingiu valores classificados como de estresse para os animais criados em ambiente sem sombreamento, mostrando a importância do ambiente de criação animal sobre o seu desempenho. Vacas da raça guzerá criadas em pasto sem sombreamento sofrem alterações fisiológicas que indicam estresse e afetam a produção de leite

O uso da mesa virtual 3D Anatomage tem como finalidade estimular e motivar os discentes, tornando as aulas de anatomia mais dinâmicas, assim como evitando os riscos á saúde, causado pelo uso do formaldeído para a conservação das peças anatômicas. Essa tecnologia possibilita a observação das peças de forma ilimitada com adição e remoção de coloração e estruturas, dissecação em todos os planos, além de possibilitar a fazer cortes transversais, horizontais, verticais e controlar a profundidade dos mesmos. Esta tecnologia também permite a observação de exames radiográficos, no qual pode ser cortado para uma melhor visualização, e assim obter uma avaliação de casos clínicos e de algumas patologias

This article presents a new architectural analysis of this building and attempts to set it within its immediate and wider archaeological and geological landscape context.     De nombreuses cardiopathies sont responsables de troubles du rythme ventriculaire ou de mort subite. La prévention de ces événements peut conduire à l'implantation d'un DAI chez les patients estimés à risque. L'identification de ces patients et les indications d'implantation du DAI sont régulièrement mises à jour. Nous avons mené un suivi chez les patients implantés d'un DAI sur une année, afin d'évaluer la prise en charge de ces patients de l'implantation du DAI et après 18 mois de suivi. L'objectif était d'évaluer le devenir de ces patients et la survenue de complications potentielles. On confirme ici les indications et la prise en charge adaptés de ces patients dans notre centre, conformes aux indications en vigueur. Les taux de complications répertoriés sont identiques voire inférieurs à ceux cités dans la littérature spécifique. Les thérapies appropriés sont nombreuses, preuve de la bonne sélection de ces patients à haut risque de mort subite. Les thérapies inappropriées ne sont pas négligeables. Elles nécessitent toujours une optimisation des traitements et de la programmation du boîtier pour éviter les récidives, toujours invalidantes pour le patient. Le DAI n'est donc pas un dispositif anodin. Il doit être proposé dans le cadre des recommandations.
