    Evaluation of gut modulatory and bronchodilator activities of Amaranthus spinosus Linn.

    Background The aqueous-methanolic extract of Amaranthus spinosus (A. spinosus Linn.,) whole plant, was studied for its laxative, spasmolytic and bronchodilator activities to validate some of its medicinal uses. Methods The crude extract of A. spinosus was studied in-vivo for bronchodilator and laxative activities and in-vitro using isolated tissue preparations which were mounted in tissue baths assembly containing physiological salt solutions, maintained at 37°C and aerated with carbogen, to assess the spasmolytic effect and to find out the possible underlying mechanisms. Results In the in-vivo experiments in mice, the administration of A. spinosus increased fecal output at doses of 100 and 300 mg/kg showing laxative activity. It also inhibited carbachol-induced bronchospasm in anesthetized rats at 1, 3, 10 and 30 mg/kg indicative of bronchodilator activity. When tested on isolated gut preparations, the plant extract showed a concentration-dependent (0.01-10.0 mg/ml) spasmogenic effect in spontaneously contracting rabbit jejunum and guinea-pig ileum. The spasmogenic effect was partially blocked in tissues pretreated with atropine (0.1 μM). When tested on K+ (80 mM)-induced sustained contractions in isolated rabbit jejunum, the plant extract caused complete relaxation and also produced a shift in the Ca++ concentration-response curves (CRCs) towards right, similar to diltiazem. In rabbit trachea, the plant extract completely inhibited K+ (80 mM) and carbachol (CCh, 1 μM)-induced contractions at 1 mg/ml but pretreatment of tissue with propranolol (1 μM), caused around 10 fold shift in the inhibitory CRCs of the plant extract constructed against CCh-induced contraction. The plant extract (up to 0.3 mg/ml) also increased both force and rate of spontaneous contractions of isolated guinea-pig atria, followed by relaxation at higher concentration (1.0-5.0 mg/ml). The cardio-stimulant effect was abolished in the presence of propranolol, similar to that of isoprenaline. Activity-directed fractionation revealed that the spasmolytic component(s) was separated in the organic fraction, whereas the spasmogenic component was concentrated in the aqueous fraction. Conclusion These results indicate that A. spinosus possesses laxative activity partially mediated through cholinergic action. The spasmolytic effect was mediated through calcium channel blocking (CCB), while bronchodilator activity through a combination of β-adrenergic and CCB pathways, which may explain the traditional uses of A. spinosus in gut and airways disorders

    Antibiotic Resistance Pattern and Molecular Epidemiology of Methicillin-Resistant Staphylococcus aureus Colonization in Burns Unit of a Tertiary Care Hospital in Peshawar, Pakistan

    Purpose: To develop strategies for the control of hospital-acquired methicillin-resistant Staphylococcus aureus (HA-MRSA) which is a serious threat to burns patients with the aid of molecular studies.Methods: Staphylococcus aureus strains were collected from the Burns Unit of Khyber Teaching Hospital (KTH) Peshawar, Pakistan from July - December 2011. Antibiotic resistance was determined according to the recommendations of Clinical Laboratory Standard Institute (CLSI). Molecular epidemiology of the S. aureus strains were determined by pulse field gel electrophoresis (PFGE).Results: PFGE identified 14 clusters which included 29 different pulso-types prevailing in the Burns Unit. Of the 29 types, 11 contained two or more strains of the same pulso-type. These MRSA isolates  were highly resistant to various kinds of penicillin and cephalosporin (85 – 100 %). Among the important anti-staphylococal agents tested, 17 % of the isolates were resistant to fusidic acid and linezolid. All the 54 strains were susceptible to vancomycin.Conclusion: Several of the same pulso-types prevail in the Burns Unit of KTH. Furthermore, 29 pulsotypes  mong the 54 strains suggest the diversity of the MRSA strains collected from burns patients.Keywords: Epidemiology, Pulso-type, Fusidic acid, Linezolid, Vancomycin, Methicillin-resistant Staphylococcus aureus, Burn

    URLLC for 5G and Beyond: Requirements, Enabling Incumbent Technologies and Network Intelligence

    The tactile internet (TI) is believed to be the prospective advancement of the internet of things (IoT), comprising human-to-machine and machine-to-machine communication. TI focuses on enabling real-time interactive techniques with a portfolio of engineering, social, and commercial use cases. For this purpose, the prospective 5{th} generation (5G) technology focuses on achieving ultra-reliable low latency communication (URLLC) services. TI applications require an extraordinary degree of reliability and latency. The 3{rd} generation partnership project (3GPP) defines that URLLC is expected to provide 99.99% reliability of a single transmission of 32 bytes packet with a latency of less than one millisecond. 3GPP proposes to include an adjustable orthogonal frequency division multiplexing (OFDM) technique, called 5G new radio (5G NR), as a new radio access technology (RAT). Whereas, with the emergence of a novel physical layer RAT, the need for the design for prospective next-generation technologies arises, especially with the focus of network intelligence. In such situations, machine learning (ML) techniques are expected to be essential to assist in designing intelligent network resource allocation protocols for 5G NR URLLC requirements. Therefore, in this survey, we present a possibility to use the federated reinforcement learning (FRL) technique, which is one of the ML techniques, for 5G NR URLLC requirements and summarizes the corresponding achievements for URLLC. We provide a comprehensive discussion of MAC layer channel access mechanisms that enable URLLC in 5G NR for TI. Besides, we identify seven very critical future use cases of FRL as potential enablers for URLLC in 5G NR

    The ligational behavior of an isatinic quinolyl hydrazone towards copper(II)- ions

    <p>Abstract</p> <p>Background</p> <p>The importance of the isatinic quinolyl hydrazones arises from incorporating the quinoline ring with the indole ring. Quinoline ring has therapeutic and biological activities whereas, the indole ring occurs in Jasmine flowers and Orange blossoms. As a ligand, the isatin moiety is potentially ambidentate and can coordinate the metal ions either through its lactam or lactim forms. In a previous study, the ligational behavior of a phenolic quinolyl hydrazone towards copper(II)- ions has been studied. As continuation of our interest, the present study is planned to check the ligational behavior of an isatinic quinolyl hydrazone.</p> <p>Results</p> <p>New homo- and heteroleptic copper(II)- complexes were obtained from the reaction of an isatinic quinolyl hydrazone (HL) with several copper(II)- salts <it>viz. </it>Clˉ, Brˉ, NO<sub>3</sub>ˉ, ClO<sub>4</sub><sup>-</sup>, SO<sub>4</sub><sup>2- </sup>and AcO<sup>-</sup>. The obtained complexes have O<sub>h</sub>, T<sub>d </sub>and D<sub>4h</sub>- symmetry and fulfill the strong coordinating ability of Clˉ, Brˉ, NO<sub>3</sub>ˉ and SO<sub>4</sub><sup>2- </sup>anions. Depending on the type of the anion, the ligand coordinates the copper(II)- ions either through its lactam (NO<sub>3</sub>ˉ and ClO<sub>4</sub><sup>-</sup>) or lactim (the others) forms.</p> <p>Conclusion</p> <p>The effect of anion for the same metal ion is obvious from either the geometry of the isolated complexes (O<sub>h</sub>, T<sub>d </sub>and D<sub>4h</sub>) or the various modes of bonding. Also, the obtained complexes fulfill the strong coordinating ability of Clˉ, Brˉ, NO<sub>3</sub>ˉ and SO<sub>4</sub><sup>2- </sup>anions in consistency with the donor ability of the anions. In case of copper(II)- acetate, a unique homoleptic complex (<b>5</b>) was obtained in which the AcO<sup>- </sup>anion acts as a base enough to quantitatively deprotonate the hydrazone. The isatinic hydrazone uses its lactim form in most complexes.</p

    Paneth cell - rich regions separated by a cluster of Lgr5+ cells initiate crypt fission in the intestinal stem cell niche

    The crypts of the intestinal epithelium house the stem cells that ensure the continual renewal of the epithelial cells that line the intestinal tract. Crypt number increases by a process called crypt fission, the division of a single crypt into two daughter crypts. Fission drives normal tissue growth and maintenance. Correspondingly, it becomes less frequent in adulthood. Importantly, fission is reactivated to drive adenoma growth. The mechanisms governing fission are poorly understood. However, only by knowing how normal fission operates can cancer-associated changes be elucidated. We studied normal fission in tissue in three dimensions using high-resolution imaging and used intestinal organoids to identify underlying mechanisms. We discovered that both the number and relative position of Paneth cells and Lgr5+ cells are important for fission. Furthermore, the higher stiffness and increased adhesion of Paneth cells are involved in determining the site of fission. Formation of a cluster of Lgr5+ cells between at least two Paneth-cell-rich domains establishes the site for the upward invagination that initiates fission

    A federated reinforcement learning framework for incumbent technologies in beyond 5G networks

    Incumbent wireless technologies for futuristic fifth generation (5G) and beyond 5G (B5G) networks, such as IEEE 802.11 ax (WiFi), are vital to provide ubiquitous ultra-reliable and low-latency communication services with massively connected devices. Amalgamating WiFi networks with 5G/B5G networks has attracted strong researcher interest over the past two decades, because over 70 percent of mobile data traffic is generated by WiFi devices. However, WiFi channel resource scarcity for 5G/B5G is becoming ever more critical. One current problem regarding channel resource allocation is channel collision handling due to increased user densities. Reinforcement learning (RL) algorithms have recently helped develop prominent behaviorist learning techniques for resource allocation in 5G/B5G networks. An agent optimizes its behavior in an RL-based algorithm based on reward and accumulated value. However, densely deployed WiFi environments are distributed and dynamic, with frequent changes. Thus, relying on individual local estimations leads to higher error variance. Therefore, this article proposes a federated RL-based channel resource allocation framework for 5G/B5G networks, and suggests collaborating learning estimates for faster learning convergence. Experimental results verify that the proposed approach optimizes WiFi performance in terms of throughput by collaborative channel access parameter selection. This study also highlights six potential applications for the proposed framework

    <p>Abstract</p> <p>Background</p> <p>The heterocyclic hydrazones constitute an important class of biologically active drug molecules. The hydrazones have also been used as herbicides, insecticides, nematocides, redenticides, and plant growth regulators as well as plasticizers and stabilizers for polymers. The importance of the phenolic quinolyl hydrazones arises from incorporating the quinoline ring with the phenolic compound; 2,4-dihydroxy benzaldehyde. Quinoline ring has therapeutic and biological activities whereas, phenols have antiseptic and disinfectants activities and are used in the preparation of dyes, bakelite and drugs. The present study is planned to check the effect of the counter anions on the type and geometry of the isolated copper(II)- complexes as well as the ligational behavior of the phenolic hydrazone; 4-[(2-(4,8-dimethylquinolin-2-yl)hydrazono)methyl] benzene-1,3-diol; (H<sub>2</sub>L).</p> <p>Results</p> <p>A phenolic quinolyl hydrazone (H<sub>2</sub>L) was allowed to react with various copper(II)- salts (Cl‾, Br‾, NO<sub>3</sub>‾, ClO<sub>4</sub>‾, AcO‾, SO<sub>4</sub><sup>2-</sup>). The reactions afforded dimeric complexes (ClO<sub>4</sub>‾, AcO‾ ), a binuclear complex (NO<sub>3</sub>‾ ) and mononuclear complexes (the others; Cl‾, Br‾, SO<sub>4</sub><sup>2-</sup>). The isolated copper(II)- complexes have octahedral, square pyramid and square planar geometries. Also, they reflect the strong coordinating ability of NO<sub>3</sub>‾, Cl‾, Br‾, AcO‾ and SO<sub>4</sub><sup>2- </sup>anions. Depending on the type of the anion, the ligand showed three different modes of bonding <it>viz</it>. (NN)<sup>0 </sup>for the mononuclear complexes (<b>3, 4, 6</b>), (NO)<sup>- </sup>with O- bridging for the dimeric complexes (<b>1, 5</b>) and a mixed mode [(NN)<sup>0 </sup>+ (NO)<sup>- </sup>with O- bridging] for the binuclear nitrato- complex (<b>2</b>).</p> <p>Conclusion</p> <p>The ligational behavior of the phenolic hydrazone (H<sub>2</sub>L) is highly affected by the type of the anion. The isolated copper(II)- complexes reflect the strong coordinating power of the SO<sub>4</sub><sup>2-</sup>, AcO‾, Br‾, Cl‾ and NO<sub>3</sub>‾ anions. Also, they reflect the structural diversity (octahedral, square pyramid and square planar) depending on the type of the counter anion.</p

    Hodgkin's lymphoma presenting with heart failure: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Cardiac involvement in malignant lymphoma is one of the least investigated subjects in oncology. This article reports a case of cardiac involvement in Hodgkin's lymphoma which presented as heart failure.</p> <p>Case presentation</p> <p>We report the case of an 8-year-old Afghan girl with Hodgkin's lymphoma. The disease presented with systemic signs and symptoms, including abdominal distension, weakness, pallor, chills, fever, generalized edema, hepatosplenomegaly and generalized lymphadenopathy, as well as signs of heart failure. Test results showed a rare form of heart metastasis.</p> <p>Conclusion</p> <p>We report a case of Hodgkin's lymphoma with metastasis to the heart, detected premortem. Although the involvement of the heart in a malignancy is relatively common, premortem detection is unusual and only few studies have reported it in the literature.</p

    The ribosome lowers the entropic penalty of protein folding

    Most proteins fold during biosynthesis on the ribosome1, and co-translational folding energetics, pathways and outcomes of many proteins have been found to differ considerably from those in refolding studies2–10. The origin of this folding modulation by the ribosome has remained unknown. Here we have determined atomistic structures of the unfolded state of a model protein on and off the ribosome, which reveal that the ribosome structurally expands the unfolded nascent chain and increases its solvation, resulting in its entropic destabilization relative to the peptide chain in isolation. Quantitative 19F NMR experiments confirm that this destabilization reduces the entropic penalty of folding by up to 30 kcal mol−1 and promotes formation of partially folded intermediates on the ribosome, an observation that extends to other protein domains and is obligate for some proteins to acquire their active conformation. The thermodynamic effects also contribute to the ribosome protecting the nascent chain from mutation-induced unfolding, which suggests a crucial role of the ribosome in supporting protein evolution. By correlating nascent chain structure and dynamics to their folding energetics and post-translational outcomes, our findings establish the physical basis of the distinct thermodynamics of co-translational protein folding

    Acral post-traumatic tumoral calcinosis in pregnancy: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Tumoral calcinosis is an uncommon disorder characterized by the development of calcified masses within the peri-articular soft tissues of large joints, but rarely occurs within the hand.</p> <p>Case presentation</p> <p>We present the case of a 31-year-old pregnant Indian woman with a three-month history of painful swelling within the tip of her right middle finger following a superficial laceration. She was otherwise well and had normal serum calcium and phosphate levels. Plain radiography demonstrated a dense, lobulated cluster of calcified nodules within the soft tissues of the volar pulp space, consistent with a diagnosis of tumoral calcinosis. This diagnosis was confirmed on the basis of the histopathological examination following surgical excision.</p> <p>Conclusion</p> <p>To the best of our knowledge, we present the only reported case of acral tumoral calcinosis within the finger, and the first description of its occurrence during pregnancy. We review the etiology, pathogenesis and treatment of tumoral calcinosis.</p
