    Validity of pre-eclampsia registration in the medical birth registry of Norway for women participating in the norwegian mother and child cohort study, 1999-2010

    Background: The Norwegian Mother and Child Cohort Study (MoBa), a prospective population-based pregnancy cohort, is a valuable database for studying causes of pre-eclampsia. Pre-eclampsia data in MoBa come from the Medical Birth Registry of Norway (MBRN); thus, we wanted to study the validity of MBRN pre-eclampsia registration for MoBa women. Methods: We selected all MoBa pregnancies with pre-eclampsia registered in the MBRN (n = 4081) and a random control group (n = 2000) without pre-eclampsia registrations. After excluding two delivery units not participating in MoBa and one no longer operating, units were asked to provide copies of antenatal charts with blood pressure and urinary measurements from all antenatal visits during pregnancy, and hospital discharge codes from thedelivery stay. We received data for 5340 pregnancies delivered 1999-2010 (87% of all eligible). We calculated positive predictive value (PPV), and sensitivity and specificity of MBRN registration, using hypertension and proteinuria on the antenatal charts and/or hospital discharge codes indicating pre-eclampsia as gold standard. Results: Overall PPV was 83.9% [95% CI 82.7, 85.1] and was higher when women were primiparous, or delivered preterm or low birthweight infants. Severe pre-eclampsia in the MBRN was found to be a true severe preeclampsia in 70% of cases. Extrapolating to the total MoBa population, the estimated sensitivity was low - 43.0% (38.7, 48.2) - while specificity was high - 99.2% (99.2, 99.3). False negative cases seemed to have mild forms of pre-eclampsia. Conclusions: PPV and specificity of pre-eclampsia registration in the MBRN during 1999-2010 was satisfactory, while sensitivity was low

Planned upgrades to DRIFT IId are detailed, along with ongoing work towards DRIFT III, which aims to be the world's first 10 m3-scale directional Dark Matter detector

    Do Genetic Variants Modify the Effect of Smoking on Risk of Preeclampsia in Pregnancy?

    Objective Maternal smoking is associated with as much as a 50% reduced risk of preeclampsia, despite increasing risk of other poor pregnancy outcomes that often co-occur with preeclampsia, such as preterm birth and fetal growth restriction. Researchers have long sought to understand whether this perplexing association is biologically based, or a result of noncausal mechanisms. We examined whether smoking-response genes modify the smoking-preeclampsia association to investigate potential biological explanations. Study Design We conducted a nested case-control study within the Norwegian Mother, Father and Child Birth Cohort (1999-2008) of 2,596 mother-child dyads. We used family-based log-linear Poisson regression to examine modification of the maternal smoking-preeclampsia relationship by maternal and fetal single nucleotide polymorphisms involved in cellular processes related to components of cigarette smoke (n = 1,915 with minor allele frequency ≥10%). We further investigated the influence of smoking cessation during pregnancy. Results Three polymorphisms showed overall (p < 0.001) multiplicative interaction between smoking and maternal genotype. For rs3765692 (TP73) and rs10770343 (PIK3C2G), protection associated with smoking was reduced with two maternal copies of the risk allele and was stronger in continuers than quitters (interaction p = 0.02 for both loci, based on testing 3-level smoking by 3-level genotype). For rs2278361 (APAF1) the inverse smoking-preeclampsia association was eliminated by the presence of a single risk allele, and again the trend was stronger in continuers than in quitters (interaction p = 0.01). Conclusion Evidence for gene-smoking interaction was limited, but differences by smoking cessation warrant further investigation. We demonstrate the potential utility of expanded dyad methods and gene-environment interaction analyses for outcomes with complex relationships between maternal and fetal genotypes and exposures.     Spin-dependent limits from the DRIFT-IId directional dark matter detector

    Data are presented from the DRIFT-IId detector operated in the Boulby Underground Science Facility in England. A 0.8 m 3 fiducial volume, containing partial pressures of 30 Torr CS 2 and 10 Torr CF 4, was exposed for a duration of 47.4 live-time days with sufficient passive shielding to provide a neutron free environment within the detector. The nuclear recoil events seen are consistent with a remaining low-level background from the decay of radon daughters attached to the central cathode of the detector. However, charge from such events must drift across the entire width of the detector, and thus display large diffusion upon reaching the readout planes of the device. Exploiting this feature, it is shown to be possible to reject energy depositions from these Radon Progeny Recoil events while still retaining sensitivity to fiducial-volume nuclear recoil events. The response of the detector is then interpreted, using the F nuclei content of the gas, in terms of sensitivity to proton spin-dependent WIMP-nucleon interactions, displaying a minimum in sensitivity cross section at 1.8 pb for a WIMP mass of 100 GeV/c 2.     Professionalism, Golf Coaching and a Master of Science Degree: A commentary

    As a point of reference I congratulate Simon Jenkins on tackling the issue of professionalism in coaching. As he points out coaching is not a profession, but this does not mean that coaching would not benefit from going through a professionalization process. As things stand I find that the stimulus article unpacks some critically important issues of professionalism, broadly within the context of golf coaching. However, I am not sure enough is made of understanding what professional (golf) coaching actually is nor how the development of a professional golf coach can be facilitated by a Master of Science Degree (M.Sc.). I will focus my commentary on these two issues

    Radon in the DRIFT-II directional dark matter TPC: emanation, detection and mitigation

    Radon gas emanating from materials is of interest in environmental science and also a major concern in rare event non-accelerator particle physics experiments such as dark matter and double beta decay searches, where it is a major source of background. Notable for dark matter experiments is the production of radon progeny recoils (RPRs), the low energy (~ 100 keV) recoils of radon daughter isotopes, which can mimic the signal expected from WIMP interactions. Presented here are results of measurements of radon emanation from detector materials in the 1 m3 DRIFT-II directional dark matter gas time projection chamber experiment. Construction and operation of a radon emanation facility for this work is described, along with an analysis to continuously monitor DRIFT data for the presence of internal 222Rn and 218Po. Applying this analysis to historical DRIFT data, we show how systematic substitution of detector materials for alternatives, selected by this device for low radon emanation, has resulted in a factor of ~ 10 reduction in internal radon rates. Levels are found to be consistent with the sum from separate radon emanation measurements of the internal materials and also with direct measurement using an attached alpha spectrometer. The current DRIFT detector, DRIFT-IId, is found to have sensitivity to 222Rn of 2.5 μBql−1 with current analysis efficiency, potentially opening up DRIFT technology as a new tool for sensitive radon assay of materials

    A Family Based Study of Carbon Monoxide and Nitric Oxide Signalling Genes and Preeclampsia

    Background: Preeclampsia is thought to originate during placentation, with incomplete remodelling and perfusion of the spiral arteries leading to reduced placental vascular capacity. Nitric oxide (NO) and carbon monoxide (CO) are powerful vasodilators that play a role in the placental vascular system. Although family clustering of preeclampsia has been observed, the existing genetic literature is limited by a failure to consider both mother and child. Methods: We conducted a nested case–control study within the Norwegian Mother and Child Birth Cohort of 1545 case-pairs and 995 control-pairs from 2540 validated dyads (2011 complete pairs, 529 missing mother or child genotype). We selected 1518 single-nucleotide polymorphisms (SNPs) with minor allele frequency >5% in NO and CO signalling pathways. We used log-linear Poisson regression models and likelihood ratio tests to assess maternal and child effects. Results: One SNP met criteria for a false discovery rate Q-value <0.05. The child variant, rs12547243 in adenylate cyclase 8 (ADCY8), was associated with an increased risk (relative risk [RR] 1.42, 95% confidence interval [CI] 1.20, 1.69 for AG vs. GG, RR 2.14, 95% CI 1.47, 3.11 for AA vs. GG, Q = 0.03). The maternal variant, rs30593 in PDE1C was associated with a decreased risk for the subtype of preeclampsia accompanied by early delivery (RR 0.45, 95% CI 0.27, 0.75 for TC vs. CC; Q = 0.02). None of the associations were replicated after correction for multiple testing. Conclusions: This study uses a novel approach to disentangle maternal and child genotypic effects of NO and CO signalling genes on preeclampsia

    Reducing DRIFT backgrounds with a submicron aluminized-mylar cathode

    Background events in the DRIFT-IId dark matter detector, mimicking potential WIMP signals, are predominantly caused by alpha decays on the central cathode in which the alpha particle is completely or partially absorbed by the cathode material. We installed a View the MathML source thick aluminized-mylar cathode as a way to reduce the probability of producing these backgrounds. We study three generations of cathode (wire, thin-film, and radiologically clean thin-film) with a focus on the ratio of background events to alpha decays. Two independent methods of measuring the absolute alpha decay rate are used to ensure an accurate result, and agree to within 10%. Using alpha range spectroscopy, we measure the radiologically cleanest cathode version to have a contamination of 3.3Âą0.1 ppt 234U and 73Âą2 ppb 238U. This cathode reduces the probability of producing an RPR from an alpha decay by a factor of 70Âą20 compared to the original stainless steel wire cathode. First results are presented from a texturized version of the cathode, intended to be even more transparent to alpha particles. These efforts, along with other background reduction measures, have resulted in a drop in the observed background rate from 500/day to 1/day. With the recent implementation of full-volume fiducialization, these remaining background events are identified, allowing for background-free operation
