    The effect of skin fatty acids on Staphylococcus aureus

    Staphylococcus aureus is a commensal of the human nose and skin. Human skin fatty acids, in particular cis-6-hexadecenoic acid (C-6-H), have high antistaphylococcal activity and can inhibit virulence determinant production. Here, we show that sub-MIC levels of C-6-H result in induction of increased resistance. The mechanism(s) of C-6-H activity was investigated by combined transcriptome and proteome analyses. Proteome analysis demonstrated a pleiotropic effect of C-6-H on virulence determinant production. In response to C-6-H, transcriptomics revealed altered expression of over 500 genes, involved in many aspects of virulence and cellular physiology. The expression of toxins (hla, hlb,hlgBC) was reduced, whereas that of host defence evasion components (cap, sspAB, katA) was increased. In particular, members of the SaeRS regulon had highly reduced expression, and the use of specific mutants revealed that the effect on toxin production is likely mediated via SaeRS

    Bistability of Slow and Fast Traveling Waves in Fluid Mixtures

    The appearence of a new type of fast nonlinear traveling wave states in binary fluid convection with increasing Soret effect is elucidated and the parameter range of their bistability with the common slower ones is evaluated numerically. The bifurcation behavior and the significantly different spatiotemporal properties of the different wave states - e.g. frequency, flow structure, and concentration distribution - are determined and related to each other and to a convenient measure of their nonlinearity. This allows to derive a limit for the applicability of small amplitude expansions. Additionally an universal scaling behavior of frequencies and mixing properties is found.     Three-Nucleon Force Effects in Nucleon Induced Deuteron Breakup: Predictions of Current Models (I)

    An extensive study of three-nucleon force effects in the entire phase space of the nucleon-deuteron breakup process, for energies from above the deuteron breakup threshold up to 200 MeV, has been performed. 3N Faddeev equations have been solved rigorously using the modern high precision nucleon-nucleon potentials AV18, CD Bonn, Nijm I, II and Nijm 93, and also adding 3N forces. We compare predictions for cross sections and various polarization observables when NN forces are used alone or when the two pion-exchange Tucson-Melbourne 3NF was combined with each of them. In addition AV18 was combined with the Urbana IX 3NF and CD Bonn with the TM' 3NF, which is a modified version of the TM 3NF, more consistent with chiral symmetry. Large but generally model dependent 3NF effects have been found in certain breakup configurations, especially at the higher energies, both for cross sections and spin observables. These results demonstrate the usefulness of the kinematically complete breakup reaction in testing the proper structure of 3N forces.Comment: 42 pages, 20 ps figures, 2 gif figure

    Spin observables in deuteron-proton radiative capture at intermediate energies

    A radiative deuteron-proton capture experiment was carried out at KVI using polarized-deuteron beams at incident energies of 55, 66.5, and 90 MeV/nucleon. Vector and tensor-analyzing powers were obtained for a large angular range. The results are interpreted with the help of Faddeev calculations, which are based on modern two- and three-nucleon potentials. Our data are described well by the calculations, and disagree significantly with the observed tensor anomaly at RCNP.Comment: 10 pages, 4 figures, submitted to PL

    Staphylococcal PknB as the First Prokaryotic Representative of the Proline-Directed Kinases

    In eukaryotic cell types, virtually all cellular processes are under control of proline-directed kinases and especially MAP kinases. Serine/threonine kinases in general were originally considered as a eukaryote-specific enzyme family. However, recent studies have revealed that orthologues of eukaryotic serine/threonine kinases exist in bacteria. Moreover, various pathogenic species, such as Yersinia and Mycobacterium, require serine/threonine kinases for successful invasion of human host cells. The substrates targeted by bacterial serine/threonine kinases have remained largely unknown. Here we report that the serine/threonine kinase PknB from the important pathogen Staphylococcus aureus is released into the external milieu, which opens up the possibility that PknB does not only phosphorylate bacterial proteins but also proteins of the human host. To identify possible human targets of purified PknB, we studied in vitro phosphorylation of peptide microarrays and detected 68 possible human targets for phosphorylation. These results show that PknB is a proline-directed kinase with MAP kinase-like enzymatic activity. As the potential cellular targets for PknB are involved in apoptosis, immune responses, transport, and metabolism, PknB secretion may help the bacterium to evade intracellular killing and facilitate its growth. In apparent agreement with this notion, phosphorylation of the host-cell response coordinating transcription factor ATF-2 by PknB was confirmed by mass spectrometry. Taken together, our results identify PknB as the first prokaryotic representative of the proline-directed kinase/MAP kinase family of enzymes

    Nucleon-deuteron elastic scattering as a tool to probe properties of three-nucleon forces

    Faddeev equations for elastic Nd scattering have been solved using modern NN forces combined with the Tucson-Melbourne two-pion exchange three-nucleon force, with a modification thereof closer to chiral symmetry and the Urbana IX three-nucleon force. Theoretical predictions for the differential cross section and several spin observables using NN forces only and NN forces combined with three-nucleon force models are compared to each other and to the existing data. A wide range of energies from 3 to 200 MeV is covered. Especially at the higher energies striking three-nucleon force effects are found, some of which are supported by the still rare set of data, some are in conflict with data and thus very likely point to defects in those three-nucleon force models.Comment: 30 pages, 14 Postscript figures; now minor changes in figures and reference

    Polarization transfer in the 16^{16}O(p,p)(p,p') reaction at forward angles and structure of the spin-dipole resonances

    Cross sections and polarization transfer observables in the 16^{16}O(p,p)(p,p') reactions at 392 MeV were measured at several angles between θlab=\theta_{lab}= 0^\circ and 14^\circ. The non-spin-flip (ΔS=0{\Delta}S=0) and spin-flip (ΔS=1{\Delta}S=1) strengths in transitions to several discrete states and broad resonances in 16^{16}O were extracted using a model-independent method. The giant resonances in the energy region of Ex=19E_x=19-27 MeV were found to be predominantly excited by ΔL=1{\Delta}L=1 transitions. The strength distribution of spin-dipole transitions with ΔS=1{\Delta}S=1 and ΔL=1{\Delta}L=1 were deduced. The obtained distribution was compared with a recent shell model calculation. Experimental results are reasonably explained by distorted-wave impulse approximation calculations with the shell model wave functions.Comment: 28 pages RevTex, including 9 figures, to be published in Phys.     Associations between the LEP -2548G/A Promoter and Baseline Weight and between LEPR Gln223Arg and Lys656Asn Variants and Change in BMI z Scores in Arab Children and Adolescents Treated with Risperidone

    Data on baseline (antipsychotic-naïve) age, weight, and height and change in these over three subsequent follow up time points up to 313.6 days (CI 303.5-323.7), were collected from 181 risperidone-treated children and adolescents (mean age 12.58 years, SD 4.99, range 2.17-17.7) attending a pediatric neurology clinic in Saudi Arabia. Owing to differences in genotypic distributions in subsamples, results are reported from the white Arabs (N=144). Age and gender-normed BMI-standardised z scores (BMI z) were calculated (lmsgrowth program). Linear regression was performed for baseline weight and BMI z, while change in BMI z was assessed using random effects ordered logistic regression. The following SNPs were analyzed: rs7799039 in the LEP promoter, rs1805094 (previously rs8179183), rs1137100 and rs1137101 in the LEPR, and rs1414334 in HTR2C. We found a nominally significant association between rs7799309 and baseline weight, adjusting for height, age, gender and diagnosis (A/G, P=0.035, β=-3.62, compared to G/G). rs1137101 (G/G, P=0.018, OR=4.13 compared to A/A) and rs1805094 C-allele carriers (P=0.019, OR=0.51) showed nominally significant associations with change in BMI z categories. Our data support and replicate previous relevant associations for these variants including with weight gain on risperidone, whilst being the first to report such associations in those of Arab ethnicity

    Measurement of Analyzing Power for Proton-Carbon Elastic Scattering in the Coulomb-Nuclear Interference Region with a 22-GeV/c Polarized Proton Beam

    The analyzing power for proton-carbon elastic scattering in the coulomb-nuclear interference region of momentum transfer, 9.0×103<t<4.1×1029.0\times10^{-3}<-t<4.1\times10^{-2} (GeV/c)2c)^{2}, was measured with a 21.7 GeV/cc polarized proton beam at the Alternating Gradient Synchrotron of Brookhaven National Laboratory. The ratio of hadronic spin-flip to non-flip amplitude, r5r_5, was obtained from the analyzing power to be Rer5=0.088±0.058\text{Re} r_5=0.088\pm 0.058 and Imr5=0.161±0.226\text{Im} r_5=-0.161\pm 0.226.Comment: 4 pages, 4 figures and 1 table.
