    Direct and indirect control of the initiation of meiotic recombination by DNA damage checkpoint mechanisms in budding yeast

    Meiotic recombination plays an essential role in the proper segregation of chromosomes at meiosis I in many sexually reproducing organisms. Meiotic recombination is initiated by the scheduled formation of genome-wide DNA double-strand breaks (DSBs). The timing of DSB formation is strictly controlled because unscheduled DSB formation is detrimental to genome integrity. Here, we investigated the role of DNA damage checkpoint mechanisms in the control of meiotic DSB formation using budding yeast. By using recombination defective mutants in which meiotic DSBs are not repaired, the effect of DNA damage checkpoint mutations on DSB formation was evaluated. The Tel1 (ATM) pathway mainly responds to unresected DSB ends, thus the sae2 mutant background in which DSB ends remain intact was employed. On the other hand, the Mec1 (ATR) pathway is primarily used when DSB ends are resected, thus the rad51 dmc1 double mutant background was employed in which highly resected DSBs accumulate. In order to separate the effect caused by unscheduled cell cycle progression, which is often associated with DNA damage checkpoint defects, we also employed the ndt80 mutation which permanently arrests the meiotic cell cycle at prophase I. In the absence of Tel1, DSB formation was reduced in larger chromosomes (IV, VII, II and XI) whereas no significant reduction was found in smaller chromosomes (III and VI). On the other hand, the absence of Rad17 (a critical component of the ATR pathway) lead to an increase in DSB formation (chromosomes VII and II were tested). We propose that, within prophase I, the Tel1 pathway facilitates DSB formation, especially in bigger chromosomes, while the Mec1 pathway negatively regulates DSB formation. We also identified prophase I exit, which is under the control of the DNA damage checkpoint machinery, to be a critical event associated with down-regulating meiotic DSB formation

    Perspectives of older women in the Netherlands: identifying motivators and barriers for healthy lifestyles and determinants of healthy aging

    BACKGROUND: Women have a higher life expectancy than men but experience more years with physical disabilities in daily life at older ages, especially women with a migration background. This pinpoints older women as an important target group for strategies that stimulate healthy lifestyle, which benefits healthy aging. Our study investigates motivators and barriers for healthy lifestyles and perspectives on determinants of healthy aging of older women. This provides essential information for developing targeted strategies. METHODS: Data was collected by semi-structured digital interviews from February till June 2021. Women aged 55 years and older living in the Netherlands (n = 34) with a native Dutch (n = 24), Turkish (n = 6) or Moroccan (n = 4) migration background were included. Two main subjects were investigated: (1) motivators and barriers on their current lifestyles regarding smoking, alcohol consumption, physical activity, diet and sleep and (2) perspectives on determinants of healthy aging. Interviews were analyzed using Krueger's framework. RESULTS: Personal health was the most common motivator for a healthy lifestyle. In addition, peer pressure and being outdoors were specific motivators for physical activity. Bad weather conditions and personal dislike to be active were specific barriers. The social environment, personal preferences and personal belief to compensate with other healthy lifestyle behaviors were barriers for low alcohol consumption. Personal preferences (liking unhealthy food and not making time) were the main barriers for a healthy diet. Sleep was not perceived as a form of lifestyle behavior, but rather as a personal trait. Since there were no smokers, specific barriers were not mentioned. For Turkish-Dutch and Moroccan-Dutch women, additional barriers and motivators were culture and religion. These were strong motivators to abstain from alcohol consumption and smoking, but a barrier for a healthy diet. With regard to perspectives on determinants of healthy aging, positive views on aging and being physically active were perceived as most important. Women often wanted to increase their physical activity or healthy diet to stimulate healthy aging. Among Turkish-Dutch and Moroccan-Dutch women, healthy aging was also perceived as something in the hands of God. CONCLUSIONS: Although motivators and barriers for a healthy lifestyle and perspectives on healthy aging vary for distinct lifestyles, personal health is a common motivator across all lifestyles. Having a migration background added culture and religion as distinct barriers and motivations. Strategies to improve lifestyle among older women should therefore have a tailored, culture sensitive approach (if applicable) for distinct lifestyle factors

    Photometric Properties of Void Galaxies in the Sloan Digital Sky Survey

    Using a nearest neighbor analysis, we construct a sample of void galaxies from the Sloan Digital Sky Survey (SDSS) and compare the photometric properties of these galaxies to the population of non-void (wall) galaxies. We trace the density field of galaxies using a volume-limited sample with z_{max}=0.089. Galaxies from the flux-limited SDSS with z\leq z_{max} and fewer than three volume-limited neighbors within 7h^{-1}Mpc are classified as void galaxies. This criterion implies a density contrast \delta \rho/ \rho < -0.6 around void galaxies. From 155,000 galaxies, we obtain a sub-sample of 13,742 galaxies with z\leq z_{max}, from which we identify 1,010 galaxies as void galaxies. To identify an additional 194 faint void galaxies from the SDSS in the nearby universe, r~ 72 h^{-1}Mpc, we employ volume-limited samples extracted from the Updated Zwicky Catalog and the Southern Sky Redshift Survey with z_{max}=0.025 to trace the galaxy distribution. Our void galaxies span a range of absolute magnitude from M_r=-13.5 to M_r=-22.5. Using SDSS photometry, we compare the colors, concentration indices, and Sersic indices of the void and wall samples. Void galaxies are significantly bluer than galaxies lying at higher density. The population of void galaxies with M_r ~ M* +1 and brighter is on average bluer and more concentrated (later type) than galaxies outside of voids. The latter behavior is only partly explained by the paucity of luminous red galaxies in voids. These results generally agree with the predictions of semi-analytic models for galaxy formation in cold dark matter models, which indicate that void galaxies should be relatively bluer, more disklike, and have higher specific star formation rates.Comment: Accepted for publication in Ap

    Influence of operating parameters on the biodegradation of steroid estrogens and nonylphenolic compounds during biological wastewater treatment processes

To access the final edited and published work see http://pubs.acs.org/doi/abs/10.1021/es901612v.This study investigated operational factors influencing the removal of steroid estrogens and nonylphenolic compounds in two sewage treatment works, one a nitrifying/denitrifying activated sludge plant and the other a nitrifying/denitrifying activated sludge plant with phosphorus removal. Removal efficiencies of >90% for steroid estrogens and for longer chain nonylphenol ethoxylates (NP4−12EO) were observed at both works, which had equal sludge ages of 13 days. However, the biological activity in terms of milligrams of estrogen removed per day per tonne of biomass was found to be 50−60% more efficient in the nitrifying/denitrifying activated sludge works compared to the works which additionally incorporated phosphorus removal. A temperature reduction of 6 °C had no impact on the removal of free estrogens, but removal of the conjugated estrone-3-sulfate was reduced by 20%. The apparent biomass sorption (LogKp) values were greater in the nitrifying/denitrifying works than those in the nitrifying/denitrifying works with phosphorus removal for both steroid estrogens and nonylphenolic compounds possibly indicating a different cell surface structure and therefore microbial population. The difference in biological activity (mg tonne−1 d−1) identified in this study, of up to seven times, suggests that there is the potential for enhancing the removal of estrogens and nonylphenols if more detailed knowledge of the factors responsible for these differences can be identified and maximized, thus potentially improving the quality of receiving waters.Public Utilities Board (Singapore), Anglian Water Ltd, Severn Trent Water Ltd, Thames Water Utilities Ltd, United Utilities 393 Plc and Yorkshire Water Services

    Optical and Near-Infrared Color Profiles in Nearby Early-Type Galaxies and The Implied Age and Metallicity Gradients

    We present results of age and metallicity gradient analysis inferred from both optical and near-infrared surface photometry. The analysis is based on a sample of 36 nearby early-type galaxies, obtained from the Early Data Release of the Sloan Digital Sky Survey and the Two Micron All Sky Survey. Surface brightness profiles were derived in each band, and used to study the color gradients of the galaxies. Using simple stellar population models with both optical and near infrared colors, we may interpret the color gradients in term of age and metallicity gradients of galaxies. Using gZdlogZmet/dlogRg_Z \equiv d \log Z_{\rm met} / d \log R and gA=dlogAge/dlogRg_A = d \log {\rm Age} / d \log R to represent the metallicity and age gradients, we found a median value of gZ=0.25±0.03g_Z=-0.25\pm 0.03 for the metallicity gradient, with a dispersion σgZ=0.19±0.02\sigma_{g_Z}=0.19\pm0.02. The corresponding values for the age gradient were gA=0.02±0.04g_A=0.02\pm 0.04 and σgA=0.25±0.03\sigma_{g_A}=0.25\pm0.03. These results are in good agreement with recent observational results, as well as with recent simulations that suggest both monolithic collapse and major merger have played important roles in the formation of early-type galaxies. Our results demonstrate the potential of using multi-waveband colors obtained from current and future optical and infrared surveys in constraining the age and metallicity gradients of early-type galaxies.Comment: 40 pages, 14 figures.
