Long-term monitoring of habitats and reef fish found inside and outside the U.S. Virgin Islands Coral Reef National Monument: a comparative assessment
The primary objective of this study was to assess the efficacy of the Virgin Islands Coral Reef National Monument (VICR), a marine protected area in St John, US Virgin Islands. Surveys of habitat and fishes inside and outside of VICR were conducted in 2003-2008. Areas outside the VICR had significantly more scleractinian corals, greater habitat complexity, and greater species richness and density of reef fishes than areas inside., Areas inside and outside the VICR exhibited significant decreases in percent scleractinian coral coverage over the study period. A contrasting trend of increasing macroalgal cover was also observed. No clear effect of the severe 2005 coral bleaching event was observed suggesting other causal factors. No obvious trends in the fish community were observed across the study period. The significant decline in habitat condition, coupled with the initial incorporation of some of the more degraded reefs into the marine protected area may result in a longer time period necessary to detect positive changes in the St. John coral reef ecosystem and associated reef fish abundance and community structure
IL-18 neutralization ameliorates obstruction-induced epithelialâmesenchymal transition and renal fibrosis
Ureteral obstruction results in renal fibrosis in part due to inflammatory injury. The role of interleukin-18 (IL-18), an important mediator of inflammation, in the genesis of renal fibrosis was studied using transgenic mice overexpressing human IL-18-binding protein. In addition, HK-2 cells were analyzed following direct exposure to IL-18 compared to control media. Two weeks after ureteral obstruction, the kidneys of wild-type mice had a significant increase in IL-18 production, collagen deposition, α-smooth muscle actin and RhoA expression, fibroblast and macrophage accumulation, chemokine expression, and transforming growth factor-ÎČ1 (TGF-ÎČ1) and tumor necrosis factor-α (TNF-α) production, whereas E-cadherin expression was simultaneously decreased. The transgenic mice with neutralized IL-18 activity exhibited significant reductions in these indicators of obstruction-induced renal fibrosis and epithelialâ mesenchymal transition, without demonstrating alterations in TGF-ÎČ1 or TNF-α activity. Similarly, the HK-2 cells exhibited increased α-smooth muscle actin expression and collagen production, and decreased E-cadherin expression in response to IL-18 stimulation without alterations in TNF-α or TGF-ÎČ1 activity. Our study demonstrates that IL-18 is a significant mediator of obstruction-induced renal fibrosis and epithelialâ mesenchymal transition independent of downstream TGF-ÎČ1 or TNF-α production
Cube law, condition factor and weight-length relationships: history, meta-analysis and recommendations
This study presents a historical review, a meta-analysis, and recommendations for users about weightâlength relationships, condition factors and relative weight equations. The historical review traces the developments of the respective concepts. The meta-analysis explores 3929 weightâlength relationships of the type W = aLb for 1773 species of fishes. It shows that 82% of the variance in a plot of log a over b can be explained by allometric versus isometric growth patterns and by different body shapes of the respective species. Across species median b = 3.03 is significantly larger than 3.0, thus indicating a tendency towards slightly positive-allometric growth (increase in relative body thickness or plumpness) in most fishes. The expected range of 2.5 < b < 3.5 is confirmed. Mean estimates of b outside this range are often based on only one or two weightâlength relationships per species. However, true cases of strong allometric growth do exist and three examples are given. Within species, a plot of log a vs b can be used to detect outliers in weightâlength relationships. An equation to calculate mean condition factors from weightâlength relationships is given as Kmean = 100aLbâ3. Relative weight Wrm = 100W/(amLbm) can be used for comparing the condition of individuals across populations, where am is the geometric mean of a and bm is the mean of b across all available weightâlength relationships for a given species. Twelve recommendations for proper use and presentation of weightâlength relationships, condition factors and relative weight are given
Differences in IV alcohol-induced dopamine release in the ventral striatum of social drinkers and nontreatment-seeking alcoholics
Background
Striatal dopamine (DA) has been implicated in alcohol use disorders, but it is still unclear whether or not alcohol can induce dopamine release in social drinkers. Furthermore, no data exist on dopamine responses to alcohol in dependent drinkers. We sought to characterize the DA responses to alcohol intoxication in moderately large samples of social drinkers (SD) and nontreatment-seeking alcoholics (NTS).
Methods
Twenty-four SD and twenty-one NTS received two [11C]raclopride (RAC) PET scans; one at rest, and one during an intravenous alcohol infusion, with a prescribed ascent to a target breath alcohol concentration (BrAC), at which it was then âclamped.â The alcohol clamp was started 5 min after scan start, with a linear increase in BrAC over 15 min to the target of 80 mg%, the legal threshold for intoxication. Target BrAC was maintained for 30 min. Voxel-wise binding potential (BPND) was estimated with MRTM2.
Results
IV EtOH induced significant increases in DA in the right ventral striatum in NTS, but not SD. No decreases in DA were observed in either group.
Conclusions
Alcohol intoxication results in distinct anatomic profiles of DA responses in SD and NTS, suggesting that in NTS, the striatal DA system may process effects of alcohol intoxication differently than in SD
An ecological correction to marine reserves boundaries in the US Virgin Islands
Marine protected areas (MPAs) are important tools for management of marine ecosystems. While desired, ecological and biological criteria are not always feasible to consider when establishing protected areas. In 2001, the Virgin Islands Coral Reef National Monument (VICR) in St. John, US Virgin Islands was established by Executive Order. VICR boundaries were based on administrative determination of Territorial Sea boundaries and land ownership at the time of the Territorial Submerged Lands Act of 1974. VICR prohibits almost all fishing and other extractive uses. Surveys of habitat and fishes inside and outside of VICR were conducted in 2002-07. Based on these surveys, areas outside VICR had significantly more hard corals; greater habitat complexity; and greater richness, abundance and biomass of reef fishes than areas within VICR, further supporting results from 2002-2004 (Monaco et al., 2007). The administrative (political) process used to establish VICR did not allow a robust ecological characterization of the area to determine the boundaries of the MPA. Efforts are underway to increase amounts of complex reef habitat within VICR by swapping a part of VICR that has little coral reef habitat for a Territorially-owned area within VICR that contains a coral reef with higher coral cover
A Survey on the Krein-von Neumann Extension, the corresponding Abstract Buckling Problem, and Weyl-Type Spectral Asymptotics for Perturbed Krein Laplacians in Nonsmooth Domains
In the first (and abstract) part of this survey we prove the unitary
equivalence of the inverse of the Krein--von Neumann extension (on the
orthogonal complement of its kernel) of a densely defined, closed, strictly
positive operator, for some in a Hilbert space to an abstract buckling problem operator.
This establishes the Krein extension as a natural object in elasticity theory
(in analogy to the Friedrichs extension, which found natural applications in
quantum mechanics, elasticity, etc.).

Radio frequency measurements of tunnel couplings and singletâtriplet spin states in Si:P quantum dots
Spin states of the electrons and nuclei of phosphorus donors in silicon are strong candidates for quantum information processing applications given their excellent coherence times. Designing a scalable donor-based quantum computer will require both knowledge of the relationship between device geometry and electron tunnel couplings, and a spin readout strategy that uses minimal physical space in the device. Here we use radio frequency reflectometry to measure singletâtriplet states of a few-donor Si:P double quantum dot and demonstrate that the exchange energy can be tuned by at least two orders of magnitude, from 20âÎŒeV to 8âmeV. We measure dotâlead tunnel rates by analysis of the reflected signal and show that they change from 100âMHz to 22âGHz as the number of electrons on a quantum dot is increased from 1 to 4. These techniques present an approach for characterizing, operating and engineering scalable qubit devices based on donors in silicon
Analysis of Microsatellite Variation in Drosophila melanogaster with Population-Scale Genome Sequencing
Genome sequencing technologies promise to revolutionize our understanding of genetics, evolution, and disease by making it feasible to survey a broad spectrum of sequence variation on a population scale. However, this potential can only be realized to the extent that methods for extracting and interpreting distinct forms of variation can be established. The error profiles and read length limitations of early versions of next-generation sequencing technologies rendered them ineffective for some sequence variant types, particularly microsatellites and other tandem repeats, and fostered the general misconception that such variants are inherently inaccessible to these platforms. At the same time, tandem repeats have emerged as important sources of functional variation. Tandem repeats are often located in and around genes, and frequent mutations in their lengths exert quantitative effects on gene function and phenotype, rapidly degrading linkage disequilibrium between markers and traits. Sensitive identification of these variants in large-scale next-gen sequencing efforts will enable more comprehensive association studies capable of revealing previously invisible associations. We present a population-scale analysis of microsatellite repeats using whole-genome data from 158 inbred isolates from the Drosophila Genetics Reference Panel, a collection of over 200 extensively phenotypically characterized isolates from a single natural population, to uncover processes underlying repeat mutation and to enable associations with behavioral, morphological, and life-history traits. Analysis of repeat variation from next-generation sequence data will also enhance studies of genome stability and neurodegenerative diseases
