    Direct Confirmation of the Asymmetry of the Cas A Supernova with Light Echoes

    We report the first detection of asymmetry in a supernova (SN) photosphere based on SN light echo (LE) spectra of Cas A from the different perspectives of dust concentrations on its LE ellipsoid. New LEs are reported based on difference images, and optical spectra of these LEs are analyzed and compared. After properly accounting for the effects of finite dust-filament extent and inclination, we find one field where the He I and H alpha features are blueshifted by an additional ~4000 km/s relative to other spectra and to the spectra of the Type IIb SN 1993J. That same direction does not show any shift relative to other Cas A LE spectra in the Ca II near-infrared triplet feature. We compare the perspectives of the Cas A LE dust concentrations with recent three-dimensional modeling of the SN remnant (SNR) and note that the location having the blueshifted He I and H alpha features is roughly in the direction of an Fe-rich outflow and in the opposite direction of the motion of the compact object at the center of the SNR. We conclude that Cas A was an intrinsically asymmetric SN. Future LE spectroscopy of this object, and of other historical SNe, will provide additional insight into the connection of explosion mechanism to SN to SNR, as well as give crucial observational evidence regarding how stars explode.Comment: 13 pages, 7 figures, accepted for publication in Ap

    Spectral Identification of an Ancient Supernova using Light Echoes in the LMC

    We report the successful identification of the type of the supernova responsible for the supernova remnant SNR 0509-675 in the Large Magellanic Cloud (LMC) using Gemini spectra of surrounding light echoes. The ability to classify outbursts associated with centuries-old remnants provides a new window into several aspects of supernova research and is likely to be successful in providing new constraints on additional LMC supernovae as well as their historical counterparts in the Milky Way Galaxy (MWG). The combined spectrum of echo light from SNR 0509-675 shows broad emission and absorption lines consistent with a supernova (SN) spectrum. We create a spectral library consisting of 26 SNe Ia and 6 SN Ib/c that are time-integrated, dust-scattered by LMC dust, and reddened by the LMC and MWG. We fit these SN templates to the observed light echo spectrum using χ2\chi^2 minimization as well as correlation techniques, and we find that overluminous 91T-like SNe Ia with \dm15<0.9 match the observed spectrum best.Comment: 12 pages, 18 Figures, to be published in Ap

    Pushing the Boundaries of Conventional Core-Collapse Supernovae: The Extremely Energetic Supernova SN 2003ma

    We report the discovery of a supernova (SN) with the highest apparent energy output to date and conclude that it represents an extreme example of the Type IIn subclass. The SN, which was discovered behind the Large Magellanic Cloud at z = 0.289 by the SuperMACHO microlensing survey, peaked at M_R = -21.5 mag and only declined by 2.9 mag over 4.7 years after the peak. Over this period, SN 2003ma had an integrated bolometric luminosity of 4 x 10^51 ergs, more than any other SN to date. The radiated energy is close to the limit allowed by conventional core-collapse explosions. Optical spectra reveal that SN 2003ma has persistent single-peaked intermediate-width hydrogen lines, a signature of interaction between the SN and a dense circumstellar medium. The light curves show further evidence for circumstellar interaction, including a long plateau with a shape very similar to the classic SN IIn 1988Z -- however, SN 2003ma is ten times more luminous at all epochs. The fast velocity measured for the intermediate-width H_alpha component (~6000 km/s) points towards an extremely energetic explosion (> 10^52 ergs), which imparts a faster blast-wave speed to the post-shock material and a higher luminosity from the interaction than is observed in typical SNe IIn.     Fission of Tubular Endosomes Triggers Endosomal Acidification and Movement

    The early endosome acts as a sorting station for internalized molecules destined for recycling or degradation. While recycled molecules are sorted and delivered to tubular endosomes, residual compartments containing molecules to be degraded undergo “maturation” before final degradation in the lysosome. This maturation involves acidification, microtubule-dependent motility, and perinuclear localization. It is currently unknown how sorting and the processes of maturation cooperate with each other. Here, we show that fission of a tubular endosome triggers the maturation of the residual endosome, leading to degradation. Use of the dynamin inhibitor dynasore to block tubular endosome fission inhibited acidification, endosomal motility along microtubules, perinuclear localization, and degradation. However, tubular endosome fission was not affected by inhibiting endosomal acidification or by depolymerizing the microtubules. These results demonstrate that the fission of recycling tubules is the first important step in endosomal maturation and degradation in the lysosome.     The First Galaxy Cluster Discovered by the VISTA Variables in the Vía Láctea Survey

    We report the first confirmed detection of the galaxy cluster VVV-J144321-611754 at very low latitudes (l = 315.°836, b = -1.°650) located in the tile d015 of the VISTA Variables in the Vía Láctea (VVV) survey. We defined the region of 30 ×30 arcmin 2 centered in the brightest galaxy finding 25 galaxies. For these objects, extinction-corrected median colors of (H-K s ) = 0.34 ± 0.05 mag, (J-H) = 0.57 ± 0.08 mag, and (J-K s ) = 0.87 ± 0.06 mag; R 1/2 = 1.59 ± 0.″16; C = 3.01 ± 0.08; and Sérsic index n = 4.63 ± 0.39 were estimated. They were visually confirmed showing characteristics of early-type galaxies in the near-IR images. An automatic clustering analysis performed in the whole tile found that the concentration of galaxies VVV-J144321-611754 is a real, compact concentration of early-type galaxies. Assuming a typical galaxy cluster with low X-ray luminosity, the photometric redshift of the brightest galaxy is z = 0.196 ± 0.025. Follow-up near-IR spectroscopy with FLAMINGOS-2 at the Gemini-South telescope revealed that the two brighter cluster galaxies have typical spectra of early-type galaxies and the estimated redshift for the brightest galaxy VVV-J144321.06-611753.9 is z = 0.234 ± 0.022 and that for VVV-J144319.02-611746.1 is z = 0.232 ± 0.019. Finally, these galaxies clearly follow the cluster red sequence in the rest-frame near-IR color-magnitude diagram with a slope similar to a galaxy cluster at a redshift of 0.2.     A Missense Mutation in a Highly Conserved Alternate Exon of Dynamin-1 Causes Epilepsy in Fitful Mice

    Dynamin-1 (Dnm1) encodes a large multimeric GTPase necessary for activity-dependent membrane recycling in neurons, including synaptic vesicle endocytosis. Mice heterozygous for a novel spontaneous Dnm1 mutation—fitful—experience recurrent seizures, and homozygotes have more debilitating, often lethal seizures in addition to severe ataxia and neurosensory deficits. Fitful is a missense mutation in an exon that defines the DNM1a isoform, leaving intact the alternatively spliced exon that encodes DNM1b. The expression of the corresponding alternate transcripts is developmentally regulated, with DNM1b expression highest during early neuronal development and DNM1a expression increasing postnatally with synaptic maturation. Mutant DNM1a does not efficiently self-assemble into higher order complexes known to be necessary for proper dynamin function, and it also interferes with endocytic recycling in cell culture. In mice, the mutation results in defective synaptic transmission characterized by a slower recovery from depression after trains of stimulation. The DNM1a and DNM1b isoform pair is highly conserved in vertebrate evolution, whereas invertebrates have only one isoform. We speculate that the emergence of more specialized forms of DNM1 may be important in organisms with complex neuronal function

    Scattered-Light Echoes from the Historical Galactic Supernovae Cassiopeia A and Tycho (SN 1572)

    We report the discovery of an extensive system of scattered light echo arclets associated with the recent supernovae in the local neighbourhood of the Milky Way: Tycho (SN 1572) and Cassiopeia A. Existing work suggests that the Tycho SN was a thermonuclear explosion while the Cas A supernova was a core collapse explosion. Precise classifications according to modern nomenclature require spectra of the outburst light. In the case of ancient SNe, this can only be done with spectroscopy of their light echo, where the discovery of the light echoes from the outburst light is the first step. Adjacent light echo positions suggest that Cas A and Tycho may share common scattering dust structures. If so, it is possible to measure precise distances between historical Galactic supernovae. On-going surveys that alert on the development of bright scattered-light echo features have the potential to reveal detailed spectroscopic information for many recent Galactic supernovae, both directly visible and obscured by dust in the Galactic plane.Comment: 4 pages, 4 color figures, accepted for publication in ApJ

    A critical role for endocytosis in Wnt signaling

    BACKGROUND: The Wnt signaling pathway regulates many processes during embryonic development, including axis specification, organogenesis, angiogenesis, and stem cell proliferation. Wnt signaling has also been implicated in a number of cancers, bone density maintenance, and neurological conditions during adulthood. While numerous Wnts, their cognate receptors of the Frizzled and Arrow/LRP5/6 families and downstream pathway components have been identified, little is known about the initial events occurring directly after receptor activation. RESULTS: We show here that Wnt proteins are rapidly endocytosed by a clathrin- and dynamin-mediated process. While endocytosis has traditionally been considered a principal mechanism for receptor down-regulation and termination of signaling pathways, we demonstrate that interfering with clathrin-mediated endocytosis actually blocks Wnt signaling at the level of β-catenin accumulation and target gene expression. CONCLUSION: A necessary component of Wnt signaling occurs in a subcellular compartment distinct from the plasma membrane. Moreover, as internalized Wnts transit partially through the transferrin recycling pathway, it is possible that a "signaling endosome" serves as a nexus for activated Wnt pathway components
