Gene expression of bacterial collagenolytic proteases in root caries

Objective: It is unknown whether bacteria play a role in the collagen matrix degradation that occurs during caries progression. Our aim was to characterize the expression level of genes involved in bacterial collagenolytic proteases in root biofilms with and without caries. Method: we collected samples from active cavitated root caries lesions (RC, n = 30) and from sound root surfaces (SRS, n = 10). Total microbial RNA was isolated and cDNA sequenced on the Illumina Hi-Seq2500. Reads were mapped to 162 oral bacterial reference genomes. Genes encoding putative bacterial collagenolytic proteases were identified. Normalization and differential expression analysis was performed on all metatranscriptomes (FDR8) but none in SRS were Pseudoramibacter alactolyticus [HMPREF0721_RS02020; HMPREF0721_RS04640], Scardovia inopinata [SCIP_RS02440] and Olsenella uli DSM7084 [OLSU_RS02990]. Conclusion: Our findings suggest that the U32 proteases may be related to carious dentine. The contribution of a small number of species to dentine degradation should be further investigated. These proteases may have potential in future biotechnological and medical applications, serving as targets for the development of therapeutic agents

Review of Matrix Metalloproteinases’ Effect on the Hybrid Dentin Bond Layer Stability and Chlorhexidine Clinical Use to Prevent Bond Failure

This review describes the relationship between dentin collagen hybrid bond layer degradation and the Matrix Metalloproteinases (MMPs) after their release by acid etch and rinse adhesives and self etching bonding adhesives that can reduce the bond stability over time. MMP-2, MMP-8 and MMP-9 are indicated as the active proteases that breakdown the collagen fibrils in the hybrid bond layer. Phosphoric acid in the acid etch and rinse bonding process and acid primers in the self etch process are implicated in the release of these proteases and their activation by several non-collagen proteins also released from dentin by the etching. MMPs are released in saliva by salivary glands, by cells in the gingival crevices to crevicular fluid and by pulpal odontoblasts cells to the dentinal fluids. These sources may affect the hybrid layer also. Evidence of the bond strength deterioration over time and the ability of Chlorhexidine to prevent bond deterioration by inhibiting MMP action are discussed. Dentin Bonding procedure utilizing Chlorhexidine for different application times and concentrations are being developed. The application of 2% Chlorhexidine to the phosphoric acid etch surface after rinsing off the acid is the only procedure that has been clinically tested for a longer period of time and shown to prevent bond strength degradation so far. The adoption of this procedure is recommended as means of improving bond stability at this time

Alkusanat Lähivertailuja 14 on Oulun yliopistossa 3.—4. toukokuuta 2003 pidetyn suomalais-virolaisen kontrastiivisen seminaarin satoa. Aurinkoiset kevätpäivät loivat oivat puitteet suomen ja viron kielestä ja kielten vertailusta kiinnostuneille. Seminaariin osallistui myös suomen ja viron kielen opiskelijoita ja muita alan harras-tajia. Seminaarin ohjelma on liitteenä.

Abstract This report is the second publication of the co-operative project Nordic-Baltic-Russian network of Finnic minority and regional languages funded by the Nordic Council of Ministers. The project participants are the Universities of Tromsø, Umeå, Stockholm, Mälardalen, Petrozavodsk, Tartu and Oulu, Võro Institute, Kväntunet, STR-T, Ruotsinsuomalaisten Keskusliitto, Karjalan Rahvahan Liitto and Võro Selts, and the project aims to follow up and support the revival and revitalization of Finnic minority languages. The project was launched in the autumn 2004, and it is coordinated by the Division of Finnish at the University of Oulu. The purpose of the network project is to promote the use of Finnic minority languages in all domains, to increase co-operation between universities and the third sector, to continue the long-term research and educational co-operation between the participating universities and to support the regional education initiated by minority organizations. The project is a continuation of the previous co-operative Academic networking in peripheral language research and higher education between the Nordic countries and the neighbouring states 1999–2001. The crucial difference compared to the aforesaid project is that the current project involves the organizations and institutes of linguistic minorities as equal participants. The previous project published a baseline report titled Kieli- ja kulttuurikontaktit and three reports under the title Tutkielmia vähemmistökielistä Jäämereltä Liivinrantaan. The current project published report IV in 2005, and this report is thus report V of the network for minority language research and education. Project publications are significant in two ways. The section of research articles is a forum for ongoing research on Finnic minority languages. It allows especially junior researches to present aspects of their doctoral dissertations. More senior researchers can also report the results of their minority projects in this section. The other section of the publication reports on the current situation of minority languages. The present report focuses especially on the themes of the Vaskna (Estonia) and Petrozavodsk conferences: the contemporary status and developmental prospects of minority language instruction as well as the operation of minority organizations

Abstract Tutkielmia oppijankielestä III (Studies on learner language III) is an addition to the series of two previous publications from the Department of Finnish Language, University of Oulu, titled Tutkielmia oppijankielestä (Studies on learner language) (2000) and Tutkielmia oppijankielestä II (Studies on learner language II) (2002). The articles in this publication are based on pro gradu theses on Finnish as a second and foreign language (S2). They focus on the communication strategies used by the learners of Finnish, the teaching and learning of spoken Finnish, the learning of the structure and lexicon of Finnish, and the order of teaching in textbooks. The articles have been reviewed by experts in the field, and edited and improved based on their recommendations. The last section of the book contains a historical review of the development of Finnish as a second and foreign language as a subject in the university of Oulu and a list of the S2 student assignments
