    Adhesion and invasion of bovine endothelial cells by Neospora caninum

    Neospora caninum is a recently identified coccidian parasite which was, until 1988, misdiagnosed as Toxoplasma gondii. It causes paralysis and death in dogs and neonatal mortality and abortion in cattle, sheep, goats and horses. The life-cycle of Neospora has not yet been elucidated. The only two stages identified so far are tissue cysts and intracellularly dividing tachyzoites. Very little is known about the biology of this species. We have set up a fluorescence-based adhesion/invasion assay in order to investigate the interaction of N. caninum tachyzoites with bovine aorta endothelial (BAE) cells in vitro. Treatment of both host cells and parasites with metabolic inhibitors determined the metabolic requirements for adhesion and invasion. Chemical and enzymatic modifications of parasite and endothelial cell surfaces were used in order to obtain information on the nature of cell surface components responsible for the interaction between parasite and host. Electron microscopical investigations defined the ultrastructural characteristics of the adhesion and invasion process, and provided information on the intracellular development of the parasite

    Identification and characterization of two repetitive non-variable antigens from African trypanosomes which are recognized early during infection

    The present paper describes two repetitive proteins representing common antigens of African trypanosomes which are non-variant and which are recognized early in infection by the host immune system. These antigens were identified by their ability to immunoreact with bovine serum taken during the early phase of a cyclic trypanosomal infection. Screening of a cDNA library from T. b. gambiense with such early infection serum identified a protein which contains a repetitive motif consisting of 68 amino acid repeat units (GM6). Immunofluorescence and immunogold electron microscopy revealed that GM6 is located on fibres which connect the microtubules of the membrane skeleton with the flagellum. A second repetitive antigen detected by this serum is MARP1 (microtubule-associated repetitive protein 1), a protein previously characterized in this laboratory as a high-molecular weight component of the membrane skeleton, which consists of more than 50 tandemly repeated, near-identical 38 amino acid repeat units. Beta-galactosidase fusion products of both proteins demonstrated a strong immunoreactivity with sera from T. b. brucei and T. congolense-infected cattle. The result from this preliminary immunological evaluation indicates a high immunodiagnostic sensititivy (90%) of the two recombinant antigens which make them interesting candidates for immunodiagnosis of trypanosomiasis in cattl

    The program was written for the Philco-2000 computer to provide a nuclear design computing tool equivalent to the IBM-704 computer program MUFT-4 and to provide a set of routines for a future spatial multigroup program, P/sub 3/ MG-1. In addition to the features of its 704 predecessor, MUFT-5 provides a more complete isotopic edit, an optional blackness coefficient edit, and the use of a complete P/sub i/->i>s library.     In vitro effects of arylimidamides against Besnoitia besnoiti infection in Vero cells

    The in vitro effects of 4 arylimidamides (DB811, DB786, DB750 and DB766) against the proliferative tachyzoite stage of the apicomplexan parasite Besnoitia besnoiti were investigated. These four compounds had been shown earlier to exhibit in vitro activities in the nanomolar range against the related apicomplexans Neospora caninum and Toxoplasma gondii. Real-time-PCR was used to assess B. besnoiti intracellular proliferation in vitro. Preliminary assessment by light microscopy identified DB811 and DB750 as the most promising compounds, while DB786 and DB766 were much less effective. Three-day-growth assays and quantitative real-time PCR was used for IC50 determination of DB811 (0·079 μM) and DB750 (0·56 μM). Complete growth inhibition was observed at 1·6 μM for DB 811 and 1·7 μM for DB750. However, when infected cultures were treated for 14 days, proliferation of parasites occurred again in cultures treated with DB750 from day 4 onwards, while the proliferation of DB811-treated tachyzoites remained inhibited. Electron microscopy of B. besnoiti-infected fibroblast cultures fixed and processed at different time-points following the initiation of drug treatments revealed that DB811 exerted a much higher degree of ultrastructural alterations compared to DB750.     Social media are implicated in many of contemporary society's most pressing issues, from influencing public opinion, to organizing social movements, and identifying economic trends. Increasing the capacity of researchers to understand the dynamics of such social, behavioral and economic phenomena will depend on reliable, curated, discoverable and accessible social media data. To that end, ICPSR will develop a new archive of curated datasets, workflows, and code for use by social science researchers for the empirical analysis of social media platforms, content, and user behavior. The goal is to provide a user-friendly, large-scale, next-generation data resource for researchers conducting data-intensive research using data from social media platforms such as Facebook, Twitter, Reddit, and Instagram.     Fish Colonization of a Newly Deployed Vessel-reef off Southeast Florida: Preliminary Results

    Fish colonization on the Ebenezer II, a 25.5m merchant marine vessel, was studied from May 2002 - July 2003. The ship was scuttled in May 2002 off Broward County, Florida at a depth of 21m and was censused 10 times during the study period using a modified Bohnsack and Bannerot visual census method. Adjacent natural reefs and the Mcallister, a nearby, 30m tugboat deployed in June 1998, were censused during the same period. Distinct changes in the fish assemblage on the Ebenezer II were observed throughout the sample period. A pioneer assemblage was observed during the first three months, characterized by the settlement of juvenile fishes «5 cm). Subsequently, numbers of juveniles decreased either through emigration, predation or growth. Resident species made up 52.5% of the total abundance but transient fish species made up 78% of the total fish biomass during the study period. Surprisingly, attraction of adult fish from both natural reefs and the Mcallister was not a major factor in assemblage fonnation. The primary adult fishes attracted to the Ebenezer II were herbivores. These fishes steadily increased in abundance throughout the study period, presumably due to increased food availability as benthic algal communities developed. A similar trend of increasing herbivores with increasing soak time was observed on the Spiegel Grove, a 153m vessel-reef sunk off Key Largo in May 2002. The fish assemblages on the artificial reefs were more similar to each other than to natural reefs. Vessel-reefs had sixty species in common, while the Ebenezer II only had thirty-nine species in common with natural reefs. Several species common to vessel-reefs were absent or rare on nearby natural reefs. This may indicate that vessel-reefs are providing early juvenile and adult habitat that is not available on natural reefs

    Isolation of Besnoitia besnoiti from infected cattle in Portugal

    Abstract Besnoitia besnoiti, an obligate intracellular protozoan parasite belonging to the phylum apicomplexa, is the causative agent of bovine besnoitiosis. Besnoitiosis is responsible for significant losses in the cattle industry of Africa and Mediterranean countries due to the high morbidity rate, abortion and infertility in males. The acute stage of disease is associated with the proliferative forms (tachyzoites) and is characterized by fever, whimpery, general weakness and swelling of the superficial lymph nodes. During the following chronic stage, a huge number of cysts are formed mainly in the subcutaneous tissues. This process is non-reversible, and chronic besnoitiosis is characterized by hyper-sclerodermia, hyperkeratosis, alopecia and, in bulls, atrophy, sclerosis and focal necrosis that cause irreversible lesions in the testis. In this paper we report on the identification of large cysts in the skin of a cow and a bull in Portugal, which presented loss of hair and enlargement and pachydermis all over the body. The observation of a two-layered cyst wall within the host cell, the encapsulation of the host cell by a large outer cyst wall, and the subcutaneous localization of the cysts within the host, were characteristic for B. besnoiti. The parasites were isolated from the infected animals and successfully propagated in Vero cells without prior passages in laboratory animals. Morphological characterization of B. besnoiti tachyzoites and the amplification of the 149 bp segment from the internal transcribed spacer 1 (ITS1), aided with specific primers, confirmed the identification of B. besnoiti.     20th to 21st Century Relative Sea and Land Level Changes in Northern California: Tectonic Land Level Changes and their Contribution to Sea-Level Rise, Humboldt Bay Region, Northern California

    Sea-level changes are modulated in coastal northern California by land-level changes due to the earthquake cycle along the Cascadia subduction zone, the San Andreas plate boundary fault system, and crustal faults. Sea-level rise (SLR) subjects ecological and anthropogenic infrastructure to increased vulnerability to changes in habitat and increased risk for physical damage. The degree to which each of these forcing factors drives this modulation is poorly resolved. We use NOAA tide gage data and ‘campaign’ tide gage deployments, Global Navigation Satellite System (GNSS) data, and National Geodetic Survey (NGS) first-order levelling data to calculate vertical land motion (VLM) rates in coastal northern California. Sea-level observations, highway level surveys, and GNSS data all confirm that land is subsiding in Humboldt Bay, in contrast to Crescent City where the land is rising. Subtracting absolute sea-level rate (~1.99 mm/year) from Crescent City (CC) and North Spit (NS) gage relative sea-level rates reveals that CC is uplifting at ~2.83 mm/year and NS is subsiding at ~3.21mm/year. GNSS vertical deformation reveals similar rates of ~2.60 mm/year of uplift at Crescent City. In coastal northern California, there is an E-W trending variation in vertical land motion that is primarily due to Cascadia megathrust fault seismogenic coupling. This interseismic subsidence also dominates the N-S variation in vertical land motion in most of the study region. There exists a second-order heterogeneous N-S trend in vertical land motion that we associate to crustal fault-related strain. There may be non-tectonic contributions to the observed VLM rates
