    Multiple equilibria describe the complete adsorption isotherms of nonporous, microporous, and mesoporous adsorbents

    The adsorption of simple gases begins with the formation of a monolayer on the pristine surface, not always followed by the formation of a second or more monolayers. Subsequently, cluster formation or cavity filling occurs, depending on the properties of the surface. The characteristically different shape of the isotherms related to these processes allows to clearly differentiate them. We analyzed argon and N2 adsorption isotherms quantitatively over the entire relative pressure range for adsorbents bearing different properties: the nonporous Stöber-type particles, the microporous zeolite L (ZL) and zeolite L filled with indigo (Indigo-ZL), and three mesoporous silica adsorbents of different pore size. The formal equilibria involved in cluster formation and in cavity filling have been derived and successfully applied to quantitatively describe the isotherms of the adsorbents. No indication regarding formation of a second monolayer on top of the first one was observed for the Stöber-type particles. Instead, cluster generation, which minimizes surface tension, starts early. The behavior of microporous ZL and of Indigo-ZL is different. A second monolayer sets up and cluster formation starts with some delay. The enthalpy of cluster formation is, however, practically identical with that seen for the Stöber-type particles. The difference between the experimental and the calculated inflection points is very small. The shapes of the isotherms seen for the mesoporous adsorbents differ significantly from those seen for the nonporous and for the microporous adsorbents. The quantitative analysis of the data proves that formation of a second monolayer is followed by filling of cavities which ends as soon as all cavity sites are filled. The sum of the individual fractional contributions, namely the monolayer formation ΘmL, the appearance of a second monolayer Θ2L on top of the first one, and the cavity filling, yields a calculated adsorption isotherm Θcalc which describes the experimental data Θexp well. The experimental and the calculated first inflection points are in excellent agreement, which is also the case for the second inflection points. The value of the cavity filling enthalpy is roughly 10% larger than that for the cluster formation seen in the nonporous and the microporous adsorbents. The volume for cavity filling is significantly smaller than the monolayer volume for the mesoporous adsorbent with a pore diameter of 2.7 nm, while it is the same or larger for pore diameters of 4.1 nm and 4.4 nm, respectively. We conclude that understanding the adsorption isotherms as signature of several sequential chemical equilibrium steps provides additional information data for clusters, cavities, and position of the inflection points, not accessible by means of the conventional models.     Multiple equilibria description of type H1 hysteresis in gas sorption isotherms of mesoporous materials

    We report argon adsorption/desorption isotherms of MCM-41 and of SBA-15. The shape of all hysteresis loops we have observed corresponds to type H1. The data have been analyzed quantitatively using the multiple equilibria description and applying the notion of metastable thermodynamic equilibrium. It is remarkable and important that for both mesoporous materials the desorption process can be understood according to this description but with a corresponding equilibrium constant. This procedure therefore allows determining the thermodynamic values for the enthalpy and the free enthalpy of cavity desorption and thus to obtain information not available so far. We observed that the MCM-41 adsorption isotherms show first an increase with the characteristic Langmuir shape, followed by the almost instantaneous filling of cavities that ends as soon as all cavities are completely filled. The SBA-15 isotherms show a characteristic Langmuir shape at low relative pressure. The pressure slightly below the inflection point at prel, infl = 0.312 marks the beginning of the growth of a second layer, before an almost instantaneous filling of cavities takes place. It is interesting to observe that the values of the enthalpy and the free enthalpy for cavity filling and cavity desorption differ by about 0.1 kJ/mol for both, MCM-41 and SBA-15. This means that the driving force for developing a hysteresis is small. The hysteresis loop is therefore driven by delicate changes occurring within the cavities, partially or completely filled by the adsorbate. The monolayer coverage and monolayer desorption processes are decoupled from the cavity filling and cavity emptying processes. Knowing thermodynamic parameters for the hysteresis loop ultimately helps to better characterize and understand the experimental observations

    A subset of ocular adnexal marginal zone lymphomas may arise in association with IgG4-related disease

    We previously suggested a relationship between ocular immunoglobulin (Ig)G4-related disease (IgG4-RD) and marginal zone lymphomas (MZLs). However, the cytokine background associated with these disorders and whether it differs between ocular adnexal MZLs with (IgG4-associated MZL) and without (IgG4-negative MZL) numerous IgG4+ plasma cells are unknown. In this study, we identified the mRNA expression pattern of Th2 and regulatory T-cell (Treg) cytokines in IgG4-RD and in IgG4-associated MZL and IgG4-negative MZL using real-time polymerase chain reaction analysis. Ocular IgG4-RD and IgG4-associated MZL exhibited significantly higher expression ratios of interleukin (IL)-4/ÎČ-actin, IL-10/ÎČ-actin, IL-13/ÎČ-actin, transforming growth factor (TGF) ÎČ1/ÎČ-actin, and FOXP3/ÎČ-actin than did IgG4-negative MZL (p < 0.05). This finding further supports our prior observations that a significant subset of ocular MZLs arises in the setting of IgG4-RD. Furthermore, the presence of a different inflammatory background in IgG4-negative MZLs suggests that IgG4-associated MZLs may have a different pathogenesis

    The neurological level of spinal cord injury and cardiovascular risk factors: a systematic review and meta-analysis.

    STUDY DESIGN Systematic review and meta-analysis. OBJECTIVE To determine the difference in cardiovascular risk factors (blood pressure, lipid profile, and markers of glucose metabolism and inflammation) according to the neurological level of spinal cord injury (SCI). METHODS We searched 5 electronic databases from inception until July 4, 2020. Data were extracted by two independent reviewers using a pre-defined data collection form. The pooled effect estimate was computed using random-effects models, and heterogeneity was calculated using I2 statistic and chi-squared test (CRD42020166162). RESULTS We screened 4863 abstracts, of which 47 studies with 3878 participants (3280 males, 526 females, 72 sex unknown) were included in the meta-analysis. Compared to paraplegia, individuals with tetraplegia had lower systolic and diastolic blood pressure (unadjusted weighted mean difference, -14.5 mmHg, 95% CI -19.2, -9.9; -7.0 mmHg 95% CI -9.2, -4.8, respectively), lower triglycerides (-10.9 mg/dL, 95% CI -19.7, -2.1), total cholesterol (-9.9 mg/dL, 95% CI -14.5, -5.4), high-density lipoprotein (-1.7 mg/dL, 95% CI -3.3, -0.2) and low-density lipoprotein (-5.8 mg/dL, 95% CI -9.0, -2.5). Comparing individuals with high- vs. low-thoracic SCI, persons with higher injury had lower systolic and diastolic blood pressure (-10.3 mmHg, 95% CI -13.4, -7.1; -5.3 mmHg 95% CI -7.5, -3.2, respectively), while no differences were found for low-density lipoprotein, serum glucose, insulin, and inflammation markers. High heterogeneity was partially explained by age, prevalent cardiovascular diseases and medication use, body mass index, sample size, and quality of studies. CONCLUSION In SCI individuals, the level of injury may be an additional non-modifiable cardiovascular risk factor. Future well-designed longitudinal studies with sufficient follow-up and providing sex-stratified analyses should confirm our findings and explore the role of SCI level in cardiovascular health and overall prognosis and survival

    Tumor markers in breast cancer - European Group on Tumor Markers recommendations

    Recommendations are presented for the routine clinical use of serum and tissue-based markers in the diagnosis and management of patients with breast cancer. Their low sensitivity and specificity preclude the use of serum markers such as the MUC-1 mucin glycoproteins ( CA 15.3, BR 27.29) and carcinoembryonic antigen in the diagnosis of early breast cancer. However, serial measurement of these markers can result in the early detection of recurrent disease as well as indicate the efficacy of therapy. Of the tissue-based markers, measurement of estrogen and progesterone receptors is mandatory in the selection of patients for treatment with hormone therapy, while HER-2 is essential in selecting patients with advanced breast cancer for treatment with Herceptin ( trastuzumab). Urokinase plasminogen activator and plasminogen activator inhibitor 1 are recently validated prognostic markers for lymph node-negative breast cancer patients and thus may be of value in selecting node-negative patients that do not require adjuvant chemotherapy.     Wheeze in Preschool Age Is Associated with Pulmonary Bacterial Infection and Resolves after Antibiotic Therapy

    BACKGROUND: Neonates with airways colonized by Haemophilus influenzae, Streptococcus pneumoniae or Moraxella catarrhalis are at increased risk for recurrent wheeze which may resemble asthma early in life. It is not clear whether chronic colonization by these pathogens is causative for severe persistent wheeze in some preschool children and whether these children might benefit from antibiotic treatment. We assessed the relevance of bacterial colonization and chronic airway infection in preschool children with severe persistent wheezing and evaluated the outcome of long-time antibiotic treatment on the clinical course in such children. METHODOLOGY/PRINCIPAL FINDINGS: Preschool children (n = 42) with severe persistent wheeze but no symptoms of acute pulmonary infection were investigated by bronchoscopy and bronchoalveolar lavage (BAL). Differential cell counts and microbiological and virological analyses were performed on BAL samples. Patients diagnosed with bacterial infection were treated with antibiotics for 2-16 weeks (n = 29). A modified ISAAC questionnaire was used for follow-up assessment of children at least 6 months after bronchoscopy. Of the 42 children with severe wheezing, 34 (81%) showed a neutrophilic inflammation and 20 (59%) of this subgroup had elevated bacterial counts (≄ 10⁎ colony forming units per milliliter) suggesting infection. Haemophilus influenzae, Streptococcus pneumoniae and Moraxella catarrhalis were the most frequently isolated species. After treatment with appropriate antibiotics 92% of patients showed a marked improvement of symptoms upon follow-up examination. CONCLUSIONS/SIGNIFICANCE: Chronic bacterial infections are relevant in a subgroup of preschool children with persistent wheezing and such children benefit significantly from antibiotic therapy

    Comprehensive genetic dissection of wood properties in a widely-grown tropical tree: Eucalyptus

    Background: Eucalyptus is an important genus in industrial plantations throughout the world and is grown for use as timber, pulp, paper and charcoal. Several breeding programmes have been launched worldwide to concomitantly improve growth performance and wood properties (WPs). In this study, an interspecific cross between Eucalyptus urophylla and E. grandis was used to identify major genomic regions (Quantitative Trait Loci, QTL) controlling the variability of WPs. Results: Linkage maps were generated for both parent species. A total of 117 QTLs were detected for a series of wood and end-use related traits, including chemical, technological, physical, mechanical and anatomical properties. The QTLs were mainly clustered into five linkage groups. In terms of distribution of QTL effects, our result agrees with the typical L-shape reported in most QTL studies, i.e. most WP QTLs had limited effects and only a few (13) had major effects (phenotypic variance explained &gt; 15%). The co-locations of QTLs for different WPs as well as QTLs and candidate genes are discussed in terms of phenotypic correlations between traits, and of the function of the candidate genes. The major wood property QTL harbours a gene encoding a Cinnamoyl CoA reductase (CCR), a structural enzyme of the monolignol-specific biosynthesis pathway. Conclusions: Given the number of traits analysed, this study provides a comprehensive understanding of the genetic architecture of wood properties in this Eucalyptus full-sib pedigree. At the dawn of Eucalyptus genome sequence, it will provide a framework to identify the nature of genes underlying these important quantitative traits. (Résumé d'auteur

    Evidence of intense chromosomal shuffling during conifer evolution

    Although recent advances have been gained on genome evolution in angiosperm lineages, virtually nothing is known about karyotype evolution in the other group of seed plants, the gymnosperms. Here, we used high-density gene-based linkagemapping to compare the karyotype structure of two families of conifers (the most abundant group of gymnosperms) separated around 290 Ma Pinaceae and Cupressaceae.We propose for the first time amodel based on the fusion of 20 ancestral chromosomal blocks that may have shaped the modern karyotpes of Pinaceae (with n=12) and Cupressaceae (with n=11). The considerable difference in modern genome organization between these two lineages contrasts strongly with the remarkable level of synteny already reported within the Pinaceae. It also suggests a convergent evolutionary mechanism of chromosomal block shuffling that has shaped the genomes of the spermatophytes. ©The Author(s) 2015.     Time at surgery during menstrual cycle and menopause affects pS2 but not cathepsin D levels in breast cancer

    Many studies have addressed the clinical value of pS2 as a marker of hormone responsiveness and of cathepsin D (Cath D) as a prognostic factor in breast cancer. Because pS2 and Cath D are both oestrogen induced in human breast cancer cell lines, we studied the influence of the menstrual cycle phase and menopausal status at the time of surgery on the levels of these proteins in breast cancer. A population of 1750 patients with breast cancer, including 339 women in menstrual cycle, was analysed. Tumoral Cath D and pS2 were measured by radioimmunoassay. Serum oestradiol (E2), progesterone (Pg), follicle-stimulating hormone (FSH) and luteinizing hormone (LH) levels at the day of surgery were used to define the hormonal phase in premenopausal women. There was a trend towards a higher mean pS2 level in the follicular phase compared with the luteal phase (17 ng mg−1and 11 ng mg−1respectively, P= 0.09). Mean pS2 was lower in menopausal patients than in women with cycle (8 ng mg−1and 14 ng mg−1respectively, P= 0.0001). No differences in mean Cath D level were observed between the different phases of the menstrual cycle, or between pre- and post-menopausal women. In the overall population, pS2 was slightly positively associated with E2 and Pg levels and negatively associated with FSH and LH, probably reflecting the link between pS2 and menopausal status. In premenopausal women, no association was found between pS2 and E2, Pg, FSH or LH levels. There were no correlations between Cath D level and circulating hormone levels in the overall population. However, in the subgroup of premenopausal women with ER-positive (ER+) tumours, E2 was slightly associated with both pS2 and Cath D, consistent with oestrogen induction of these proteins in ER+ breast cancer cell lines. There are changes in pS2 level in breast cancer throughout the menstrual cycle and menopause. This suggests that the choice of the pS2 cut-off level should take the hormonal status at the time of surgery into account.
