    Síndrome do cão tremedor

    Background:  :  :  : The Shaker Dog Syndrome manifests itself as generalized tremors that usually affect the head and body of the patient. The tremors increase with movement and decrease at rest, and it may cease during sleep. The disorder develops mostly in 1-year to 5-year-old animals weighing below 15 kg. The diagnosis is based on the exclusion of other possible causes of tremors and a positive response to treatment with corticosteroids. The treatment consists of immunosuppressant doses of corticosteroids and it can be associated to decreasing diazepam doses during 8 to 12 weeks. Literature data are very rare and cannot be found in Brazil. Consequently, this study aimed at describing a Shaker Dog Syndrome case that is responsive to corticosteroids. Case: A 2-year-old mongrel female dog, weighting 7.5 kg, was referred to the Hospital de Clínicas Veterinárias da Universidade Federal do Rio Grande do Sul, presenting generalized tremors for 5 days, which would cease during sleep. In the neurological examination, the cranial nerve exam and the test for postural reactions were considered normal. The hematological and biochemical profile did not show alterations. The cervical spine radiography did not display bone alterations. A density of 1.016 was observed at the physical examination of the cerebrospinal fluid. The chemical examination revealed a glucose level of 100 mg/dL, negative bilirubin, negative ketones, absence of occult blood, pH 8.0, presence of protein traces, normal urobilinogen, negative reactive protein C (PCR), normal creatine kinase (CK) and aspartate aminotransferase (AST). The result of Pandy’s test was negative. Oral treatment with prednisone at a dose of 1.5 mg.kg-1 BID and diazepam at a dose of 0.5 mg.kg-1 BID was instituted, this latter during 4 days. The corticoid dose was maintained until total disappearance of the tremors, which occurred within 24 days. Afterwards, a gradual reduction of 50% of the dose was made every 7 days, during 4 weeks. Discussion: During anamnesis, tremors induced by trauma and medication were ruled out. Myelin abnormalities were also improbable, as they cause congenital tremors and do not quickly respond to corticosteroids. A colorless aspect and absence of occult blood and bilirubin was observed in the LCR analysis. The absence of turbidity and the presence of protein traces in this sample demonstrate normality (38.7 mg/dL ± 15.49). Chemical and physical exams did not demonstrate alterations – pH 8.0 (8.30 ± 0.34) and a density of 1.016 (1007 ± 1.83), respectively. The glucose concentration in the cerebrospinal fluid was not reduced. The results obtained from Pandy’s and reactive protein C tests were negative. The analysis of AST and CK enzymes was normal. Since the values obtained from enzymatic and protein evaluations of the cerebrospinal fluid were normal, they suggest an absence of infectious and degenerative processes. The immunosuppressant prednisone dose recommended by literature is variable; thus, the smallest corticosteroid dose indicated was selected, which is 1.5 mg.kg-1 BID. An association with diazepam was made to reduce tremors. Usually, clinical improvement can occur within a few days, but the remission of clinical signs can take weeks. In the present case, the reduction of tremors occurred on the fourth day, with the complete disappearance in 24 days, without recurrence within a clinical follow-up period of 2 years

    Avaliação do endotélio corneano suíno por microscopia eletrônica de varredura após aplicação de azul brilhante a 0,05% na câmara anterior : estudo in vitro

    The aim was to investigate the ultrastructural changes in the corneal endothelium of pigs induced by intracameral 0.05% brilliant blue. Twenty swine corneas were separated into two groups, the right eye bulbs (control group) and the left eye bulbs (experimental group) of the same animal. All the eye bulbs were evaluated with specular microscopy. The cornea of the right eye bulbs was excised and in the left eye bulbs 0.2ml of 0.05% brilliant blue vital dye (OPTH-blue®) was injected into the anterior chamber, where it remained for one minute. Then the anterior chamber was cleaned with a balanced salt solution injection and the cornea was excised too. All the corneas were evaluated by scanning electron microscopy to evaluate the changes on the endothelium caused by the brilliant blue dye. There were no significant differences between the right corneal endothelium cells and the left corneal endothelium cells with scanning electron microscopy after intracameral use of 0.05% brilliant blue dye. The 0.05% brilliant blue dye concentration did not cause deleterious effects for the swine corneal endothelium after intracameral use and can be a choice for safe staining of the anterior capsule of the lens in cataract surgery.[Avaliação do endotélio corneano suíno por microscopia eletrônica de varredura após aplicação de azul brilhante a 0,05% na câmara anterior: estudo in vitro.] Com o objetivo de avaliar as alterações ultraestruturais no endotélio corneano suíno induzidas pela aplicação intracameral do corante azul brilhante 0,05%, vinte córneas suínas foram separadas em dois grupos: olhos direitos (grupo controle) e olhos esquerdos (grupo experimental). Inicialmente todos os bulbos oculares foram avaliados por microscopia especular. As córneas dos bulbos oculares do grupo controle foram excisadas, enquanto nos bulbos oculares do grupo experimental foram injetados 0,2ml do corante vital azul brilhante 0,05% na câmara anterior e mantido por um minuto. Após esse período a câmara anterior foi lavada com solução salina balanceada e as córneas foram excisadas. Todas as córneas excisadas foram avaliadas por microscopia eletrônica de varredura para verificar a ocorrência possíveis alterações no endotélio corneano causadas pelo corante. Não foram observadas diferenças ultraestruturais durante a avaliação endotelial por microscopia eletrônica de varredura das córneas do grupo controle e experimental após a aplicação do azul brilhante 0,05% na câmara anterior. O corante azul brilhante na concentração 0,05% não causou danos endoteliais após seu uso intracameral em suínos e pode ser uma alternativa segura para a coloração da cápsula anterior da lente na cirurgia de catarata

    Reparação de perfuração ocular em um cão usando pericárdio homólogo preservado em glicerina

    As lesões de córnea ocorrem frequentemente em cães. Várias técnicas cirúrgicas são propostas para reparar perfuração de córnea, incluindo flapes conjuntivais, ceratoplastias, enxertos corneanos e implantes biológicos. Um caso de perfuração ocular em um cão tratado com pericárdio homólogo conservado em glicerina é apresentado. Oito anos de idade, macho, Pit Bull foi encaminhado ao Serviço de Oftalmologia do Hospital de Clínicas Veterinárias da Universidade Federal do Rio Grande do Sul (UFRGS) com 12 horas de perfuração corneana. O exame oftálmico revelou desconforto ocular, epífora, hiperemia conjuntival e perfuração corneana central com 5mm no olho direito. A correção cirúrgica foi realizada utilizando pericárdio homólogo conservado em glicerina. O uso de pericárdio homólogo conservado em glicerina foi efetivo na reparação do defeito corneano, preservando as funções e a anatomia oculares

    Morphology of Endothelial Cells from Different Regions of the Swine Cornea

    Background: The corneal endothelium is a monolayer of polygonal cells which constitute the last layer of the cornea. The integrity of this layer is critical to cornea transparency. The characterization of normal corneal endothelial morphology is important not only to clinical evaluation but also to selection of areas of the cornea with better quality to be employed as donor tissue. The aim of the present study was to evaluate the morphology of endothelial cells from different regions of the swine cornea after alizarin red staining using optical microscopy.Materials, Methods & Results: Twenty-four healthy eyes from 12 swine Large White breed, with 14-month-old, males or females obtained from a licensed Brazilian commercial slaughterhouse were studied. Immediately after humane slaughter, the eyes were enucleated and submitted to ophthalmic examination. Eyes with signs of diseases of the anterior segment were excluded. The cornea, with 3 mm of the sclera, was removed and placed on a glass microscope slide with the endothelial side up. Four radial incisions were made in the periphery of the cornea to better accommodate the cornea on the microscope slide. Alizarin red was diluted in isotonic solution (0.2 g/100 mL) and the pH was adjusted to 4.2 with hydrochloric acid. Three drops of alizarin red were placed on the corneal endothelium. After 90 s, the dye was removed from the cornea with balanced saline solution. The corneal endothelium was examined and photographed using an optical microscope. All evaluations were performed by the same investigator. Photomicrographs were taken of central, superior, inferior, nasal and temporal corneal areas. Parameter studied included endothelial cell morphology. For the statistical analysis, was employed the ANOVA variance test (repeated measures). Differences were considered statistically significant at P < 0.05. Normal endothelium cells were mainly hexagonal (83.7%), pentagonal (7.45%) and heptagonal (8.8%), with a minimal number of cells of other shapes present. There were no significant statistical differences in the proportion of the morphology and the different regions of the cornea (P = 0.31).Discussion: Different techniques are available for the analysis of corneal endothelium, including mainly scanning electron microscopy, specular microscopy and optical microscopy. The analysis of the morphology of corneal endothelium with an optic microscope after staining with alizarin red has been described as an effective, rapid and cost-efficient method, since this dye blends the borated cells, allowing identification. In the present study, using optical microscopy and coloration with alizarin red it was possible to explore and to obtain images of the swine endothelium of all regions of the cornea.  The analysis of the cellular morphology or the percentage of hexagonal cells are among the main parameters used to evaluate the health of the corneal endothelium. In this study, the endothelium had the predominance of the hexagonal shape in all regions studied. In swine, there are no studies evaluating the shape of the endothelial cells in the five different regions of the cornea. This study has demonstrated that the parameters evaluated in swine did not differ significantly between the various places of the cornea

    Corneal pigmentation and vascularization eventually result in blindness in dogs. Pigmentary keratitis describes a relatively common presentation comprising the deposition of melanin in the cornea and conjunctival surface associated with chronic inflammation. Cryosurgery is indicated as a treatment for pigmentary keratitis in dogs. Due to melanocytes sensitivity to cold, cryosurgery is a viable treatment for severe refractory corneal pigmentation. The aim of this work was to evaluate the use of dimethyl ether in the treatment of pigmentary keratitis in 14 eyes of seven Pug dogs. Follow-up occurred after 30 days in four animals and six months in three animals. In all treated animals, there was a reduction in corneal pigmentation. Cryosurgery causes intracellular and extracellular ice crystal formation and other mechanisms that result in rupture and death of the melanocytes. The technique used was easy to perform, has a low cryogen cost and has few undesirable or serious side effects. However, after 30 days repigmentation occured in treated patients. Cryosurgery shows good results in the first four weeks, but partial recurrence occurred in all cases.A pigmentação e a vascularização da córnea eventualmente resultam em cegueira em cães. A ceratite pigmentar é descrita com uma apresentação relativamente comum que compreende a deposição de melanina na córnea e na superfície conjuntival associada à inflamação crônica. A criocirurgia é indicada como tratamento para ceratite pigmentar em cães. Devido à sensibilidade dos melanócitos ao frio, a criocirurgia é um tratamento viável para pigmentação corneana refratária grave. O objetivo deste trabalho foi avaliar o uso do éter dimetílico no tratamento da ceratite pigmentar em 14 olhos de sete cães da raça Pug. O acompanhamento foi de 30 dias em quatro animais e seis meses em três animais. Em todos os animais tratados, houve redução da pigmentação da córnea. A criocirurgia causa a formação de cristais de gelo intracelular e extracelular e outros mecanismos que resultam na ruptura e morte dos melanócitos. A técnica utilizada foi de fácil execução, baixo custo de criogenia e poucos efeitos colaterais indesejáveis ou graves. No entanto, após 30 dias, ocorreu repigmentação nos pacientes tratados. A criocirurgia apresenta bons resultados nas primeiras quatro semanas, mas ocorreu recidiva parcial em todos os casos
