    Parasitism strategies of the Fan-tailed Cuckoo Cacomantis flabelliformis

    The Fan-tailed Cuckoo Cacomantis flabelliformis inhabits thick forest, and nothing is known about how it finds the nests of its hosts, or whether it monitors the nests that it does find. We studied this cuckoo, and its hosts, for 8 breeding seasons between 2007 (August 2007–January 2008 inclusive) and 2014 (August 2014–January 2015 inclusive) on a 10-ha site in a coastal forest of south-eastern Australia, near Moruya, New South Wales. For three of these seasons, nests were monitored with cameras. The Cuckoo was recorded at the nests of only its four putative hosts at the site (White-browed Scrubwren Sericornis frontalis, Brown Thornbill Acanthiza pusilla, Superb Fairy-wren Malurus cyaneus and Variegated Fairy-wren M. lamberti). It parasitised only the nests of the White-browed Scrubwren and Brown Thornbill, but it removed either eggs or young from all other nests at which it was recorded. There was no correlation between any measure of nest activity for a host species, and parasitism of that species, and cameras at nests recorded no evidence of nest monitoring by the Cuckoo. We conclude that individual Cuckoos may be hostspecific, and that the parasitism strategy is enigmatic, but is possibly haphazard and inefficient. As a result, the Fantailed Cuckoo finds most host nests too late for successful parasitism, it spoils them to re-instigate building, and is by default a major nest-predator

    Standardization and methodological considerations for the isometric mid-thigh pull

    The isometric mid-thigh pull (IMTP) is commonly used to assess an athlete’s force generation ability. This test is highly reliable and is simple and relatively quick to perform. The data that can be determined from the force-time curves generated by the test have been shown to be closely related to performance capacities in a variety of dynamic athletic tasks. However, within the scientific literature there are inconsistencies in the data collection procedures and methods used for data analysis that may impact the resultant output and the ability to compare and generalize results. Therefore, the primary aim of this review is to identify the differences in IMTP testing procedures and data analysis techniques, while identifying the potential impact this may have on the data collected. The secondary aim is to provide recommendations for the standardization of testing procedures to ensure that future IMTP data is of maximal benefit to practitioners and researchers

    Optimization of a non-activating medium for short-term chilled storage of barramundi (Lates calcarifer) testicular spermatozoa

    Reliable short-term chilled sperm storage is a critical prerequisite to using advanced reproductive techniques for captive breeding of barramundi (Asian sea bass; Lates calcarifer). Marine Ringer's solution (MRS) is a common non-activating medium (NAM) and has previously been used to store sperm from wild-caught barramundi. However, MRS-stored spermatozoa from captive-bred barramundi were observed to lyse within 30 min incubation. Therefore, this study aimed to optimize the composition of NAM for short-term chilled storage by characterizing and mimicking the biochemical profile of seminal and blood plasma of captive-bred barramundi. To further understand the effect of each component, osmolality was first examined to determine its effect on sperm viability. Thereafter, the effects of NaHCO3, pH, and Na+ and K+ concentrations on sperm motility were investigated. Optimization of the NAM formula was achieved through iterative adaptions. The increase in NAM osmolality from 260 to 400 mOsm/kg led to a significant improvement in sperm viability. Moreover, using HEPES instead of NaHCO3 as buffering agent significantly enhanced sperm motility and velocity. As a result, sperm samples diluted with optimized NAM (185 mM NaCl, 5.1 mM KCl, 1.6 mM CaCl2·2H2O, 1.1 mM MgSO4·7H2O, 10.0 mM HEPES, 5.6 mM D+ glucose, 400 mOsm/kg, pH 7.4) and stored at 4 °C showed no significant loss in total motility for up to 48 h and retained progressive motility for up to 72 h. The optimized NAM developed in this study significantly extended the functional lifespan of spermatozoa during chilled storage, permitting the ongoing development of advanced reproductive technologies for barramundi

    The Agreement Between a Portable Contact-Mat and Force-Plates During Bilateral Vertical Jumps

Force plates are commonly used when assessing vertical jumping performance but are not always affordable or practical tools for all testing situations. Twenty-four participants volunteered to take part in a study investigating the agreement between bilateral force plates and a new commercially available contact mat that records jump height, flight-time (FT), and FT of individual limbs during both countermovement (CMJ) and squat (SJ) jumps. Each participant performed six jumps of each type while standing on a contact mat placed upon a pair of in-ground force plates. When compared to the force plate via ordinary least products regression, the contact mat agreed with force plate CMJ and SJ jump height, individual limb FT during CMJs, and left-leg FT during SJs. The bilateral contact mat provided valid assessment of individual limb FT during CMJs, but not SJs. Practitioners can therefore use a bilateral contact mat interchangeably with bilateral force plates to measure SJ and CMJ performance

    Phosphorus bioavailability in soil profiles of a long-term fertilizer experiment: The evaluation of their bioaccessibility

    Global agricultural productivity depends on the use of finite phosphorus (P) resources of which not only the topsoil, but also subsoil, can hold immense reserves. To assess potential soil contribution to plant nutrition, we compared the P status of Stagnic Cambisol profiles in experimental plots that received different P fertilizer applications (control, triple superphosphate (TSP), compost, compost+TSP) for 16 years. Sequential fractionation was combined with P K-edge X-ray absorption near edge structure (XANES) spectroscopy to identify the chemical P speciation. Fertilized topsoils (21 to 69 kg P ha-1 a-1) showed P reserves larger by a factor of 1.2 to 1.4, and subsoil P reserves larger by a factor of 1.3 to 1.5 than those of the control. P-XANES revealed the predominance of inorganic P species such as moderately labile Fe- (46 to 92%), Al- (0 to 40%), and Ca- (0 to 15%) P compounds besides organic P (0 to 13%) in all treatments. The fertilizer application slightly altered P speciation throughout the profiles, but the type of fertilizer had no significant effect on it. Optimal plant growth requirements are restricted by the exchangeable P from the solid phase within the soil solution. Therefore, ongoing research focuses on the accessibility of P from P loaded amorphous Fe- and Al-hydroxides, previously identified as the predominant abiotic P forms. To assess their P desorption potential, P-33 rhizotron experiments combined with P-33 isotopic exchange kinetics (IEK) are underway. Preliminary results indicated that besides differences in P binding capacity of soil hydroxides, physical soil parameters, such as the matric potential, strongly control soil P availability, thus plant P acquisition rates can vary among different soil types. Our results gained new detailed information about P bioavailability under agricultural practice. The investigations towards P bioaccessibility may contribute to improved interpretation of soil P tests and reduced fertilizer recommendations

Within the tonotopic organization of the inferior colliculus two frequency ranges are well represented: a frequency range within that of the echolocation signals from 50 to 100 kHz, and a frequency band below that of the echolocation sounds, from 10 to 35 kHz. The frequency range between these two bands, from about 40 to 50 kHz is distinctly underrepresented (Fig. 3B). 2. Units with BFs in the lower frequency range (10–25 kHz) were most sensitive with thresholds of -5 to -11 dB SPL, and units with BFs within the frequency range of the echolocation signals had minimal thresholds around 0 dB SPL (Fig. 1). 3. In the medial part of the rostral inferior colliculus units were encountered which preferentially or exclusively responded to noise stimuli. — Seven neurons were found which were only excited by human breathing noises and not by pure tones, frequency modulated signals or various noise bands. These neurons were considered as a subspeciality of the larger sample of noise-sensitive neurons. — The maximal auditory sensitivity in the frequency range below that of echolocation, and the conspicuous existence of noise and breathing-noise sensitive units in the inferior colliculus are discussed in context with the foraging behavior of vampire bats

    A Simplified, Sequential, Phosphorus Fractionation Method

    Hedley et al. (1982) developed what has become the most widely used (and modified), phosphorus (P) fractionation technique. It consists of sequential extraction of increasingly less phytoavailable P pools. Extracts are centrifuged at up to 25000 g (RCF) and filtered to 0.45 μm to ensure that soil is not lost between extractions. In attempting to transfer this method to laboratories with limited facilities, it was considered that access to high-speed centrifuges, and the cost of frequent filtration may prevent adoption of this P fractionation technique. The modified method presented here was developed to simplify methodology, reduce cost, and therefore increase accessibility of P fractionation technology. It provides quantitative recovery of soil between extractions, using low speed centrifugation without filtration. This is achieved by increasing the ionic strength of dilute extracts, through the addition of NaCl, to flocculate clay particles. Addition of NaCl does not change the amount of P extracted. Flocculation with low speed centrifugation produced extracts comparable with those having undergone filtration (0.025 μm). A malachite green colorimetric method was adopted for inorganic P determination, as this simple manual method provides high sensitivity with negligible interference from other anions. This approach can also be used for total P following digestion, alternatively non-discriminatory methods, such as inductively coupled plasma atomic emission spectroscopy, may be employed

    RIPK1-mediated immunogenic cell death promotes anti-tumour immunity against soft-tissue sarcoma.

    Drugs that mobilise the immune system against cancer are dramatically improving care for many people. Dying cancer cells play an active role in inducing anti-tumour immunity but not every form of death can elicit an immune response. Moreover, resistance to apoptosis is a major problem in cancer treatment and disease control. While the term "immunogenic cell death" is not fully defined, activation of receptor-interacting serine/threonine-protein kinase 1 (RIPK1) can induce a type of death that mobilises the immune system against cancer. However, no clinical treatment protocols have yet been established that would harness the immunogenic potential of RIPK1. Here, we report the first pre-clinical application of an in vivo treatment protocol for soft-tissue sarcoma that directly engages RIPK1-mediated immunogenic cell death. We find that RIPK1-mediated cell death significantly improves local disease control, increases activation of CD8+ T cells as well as NK cells, and enhances the survival benefit of immune checkpoint blockade. Our findings warrant a clinical trial to assess the survival benefit of RIPK1-induced cell death in patients with advanced disease at limb extremities
