    Current situation and factors influencing elderly care in community day care centers: a cross-sectional study

    BackgroundThe latest census data show that people over 60 years of age account for about 18.7% of the total population in China, and the aging of the population has become an irreversible trend in the 21st century. This study aimed to investigate the current status and factors influencing the care of the elderly in community day care centers in order to lay the foundation for the development of better services in community day care centers.MethodsThis study was a cross-sectional survey using convenience sampling in Nanjing, China. The survey instrument was the Day care and Elderly Care Service Needs Questionnaire, which included the Ability of Daily Living Assessment (ADL), the Xiao Shuiyuan Social Support Rating Scale (SSRS) and the Day care Elderly Care Service Needs Survey Form, and a general information survey.ResultsA total of 450 elderly people in day care centers were surveyed. The elderly had different levels of demand for day care services, especially regarding daily care. Correlation analyses indicated that age (r = 0.619), education level (r = 0.616), source of income (r = 0.582), caregiver (r = 0.557), satisfaction with care service (r = 0.603), and degree of ADL (r = 0.629) were correlated with the need for elderly day care services (all p < 0.05). The factors influencing the demand for day care services encompassed age, education level, income source, caregiver, satisfaction with service, and ADL (all p < 0.05).ConclusionElderly care services in community day care centers are mainly based on daily and spiritual comfort, and the needs of the elderly are influenced by many factors. Timely nursing care policies and measures that target these factors are needed to improve elderly care

    Genome-Wide Association Study Identifies Candidate Genes Related to Seed Oil Composition and Protein Content in Gossypium hirsutum L.

    Cotton (Gossypium spp.) is a leading natural fiber crop and an important source of vegetable protein and oil for humans and livestock. To investigate the genetic architecture of seed nutrients in upland cotton, a genome-wide association study (GWAS) was conducted in a panel of 196 germplasm resources under three environments using a CottonSNP80K chip of 77,774 loci. Relatively high genetic diversity (average gene diversity being 0.331) and phenotypic variation (coefficient of variation, CV, exceeding 3.9%) were detected in this panel. Correlation analysis revealed that the well-documented negative association between seed protein (PR) and oil may be to some extent attributable to the negative correlation between oleic acid (OA) and PR. Linkage disequilibrium (LD) was unevenly distributed among chromosomes and subgenomes. It ranged from 0.10–0.20 Mb (Chr19) to 5.65–5.75 Mb (Chr25) among the chromosomes and the range of Dt-subgenomes LD decay distances was smaller than At-subgenomes. This panel was divided into two subpopulations based on the information of 41,815 polymorphic single-nucleotide polymorphism (SNP) markers. The mixed linear model considering both Q-matrix and K-matrix [MLM(Q+K)] was employed to estimate the association between the SNP markers and the seed nutrients, considering the false positives caused by population structure and the kinship. A total of 47 SNP markers and 28 candidate quantitative trait loci (QTLs) regions were found to be significantly associated with seven cottonseed nutrients, including protein, total fatty acid, and five main fatty acid compositions. In addition, the candidate genes in these regions were analyzed, which included three genes, Gh_D12G1161, Gh_D12G1162, and Gh_D12G1165 that were most likely involved in the control of cottonseed protein concentration. These results improved our understanding of the genetic control of cottonseed nutrients and provided potential molecular tools to develop cultivars with high protein and improved fatty acid compositions in cotton breeding programs through marker-assisted selection

    De novo sequencing of circulating miRNAs identifies novel markers predicting clinical outcome of locally advanced breast cancer

    <p>Abstract</p> <p>Background</p> <p>MicroRNAs (miRNAs) have been recently detected in the circulation of cancer patients, where they are associated with clinical parameters. Discovery profiling of circulating small RNAs has not been reported in breast cancer (BC), and was carried out in this study to identify blood-based small RNA markers of BC clinical outcome.</p> <p>Methods</p> <p>The pre-treatment sera of 42 stage II-III locally advanced and inflammatory BC patients who received neoadjuvant chemotherapy (NCT) followed by surgical tumor resection were analyzed for marker identification by deep sequencing all circulating small RNAs. An independent validation cohort of 26 stage II-III BC patients was used to assess the power of identified miRNA markers.</p> <p>Results</p> <p>More than 800 miRNA species were detected in the circulation, and observed patterns showed association with histopathological profiles of BC. Groups of circulating miRNAs differentially associated with ER/PR/HER2 status and inflammatory BC were identified. The relative levels of selected miRNAs measured by PCR showed consistency with their abundance determined by deep sequencing. Two circulating miRNAs, miR-375 and miR-122, exhibited strong correlations with clinical outcomes, including NCT response and relapse with metastatic disease. In the validation cohort, higher levels of circulating miR-122 specifically predicted metastatic recurrence in stage II-III BC patients.</p> <p>Conclusions</p> <p>Our study indicates that certain miRNAs can serve as potential blood-based biomarkers for NCT response, and that miR-122 prevalence in the circulation predicts BC metastasis in early-stage patients. These results may allow optimized chemotherapy treatments and preventive anti-metastasis interventions in future clinical applications.</p

    One-step Method to Fabricate Poly(ethylene terephthalate)/Gd(OH)3 Magnetic Nanofibers tTowards MRI-active Materials with High T1 Relaxivity and Long-term Visibility

    Magnetic resonance imaging (MRI)-active polymers exhibit unique advantages for in vivo diagnosis. Here, in order to endow electrospun fibers with long-term T1 positive MRI visibility, MRI contrast agent (CA), Gd(OH)3, is introduced in a new, extremely convenient method. Crucially, GdCl3 is reacted with NaOH in situ during electrospinning, with flexibility to deliver both well-dispersed and aggregated Gd(OH)3 clusters within a poly(ethylene terephthalate) (PET) matrix. T1 and T2 relaxivities of Gd(OH)3 in PET nanofibers are studied. Well-dispersed Gd(OH)3 (sub-nanometer in size) exhibits 34 times higher T1 relaxivity than aggregated nanoparticles when embedded within the fibers. The morphology, structure, magnetic properties, tensile properties, imaging performance and biosafety of the PET/Gd(OH)3 composite fibers are evaluated to identify the optimum conditions to produce new materials with balanced properties, excellent in vivo positive contrast and approximately 139 days imaging lifetime. Comparing this sample with a commercial CA, only 0.32 wt.% Gd loading is needed to attain similar MRI signal intensity. In summary, PET/Gd(OH)3 long-term MRI-active fibers show great potential for future biomedical applications and the study also provides a promising new general strategy to enhance the MRI T1 positive contrast of electrospun fibers of a whole host of other systems

    HCV 6a Prevalence in Guangdong Province Had the Origin from Vietnam and Recent Dissemination to Other Regions of China: Phylogeographic Analyses

    Recently in China, HCV 6a infection has shown a fast increase among patients and blood donors, possibly due to IDU linked transmission.We recruited 210 drug users in Shanwei city, Guangdong province. Among them, HCV RNA was detected in 150 (71.4%), both E1 and NS5B genes were sequenced in 136, and 6a genotyped in 70. Of the 6a sequences, most were grouped into three clusters while 23% represent emerging strains. For coalescent analysis, additional 6a sequences were determined among 21 blood donors from Vietnam, 22 donors from 12 provinces of China, and 36 IDUs from Liuzhou City in Guangxi Province. Phylogeographic analyses indicated that Vietnam could be the origin of 6a in China. The Guangxi Province, which borders Vietnam, could be the first region to accept 6a for circulation. Migration from Yunnan, which also borders Vietnam, might be equally important, but it was only detected among IDUs in limited regions. From Guangxi, 6a could have further spread to Guangdong, Yunnan, Hainan, and Hubei provinces. However, evidence showed that only in Guangdong has 6a become a local epidemic, making Guangdong the second source region to disseminate 6a to the other 12 provinces. With a rate of 2.737×10⁻³ (95% CI: 1.792×10⁻³ to 3.745×10⁻³), a Bayesian Skyline Plot was portrayed. It revealed an exponential 6a growth during 1994-1998, while before and after 1994-1998 slow 6a growths were maintained. Concurrently, 1994-1998 corresponded to a period when contaminated blood transfusion was common, which caused many people being infected with HIV and HCV, until the Chinese government outlawed the use of paid blood donations in 1998.With an origin from Vietnam, 6a has become a local epidemic in Guangdong Province, where an increasing prevalence has subsequently led to 6a spread to many other regions of China

    Fabrication of Magnetic Catalyst Fe3O4-SiO2-V3 and Its Application on Lignin Extraction from Corncob in Deep Eutectic Solvent

    Fe3O4-SiO2-V3 was prepared by deposited H6PMo9V3O40 on Fe3O4-SiO2 and employed as a catalyst to extract lignin from corncob in deep eutectic solvent (choline chloride/lactic acid = 1/10). Batch experiments were conducted in an autoclave under the conditions of 500 kPa, 90–130 °C and 15 h, while the dosage of the catalyst was set as a variable. Results indicated that the catalyst could effectively improve the qualities of the lignin, while the characteristics of the lignin showed prominent changes with the participation of the catalyst: the extraction rate increased from 71.65% to 98.13%, the purity was improved from 85.62% to 97.09%, and both the number average molecular weight and the weight average molecular weight also decreased significantly. Besides, the molecular distribution of the lignin achieved from the CC-LA-Fe-Si-V3 reaction system was found to be more highly concentrated (Polydispersity index = 1.746). Results from 2D NMR HSQC analysis indicated that lignin fractions achieved from the CC-LA-Fe-Si-V3 system showed distinct destruction involving C2-H2 in guaiacyl units (G), C5-H5 in guaiacyl units (G), and the Cγ-Hγ in γ-hydroxylated β-O-4′ substructures, but little changes in the Cγ-Hγ in phenylcoumaran substructures
