    Measles virus V protein blocks Jak1-mediated phosphorylation of STAT1 to escape IFN-α/β signaling

    AbstractViruses have evolved various strategies to escape the antiviral activity of type I interferons (IFN-α/β). For measles virus, this function is carried by the polycistronic gene P that encodes, by an unusual editing strategy, for the phosphoprotein P and the virulence factor V (MV-V). MV-V prevents STAT1 nuclear translocation by either sequestration or phosphorylation inhibition, thereby blocking IFN-α/β pathway. We show that both the N- and C-terminal domains of MV-V (PNT and VCT) contribute to the inhibition of IFN-α/β signaling. Using the two-hybrid system and co-affinity purification experiments, we identified STAT1 and Jak1 as interactors of MV-V and demonstrate that MV-V can block the direct phosphorylation of STAT1 by Jak1. A deleterious mutation within the PNT domain of MV-V (Y110H) impaired its ability to interact and block STAT1 phosphorylation. Thus, MV-V interacts with at least two components of IFN-α/β receptor complex to block downstream signaling

    Abstract Tissue samples from mosquitoes artificially infected with Zika virus and shown to be positive by RT-qPCR were reexamined by RT-PCR. Using these samples we compared the two methods employed in virus RNA detection for vector competence studies. Results demonstrated that, albeit useful, RT-PCR gave false negatives with low viral loads (< 106 RNA copies/ml)

    Social Perceptions of Forest Ecosystem Services in the Democratic Republic of Congo

    The forests of the Albertine Rift are known for their high biodiversity and the important ecosystem services they provide to millions of inhabitants. However, their conservation and the maintenance of ecosystem service delivery is a challenge, particularly in the Democratic Republic of the Congo. Our research investigates how livelihood strategy and ethnicity affects local perceptions of forest ecosystem services. We collected data through 25 focus-group discussions in villages from distinct ethnic groups, including farmers (Tembo, Shi, and Nyindu) and hunter-gatherers (Twa). Twa identify more food-provisioning services and rank bush meat and honey as the most important. They also show stronger place attachment to the forest than the farmers, who value other ecosystem services, but all rank microclimate regulation as the most important. Our findings help assess ecosystem services trade-offs, highlight the important impacts of restricted access to forests resources for Twa, and point to the need for developing alternative livelihood strategies for these communities

    The use of thermogravimetric analysis-mass spectrometry (TGMS) to study the state of preservation of archaeological bones has been investigated. As part of a collaborative multi-analytical study, bones exhumed from graves of the late Roman period in France and Italy were examined. A decrease in organic matter for the archaeological bones compared to that for new bone was confirmed, demonstrating that diagenesis of aged bones can be detected using TGMS. Different amounts of collagen were determined for bones from different graves and could, for the majority of specimens, be correlated with the visually observed preservation states

    A study of ochres from an Australian aboriginal bark painting using thermal methods

    The potential of thermogravimetric analysis (TG) as a tool for the characterisation of ochre paint used in indigenous Australian bark paintings has been investigated. TG has been combined with differential scanning calorimetry (DSC) and mass spectrometry (MS) to identify and quantify the main inorganic and organic components present in the paints. The results obtained were supported by comparison with infrared spectra and XRD data obtained for the same specimens. The potential of thermal methods for the characterisation ochres has been demonstrated, with subtle differences between small samples being able to be identified. © 2011 Akadémiai Kiadó, Budapest, Hungary
