    A Longitudinal Analysis about the Effect of Air Pollution on Astigmatism for Children and Young Adults

    Purpose: This study aimed to investigate the correlation between air pollution and astigmatism, considering the detrimental effects of air pollution on respiratory, cardiovascular, and eye health. Methods: A longitudinal study was conducted with 127,709 individuals aged 4-27 years from 9 cities in Guangdong Province, China, spanning from 2019 to 2021. Astigmatism was measured using cylinder values. Multiple measurements were taken at intervals of at least 1 year. Various exposure windows were used to assess the lagged impacts of air pollution on astigmatism. A panel data model with random effects was constructed to analyze the relationship between pollutant exposure and astigmatism. Results: The study revealed significant associations between astigmatism and exposure to carbon monoxide (CO), nitrogen dioxide (NO2), and particulate matter (PM2.5) over time. A 10 {\mu}g/m3 increase in a 3-year exposure window of NO2 and PM2.5 was associated with a decrease in cylinder value of -0.045 diopters and -0.017 diopters, respectively. A 0.1 mg/m3 increase in CO concentration within a 2-year exposure window correlated with a decrease in cylinder value of -0.009 diopters. No significant relationships were found between PM10 exposure and astigmatism. Conclusion: This study concluded that greater exposure to NO2 and PM2.5 over longer periods aggravates astigmatism. The negative effect of CO on astigmatism peaks in the exposure window of 2 years prior to examination and diminishes afterward. No significant association was found between PM10 exposure and astigmatism, suggesting that gaseous and smaller particulate pollutants have easier access to human eyes, causing heterogeneous morphological changes to the eyeball

    Eg5 Overexpression Is Predictive of Poor Prognosis in Hepatocellular Carcinoma Patients

    Eg5 (kinesin spindle protein) plays an essential role in mitosis. Inhibition of Eg5 function results in cell cycle arrest at mitosis, which leads to cell death. To date, Eg5 expression and its prognostic significance have not been studied in hepatocellular carcinoma (HCC). In this study, 26 freshly frozen HCC tissue samples and matched peritumoral tissue samples were evaluated with a one-step qPCR test and immunohistochemical (IHC) analysis was conducted on 156 HCC samples to investigate the relationships among Eg5 expression, clinicopathological factors, and prognosis. Eg5 mRNA and protein expression levels were significantly higher in HCC tissues relative to matched noncancerous tissues (p<0.05). High Eg5 protein expression was significantly related to liver cirrhosis (p=0.038) and TNM stage (p=0.008). Kaplan-Meier survival and Cox regression analyses revealed that Eg5 overexpression (p=0.001), liver cirrhosis (p=0.009), and TNM stage (p=0.025) were independent prognostic factors for overall survival. These findings indicate that Eg5 expression can be used as a biomarker of poor prognosis and as a novel therapeutic target for HCC

    Role of N6-methyladenosine-related lncRnas in pseudoexfoliation glaucoma

    ABSTRACTTo explore the role of lncRNA m6A methylation modification in aqueous humour (AH) of patients with pseudoexfoliation glaucoma (PXG). Patients with open-angle PXG under surgery from June 2021 to December 2021 were selected. Age- and gender-matched patients with age-related cataract (ARC) were chosen as control. Patients underwent detailed ophthalmic examinations. 0.05–0.1 ml AH were extracted during surgery for MeRIP-Seq and RNA-Seq. Joint analysis was used to screen lncRNAs with differential m6A methylation modification and expression. Online software tools were used to draw lncRNA-miRNA-mRNA network (ceRNA). Expression of lncRNAs and mRNAs was confirmed using quantitative real-time PCR. A total of 4151 lncRNAs and 4386 associated m6A methylation modified peaks were identified in the PXG group. Similarly, 2490 lncRNAs and 2595 associated m6A methylation modified peaks were detected in the control. Compared to the ARC group, the PXG group had 234 hypermethylated and 402 hypomethylated m6A peaks, with statistically significant differences (| Fold Change (FC) |≥2, p < 0.05). Bioinformatic analysis revealed that these differentially methylated lncRNA enriched in extracellular matrix formation, tight adhesion, TGF- β signalling pathway, AMPK signalling pathway, and MAPK signalling pathway. Joint analysis identified 10 lncRNAs with differential m6A methylation and expression simultaneously. Among them, the expression of ENST000000485383 and ROCK1 were confirmed downregulated in the PXG group by RT-qPCR. m6A methylation modification may affect the expression of lncRNA and participate in the pathogenesis of PXG through the ceRNA network.     Rural Residents in China Are at Increased Risk of Exposure to Tick-Borne Pathogens Anaplasma phagocytophilum and Ehrlichia chaffeensis

    As emerging tick born rickettsial diseases caused by A. phagocytophilum and E. chaffeensis, anaplasmosis and ehrlichiosis have become a serious threat to human and animal health throughout the world. In particular, in China, an unusual transmission of nosocomial cases of human granulocytic anaplasmosis occurred in Anhui Province in 2006 and more recent coinfection case of A. phagocytophilum and E. chaffeensis was documented in Shandong Province. Although the seroprevalence of human granulocytic anaplasmosis (former human granulocytic ehrlichiosis, HGE) has been documented in several studies, these data existed on local investigations, and also little data was reported on the seroprevalence of human monocytic ehrlichiosis (HME) in China. In this cross-sectional epidemiological study, indirect immunofluorescence antibody assay (IFA) proposed by WHO was used to detect A. phagocytophilum and E. chaffeensis IgG antibodies for 7,322 serum samples from agrarian residents from 9 provinces/cities and 819 urban residents from 2 provinces. Our data showed that farmers were at substantially increased risk of exposure. However, even among urban residents, risk was considerable. Seroprevalence of HGA and HME occurred in diverse regions of the country and tended to be the highest in young adults. Many species of ticks were confirmed carrying A. phagocytophilum organisms in China while several kinds of domestic animals including dog, goats, sheep, cattle, horse, wild rabbit, and some small wild rodents were proposed to be the reservoir hosts of A. phagocytophilum. The broad distribution of vector and hosts of the A. phagocytophilum and E. chaffeensis, especially the relationship between the generalized susceptibility of vectors and reservoirs and the severity of the disease’s clinical manifestations and the genetic variation of Chinese HGA isolates in China, is urgently needed to be further investigated
