    Book description: The relationship between states, societies, and individuals in Central and Eastern Europe has been characterised by periods of change and redefinition. The current political, economic, social and cultural climate necessitates a discussion of these issues, both past and present. It is this theme which the proposed publication intends to discuss using a selection of papers given at the 5 th Annual Postgraduate Conference on Central and Eastern Europe held at the UCL School of Slavonic and East European Studies (SSEES) in 2003. The papers represent work from young international scholars from Europe and North America writing on Central and Eastern Europe. The book consists of seven papers and develops an interdisciplinary framework reflecting the range of topics discussed during the conference. It embraces the regional breadth of Central and Eastern Europe containing analyses of Russia, the former Soviet Republics, Central Europe and South Eastern Europe. The papers chosen cover a variety of fields and adopt a corresponding range of approaches with a view to assessing from a multidisciplinary perspective the relationship between state, society and individuals. The papers in the book have been ordered chronologically. The volume starts with an analysis by Julia Mannherz of social conflict in late imperial Russia and moves on to Sergei Zhuk’s discussion of the Stundist movement in Ukraine. The third paper from Stefan Detchev is a discussion of the late-nineteenth-century politics of commemoration surrounding the Bulgarian war of independence. The theme of the politics of commemoration is also present in Andrzej Michalczyk’s analysis of the commemoration of the plebiscite in Silesia by Germans and Poles during the interwar period. Michalczyk examines how a shared event is commemorated and interpreted differently by the two national groups. The idea of common and shared histories is further developed by Rüdiger Ritter in his study of the history and the historiography of post-Communist Poland, Belarus and Lithuania. The move into the contemporary period is completed in the final two papers. The use of historical imagery for political purposes is explored in Markus Wien’s study of the King Simeon II Party in Bulgaria as well as the way in which the historical image of the monarchy has been changed for political purposes during the transition from communism to democracy. The final paper by Maria Aluchna continues the discussion of the process of transition by examining the economic transformation from a communist command economic system to a modern capitalist economy

    Physical Modeling of Process Forces in Grinding

    This paper deals with material removal mechanisms in grinding by considering single grit-workpiece interactions. Individual investigations were performed both experimentally and using finite element simulations. Firstly, a comparison between the Johnson-Cooke material model and a Crystal Plasticity finite element method was performed with the help of micro-indentation experiments. Here the research question was answered if an anisotropic material model better describe the grinding process and process forces compared to an isotropic material model. Secondly, four discretization approaches were employed: pure Lagrangian (LAG), Arbitrary Lagrange Eulerian (ALE), Particle Finite Element Method (PFEM), and Smooth Particle Hydrodynamics (SPH), to simulate a micro-cutting operation of A2024 T351 aluminium. This study aims to compare the conventional approaches (LAG and ALE) to newer approaches (PFEM and SPH). The orthogonal cutting models were benchmarked against a micro-cutting experiment presented in literature, by comparing the obtained cutting and passive forces. The study was then extended to negative rake angles to study the effect on the discretization approaches for grinding. Thirdly, scratch experiments were investigated for a brittle material sodalime glass and A2024 T351 aluminium. Effects of the linear speed of the device, depth of cut, and conical tool angle were analyzed and tendencies are built. Finally, a realistic simulation of the manufacturing process of a grinding wheel was developed, starting with the raw material, compression, sintering, and dressing until the final grinding surface. As a result of the simulations, virtual grinding wheel topographies can be visualized and analyzed with regard to the output variables from grinding wheels such as bonding strength and static grain count. The individual research studies help in understanding the material removal mechanisms in a single grit scratch process as well as in the understanding of the overall grinding wheel topography. This in turn helps in the developing an overall physical force model for scratching/grinding to predict mechanical output parameters and hence reduce the need for experimentation

    Subcellular distribution and expression of cofilin and ezrin in human colon adenocarcinoma cell lines with different metastatic potential

    The dynamic reorganization of the actin cytoskeleton is regulated by a number of actin binding proteins (ABPs). Four human colon adenocarcinoma cell lines – parental and three selected sublines, which differ in motility and metastatic potential, were used to investigate the expression level and subcellular localization of selected ABPs. Our interest was focused on cofilin and ezrin. These proteins are essential for cell migration and adhesion. The data received for the three more motile adenocarcinoma sublines (EB3, 3LNLN, 5W) were compared with those obtained for the parental LS180 adenocarcinoma cells and fibroblastic NRK cells. Quantitative densitometric analysis and confocal fluorescence microscopy were used to examine the expression levels and subcellular distribution of the selected ABPs. Our data show distinct increase in the level of cofilin in adenocarcinoma cells accompanied by the reduction of inactive phosphorylated form of cofilin. In more motile cells, cofilin was accumulated at cellular periphery in co-localization with actin filaments. Furthemore, we indicated translocation of ezrin towards the cell periphery within more motile cells in comparison with NRK and parental adenocarcinoma cells

    Nucleosomes in serum as a marker for cell death

    The concentration of nucleosomes is elevated in blood of patients with diseases which are associated with enhanced cell death. In order to detect these circulating nucleosomes, we used the Cell Death Detection-ELISA(Plus) (CDDE) from Roche Diagnostics (Mannheim, Germany) (details at http:\textbackslash{}\textbackslash{}biochem.roche.com). For its application in liquid materials we performed various modifications: we introduced a standard curve with nucleosome-rich material, which enabled direct quantification and improved comparability of the values within (CVinterassay:3.0-4.1%) and between several runs (CVinterassay:8.6-13.5%), and tested the analytical specificity of the ELISA. Because of the fast elimination of nucleosomes from circulation and their limited stability, we compared plasma and serum matrix and investigated in detail the pre-analytical handling of serum samples which can considerably influence the test results. Careless venipuncture producing hemolysis, delayed centrifugation and bacterial contamination of the blood samples led to false-positive results; delayed stabilization with EDTA and insufficient storage conditions resulted in false-negative values. At temperatures of -20 degreesC, serum samples which were treated with 10 mM EDTA were stable for at least 6 months. In order to avoid possible interfering factors, we recommend a schedule for the pre-analytical handling of the samples. As the first stage, the possible clinical application was investigated in the sera of 310 persons. Patients with solid tumors (n = 220; mean = 361 Arbitrary Units (AU)) had considerably higher values than healthy persons (n = 50; mean = 30 AU; P = 0.0001) and patients with inflammatory diseases (n = 40; mean = 296 AU; p = 0.096). Within the group of patients with tumors, those in advanced stages (UICC 4) showed significantly higher values than those in early stages (UICC 1-3) (P = 0.0004)

    The expression levels of three raft-associated molecules in cultivated vascular cells are dependent on culture conditions

    Relaying a signal across the plasma membrane requires functional connections between the partner molecules. Membrane microdomains or lipid rafts provide an environment in which such specific interactions can take place. The integrity of these sites is often taken for granted when signalling pathways are investigated in cell culture. However, it is well known that smooth muscle and endothelial cells undergo cytoskeletal rearrangements during monolayer culturing. Likewise affected - and with potentially important consequences for signalling events - is the organization of the plasma membrane. The expression levels of three raft markers were massively upregulated, and raft-associated 5′-nucleotidase activity increased in conventional monolayer cultures as compared with a spheroidal coculture model, shown to promote the differentiation of endothelial cells. Our data point to a shift of raft components in monolayer cultures and demonstrate potential advantages of the spheroid coculture system for investigation of raft-mediated signalling events in endothelial cell

    AbstractLaminin and fibronectin, but not collagen, affect the AMPase activity of the purified transmembrane protein 5'-nucleotidase. Laminin stimulates whereas fibronectin inhibits the AMPase activity of this ectoenzyme. The AMPase-modulating effects by these components of the extracellular matrix require a preincubation period of several hours when detergent-solubilized 5'-nucleotidase is employed, they can, however, instantaneously be elicited with liposome-incorporated 5/-nucleotidase
