    Fungal diversity in floral and honeydew honeys

    Studying fungal diversity in various environmental samples provides us with valuable knowledge about the occurrence of fungi of medical and ecological importance. Moreover, fungal composition may also characterise well the botanical and geographical source of food products, such as the origin of the spore enriched honeydew honeys. Thereby, we identified a wide spectrum of fungi found in 100 of honey samples from various geographical sources – most of them were from Italy, Greece and Hungary. Our honeydew honeys had a higher mean of the number of spore types found in them than floral honeys had. Statistically significant differences in diversity were found regarding the botanical source (p = 1.29 × 10–9) and the climatic classification (p = 2.28 × 10–2) according to Kruskal– Wallis rank sum tests. Most frequently encountered genera included ubiquitous saprotrophic species (Alternaria, Cladosporium, Epicoccum nigrum, Stemphylium), both in floral and honeydew honeys. On the other hand, certain sooty moulds like Aureobasidium pullulans, Tripospermum and Capnobotrys were rather present in different types of honeydew honeys. Metschnikowia reukaufii, the nectar inhabiting yeast reached considerably high quantities in floral honey samples. Present findings encourage further studies on quantifying the occurrence and the indicator value of specific fungal elements in honey, concerning its origin

    Complex behavioural changes after odour exposure in Drosophila larvae

    A variety of odorants attract Drosophila larvae, although this behaviour can be modulated by experience. For instance, larvae pre-exposed to an attractive odorant may subsequently display less attraction towards the same compound. In previous reports, this phenomenon has been interpreted as a drop in olfactory sensitivity, caused by sensory adaptation. We tried to elucidate the basis of this behavioural modification by pre-exposing larvae to various odours. After multiple pre-exposure cycles larvae were repulsed by initially attractive odours, and pre-exposure did not change the threshold concentration driving a behavioural response. We therefore believe that sensitivity to the odorant was only slightly affected in our protocol. Our results thus do not support the previous interpretation and rather suggest that olfactory pre-exposure induces a change in the hedonic value of the odour. Although we did not succeed in elucidating the exact nature of the underlying mechanism, we can reject an association of the odour with the absence of food as an interpretation of the observed behavioural changes; this is because addition of food did not abolish the repulsion to the pre-exposed odour.     Effect of Lactoferrin on Clinical Outcomes of Hospitalized Patients with COVID-19: The LAC Randomized Clinical Trial

    As lactoferrin is a nutritional supplement with proven antiviral and immunomodulatory abilities, it may be used to improve the clinical course of COVID-19. The clinical efficacy and safety of bovine lactoferrin were evaluated in the LAC randomized double-blind placebo-controlled trial. A total of 218 hospitalized adult patients with moderate-to-severe COVID-19 were randomized to receive 800 mg/die oral bovine lactoferrin (n = 113) or placebo (n = 105), both given in combination with standard COVID-19 therapy. No differences in lactoferrin vs. placebo were observed in the primary outcomes: the proportion of death or intensive care unit admission (risk ratio of 1.06 (95% CI 0.63–1.79)) or proportion of discharge or National Early Warning Score 2 (NEWS2) ≤ 2 within 14 days from enrollment (RR of 0.85 (95% CI 0.70–1.04)). Lactoferrin showed an excellent safety and tolerability profile. Even though bovine lactoferrin is safe and tolerable, our results do not support its use in hospitalized patients with moderate-to-severe COVID-19

    PTPA variants and impaired PP2A activity in early-onset parkinsonism with intellectual disability

    The protein phosphatase 2A complex (PP2A), the major Ser/Thr phosphatase in the brain, is involved in a number of signalling pathways and functions, including the regulation of crucial proteins for neurodegeneration, such as alpha-synuclein, tau and LRRK2. Here, we report the identification of variants in the PTPA/PPP2R4 gene, encoding a major PP2A activator, in two families with early-onset parkinsonism and intellectual disability. We carried out clinical studies and genetic analyses, including genome-wide linkage analysis, whole-exome sequencing, and Sanger sequencing of candidate variants. We next performed functional studies on the disease-associated variants in cultured cells and knock-down of ptpa in Drosophila melanogaster. We first identified a homozygous PTPA variant, c.893T&gt;G (p.Met298Arg), in patients from a South African family with early-onset parkinsonism and intellectual disability. Screening of a large series of additional families yielded a second homozygous variant, c.512C&gt;A (p.Ala171Asp), in a Libyan family with a similar phenotype. Both variants co-segregate with disease in the respective families. The affected subjects display juvenile-onset parkinsonism and intellectual disability. The motor symptoms were responsive to treatment with levodopa and deep brain stimulation of the subthalamic nucleus. In overexpression studies, both the PTPA p.Ala171Asp and p.Met298Arg variants were associated with decreased PTPA RNA stability and decreased PTPA protein levels; the p.Ala171Asp variant additionally displayed decreased PTPA protein stability. Crucially, expression of both variants was associated with decreased PP2A complex levels and impaired PP2A phosphatase activation. PTPA orthologue knock-down in Drosophila neurons induced a significant impairment of locomotion in the climbing test. This defect was age-dependent and fully reversed by L-DOPA treatment. We conclude that bi-allelic missense PTPA variants associated with impaired activation of the PP2A phosphatase cause autosomal recessive early-onset parkinsonism with intellectual disability. Our findings might also provide new insights for understanding the role of the PP2A complex in the pathogenesis of more common forms of neurodegeneration.</p

    Architecture of the primary taste center of Drosophila melanogaster larvae

    A simple nervous system combined with stereotypic behavioral responses to tastants, together with powerful genetic and molecular tools, have turned Drosophila larvae into a very promising model for studying gustatory coding. Using the Gal4/UAS system and confocal microscopy for visualizing gustatory afferents, we provide a description of the primary taste center in the larval central nervous system. Essentially, gustatory receptor neurons target different areas of the subesophageal ganglion (SOG), depending on their segmental and sensory organ origin. We define two major and two smaller subregions in the SOG. One of the major areas is a target of pharyngeal sensilla, the other one receives inputs from both internal and external sensilla. In addition to such spatial organization of the taste center, circumstantial evidence suggests a subtle functional organization: aversive and attractive stimuli might be processed in the anterior and posterior part of the SOG, respectively. Our results also suggest less coexpression of gustatory receptors than proposed in prior studies. Finally, projections of putative second-order taste neurons seem to cover large areas of the SOG. These neurons may thus receive multiple gustatory inputs. This suggests broad sensitivity of secondary taste neurons, reminiscent of the situation in mammals

    The discrimination of honey origin using melissopalynology and Raman spectroscopy techniques coupled with multivariate analysis

    Honey traceability to food quality is required by consumers and food control institutions. Melissopalynologists traditionally use percentages of nectariferous pollens to discriminate the botanical origin and the entire pollen spectrum (presence/absence, type and quantities and association of some pollen types) to determinate the geographical origin of honeys. To improve melissopalynological routine analysis, principal components analysis (PCA) was used. A remarkable and innovative result was that the most significant pollens for the traditional discrimination of the botanical and geographical origin of honeys were the same as those individuated with the chemometric model. The reliability of assignments of samples to honey classes was estimated through explained variance (85%). This confirms that the chemometric model properly describes the melissopalynological data. With the aim to improve honey discrimination, FT-microRaman spectrography and multivariate analysis were also applied. Well performing PCA models and good agreement with known classes were achieved.
