    Domain interactions within Fzo1 oligomers are essential for mitochondrial fusion

    Mitofusins are conserved GTPases essential for the fusion of mitochondria. These mitochondrial outer membrane proteins contain a GTPase domain and two or three regions with hydrophobic heptad repeats, but little is known about how these domains interact to mediate mitochondrial fusion. To address this issue, we have analyzed the yeast mitofusin Fzo1p and find that mutation of any of the three heptad repeat regions (HRN, HR1, and HR2) leads to a null allele. Specific pairs of null alleles show robust complementation, indicating that functional domains need not exist on the same molecule. Biochemical analysis indicates that this complementation is due to Fzo1p oligomerization mediated by multiple domain interactions. Moreover, we find that two non-overlapping protein fragments, one consisting of HRN/GTPase and the other consisting of HR1/HR2, can form a complex that reconstitutes Fzo1p fusion activity. Each of the null alleles disrupts the interaction of these two fragments, suggesting that we have identified a key interaction involving the GTPase domain and heptad repeats essential for fusion

    Significant improvements in structural efficiency can be achieved by the utilization of advanced composites for construction of aircraft secondary structures. Careful evaluation of alternate designs and materials for the L-1011 advanced composite inboard aileron has led to the selection of several unique material combinations and easily manufactured structural configurations. The advanced composite aileron is a direct replacement for the metal aileron with a weight savings of 23 percent. Due to the configurational simplicity of the components within the composite aileron, and because it contains 50 percent fewer parts and fasteners than the metal aileron, it is predicted that the composite aileron will be cost competitive with the metal aileron in a production environment. Structural analysis of the composite aileron, in conjunction with the design data, concept verification, and ground tests, indicates that the composite aileron design meets or exceeds structural requirements

    Numerous design concepts, materials, and manufacturing methods were investigated analytically and empirically for the covers and spars of a transport wing box. This information was applied to the design, analysis, and fabrication of a full-scale section of a transport wing box. A blade-stiffened design was selected for the upper and lower covers of the box. These covers have been constructed using three styles of AS4/974 prepreg fabrics. The front and rear T-stiffened channel spars were filament wound using AS4/1806 towpreg. Covers, ribs, and spars were assembled using mechanical fasteners. When they are completed later this year, the tests on the technology integration box beam will demonstrate the structural integrity of an advanced composite wing design which is 25 percent lighter than the metal baseline

    The WD40 protein Caf4p is a component of the mitochondrial fission machinery and recruits Dnm1p to mitochondria

    The mitochondrial division machinery regulates mitochondrial dynamics and consists of Fis1p, Mdv1p, and Dnm1p. Mitochondrial division relies on the recruitment of the dynamin-related protein Dnm1p to mitochondria. Dnm1p recruitment depends on the mitochondrial outer membrane protein Fis1p. Mdv1p interacts with Fis1p and Dnm1p, but is thought to act at a late step during fission because Mdv1p is dispensable for Dnm1p localization. We identify the WD40 repeat protein Caf4p as a Fis1p-associated protein that localizes to mitochondria in a Fis1p-dependent manner. Caf4p interacts with each component of the fission apparatus: with Fis1p and Mdv1p through its NH2-terminal half and with Dnm1p through its COOH-terminal WD40 domain. We demonstrate that mdv1{Delta} yeast contain residual mitochondrial fission due to the redundant activity of Caf4p. Moreover, recruitment of Dnm1p to mitochondria is disrupted in mdv1{Delta} caf4{Delta} yeast, demonstrating that Mdv1p and Caf4p are molecular adaptors that recruit Dnm1p to mitochondrial fission sites. Our studies support a revised model for assembly of the mitochondrial fission apparatus

    Simultaneous measurements of particulate and gas-phase water-soluble organic carbon concentrations at remote and urban-influenced locations

    The sources, sinks, and overall importance of watersoluble organic carbon (WSOC) in the atmosphere are not well understood. Although the primary historical focus has been on particulate WSOC (WSOCP), here we also present results obtained using a newly developed technique that additionally measures gas-phase water-soluble organic carbon (WSOCG). These first-of-their-kind measurements show that WSOCG can often be more than ten times larger than WSOCP at both urban and remote locations. The average fraction of WSOC residing in the gas phase (fg = WSOCG/(WSOCG + WSOCP)) at five various field sites ranged from 0.64 to 0.93, implying significant differences in WSOC phase partitioning between locations. At Houston, TX, and Summit, Greenland, a repeatable diurnal pattern was observed, with minimum values for fg occurring at night. These trends likely are due, at least in part, to temperature and/or relative humidity related gas-to-particle partitioning. These coincident measurements of WSOC in both the gas and particle phases indicate that a relatively large reservoir of water-soluble organic mass is not taken into account by measurements focused only on WSOCP. In addition, a significant amount of WSOCG is available to form WSOCP or enter cloud droplets depending on the chemical and physical properties of the droplets and/or aerosols present.     Acoustic waves and heating due to molecular energy transfer in an electric discharge CO laser

    This paper summarizes analytical studies and the interpretation of experimental results for the compression and rarefaction waves generated in the cavity of a pulsed CO electric discharge laser. A one-dimensional analysis of acoustic waves is applied to a transversely excited laser. The influences of heating in the cathode fall, heat transfer to the cathode, flow through both the anode and cathode, and bulk heating of the plasma are included. The analysis is used to relate the bulk heating rate to observable features of the pressure and density waves. Data obtained from interferograms and reported elsewhere are used to infer the bulk heating rates in a pulsed CO laser. Results are presented for CO/Ar, CO/N2, and N2 plasmas. Comparison of the data with recent theoretical results for the heating due to electron/ neutral collisions and the anharmonic defect associated with V-V energy transfer shows substantial differences at lower values of total energy deposition. The change of heating with E/N is in fairly good agreement with predicted values
