Effects of Rainfall on E. coli Concentrations at Door County, Wisconsin Beaches
Rainfall and its associated storm water runoff have been associated with transport of many pollutants into beach water. Fecal material, from a variety of animals (humans, pets, livestock, and wildlife), can wash into beach water following rainfall and result in microbial contamination of the beach. Many locales around the world issue pre-emptive beach closures associated with rainfall. This study looked at eight beaches located in Door County, Wisconsin, on Lake Michigan to determine the impact of rainfall on E. coli concentrations in beach water. Water samples were collected from beach water and storm water discharge pipes during rainfall events of 5 mm in the previous 24 hours. Six of the eight beaches showed a significant association between rainfall and elevated beach water E. coli concentrations. The duration of the impact of rainfall on beach water E. coli concentrations was variable (immediate to 12 hours). Amount of rainfall in the days previous to the sampling did not have significant impact on the E. coli concentrations measured in beach water. Presence of storm water conveyance pipes adjacent to the beach did not have a uniform impact on beach water E. coli concentrations. This study suggests that each beach needs to be examined on its own with regard to rain impacts on E coli concentrations in beach water
Student understanding of rotational and rolling motion concepts
We investigated the common difficulties that students have with concepts
related to rotational and rolling motion covered in the introductory physics
courses. We compared the performance of calculus- and algebra-based
introductory physics students with physics juniors who had learned rotational
and rolling motion concepts in an intermediate level mechanics course.
Interviews were conducted with six physics juniors and ten introductory
students using demonstration-based tasks. We also administered free-response
and multiple-choice questions to a large number of students enrolled in
introductory physics courses, and interviewed six additional introductory
students on the test questions (during the test design phase). All students
showed similar difficulties regardless of their background, and higher
mathematical sophistication did not seem to help acquire a deeper
understanding. We found that some difficulties were due to related difficulties
with linear motion, while others were tied specifically to the more intricate

Extent and Volume of Lava Flows Erupted at 9°50′N, East Pacific Rise in 2005–2006 From Autonomous Underwater Vehicle Surveys
Seafloor volcanic eruptions are difficult to directly observe due to lengthy eruption cycles and the remote location of mid-ocean ridges. Volcanic eruptions in 2005–2006 at 9°50′N on the East Pacific Rise have been well documented, but the lava volume and flow extent remain uncertain because of the limited near-bottom bathymetric data. We present near-bottom data collected during 19 autonomous underwater vehicle (AUV) Sentry dives at 9°50′N in 2018, 2019, and 2021. The resulting 1 m-resolution bathymetric grid and 20 cm-resolution sidescan sonar images cover 115 km2, and span the entire area of the 2005–2006 eruptions, including an 8 km2 pre-eruption survey collected with AUV ABE in 2001. Pre- and post-eruption surveys, combined with sidescan sonar images and seismo-acoustic impulsive events recorded during the eruptions, are used to quantify the lava flow extent and to estimate changes in seafloor depth caused by lava emplacement. During the 2005–2006 eruptions, lava flowed up to ∼3 km away from the axial summit trough, covering an area of ∼20.8 km2; ∼50% larger than previously thought. Where pre- and post-eruption surveys overlap, individual flow lobes can be resolved, confirming that lava thickness varies from ∼1 to 10 m, and increases with distance from eruptive fissures. The resulting lava volume estimate indicates that ∼57% of the melt extracted from the axial melt lens probably remained in the subsurface as dikes. These observations provide insights into recharge cycles in the subsurface magma system, and are a baseline for studying future eruptions at the 9°50′N area.publishedVersio

Bayesian timing analysis of giant flare of SGR 1806-20 by RXTE PCA
By detecting high frequency quasi-periodic oscillations (QPOs) and estimating
frequencies of them during the decaying tail of giant flares from Soft
Gamma-ray Repeaters (SGRs) useful constraints for the equation of state (EoS)
of superdense matter may be obtained via comparison with theoretical
predictions of eigenfrequencies. We used the data collected by the Rossi X-Ray
Timing Explorer (RXTE/XTE) Proportional Counter Array (PCA) of a giant flare of
SGR 1806-20 on 2004 Dec 27 and applied a Bayesian periodicity detection method
(Gregory & Loredo, 1992) for the search of oscillations of transient nature. In
addition to the already detected frequencies, we found a few new frequencies
(f_{QPOs} ~ 16.9, 21.4, 36.4, 59.0, 116.3 Hz) of oscillations predicted by
Colaiuda et al. (2009) based on the APR_{14} EoS (Akmal et al., 1998) for SGR

Prospective Study of Antibiotic Prophylaxis for Prostate Biopsy Involving >1100 Men
We aimed to compare infection rates for two 3-day antibiotic prophylaxis regimens for transrectal ultrasound-guided prostate biopsy (TRUSgbp) and demonstrate local microbiological trends. In 2008, 558 men and, in 2009, 625 men had TRUSgpb. Regimen 1 (2008) comprised 400 mg Ofloxacin immediately before biopsy and 200 mg 12-hourly for 3 days. Regimen 2 (2009) comprised Ofloxacin 200 mg 12-hourly for 3 days commencing 24 hours before biopsy. 20/558 (3.6%) men had febrile episodes with regimen 1 and 10/625 (1.6%) men with regimen 2 (P = 0.03). E. coli was the most frequently isolated organism. Overall, 7/13 (54%) of positive urine cultures were quinolone resistant and (5/13) 40% were multidrug resistant. Overall, 5/9 (56%) patients with septicaemia were quinolone resistant. All patients were sensitive to Meropenem. There was 1 (0.2%) death with regimen 1. Commencing Ofloxacin 24 hours before TRUSgpb reduced the incidence of febrile episodes significantly. We observed the emergence of quinolone and multidrug-resistant E. coli. Meropenem should be considered for unresolving sepsis
Emulating Natural Disturbances for Declining Late- Successional Species: A Case Study of the Consequences for Cerulean Warblers (Setophaga cerulea)
Forest cover in the eastern United States has increased over the past century and while some late-successional species have benefited from this process as expected, others have experienced population declines. These declines may be in part related to contemporary reductions in small-scale forest interior disturbances such as fire, windthrow, and treefalls. To mitigate the negative impacts of disturbance alteration and suppression on some late-successional species, strategies that emulate natural disturbance regimes are often advocated, but large-scale evaluations of these practices are rare. Here, we assessed the consequences of experimental disturbance (using partial timber harvest) on a severely declining late-successional species, the cerulean warbler (Setophaga cerulea), across the core of its breeding range in the Appalachian Mountains. We measured numerical (density), physiological (body condition), and demographic (age structure and reproduction) responses to three levels of disturbance and explored the potential impacts of disturbance on source-sink dynamics. Breeding densities of warblers increased one to four years after all canopy disturbances (vs. controls) and males occupying territories on treatment plots were in better condition than those on control plots. However, these beneficial effects of disturbance did not correspond to improvements in reproduction; nest success was lower on all treatment plots than on control plots in the southern region and marginally lower on light disturbance plots in the northern region. Our data suggest that only habitats in the southern region acted as sources, and interior disturbances in this region have the potential to create ecological traps at a local scale, but sources when viewed at broader scales. Thus, cerulean warblers would likely benefit from management that strikes a landscape-level balance between emulating natural disturbances in order to attract individuals into areas where current structure is inappropriate, and limiting anthropogenic disturbance in forests that already possess appropriate structural attributes in order to maintain maximum productivity
Therapy with high-dose Interleukin-2 (HD IL-2) in metastatic melanoma and renal cell carcinoma following PD1 or PDL1 inhibition
Abstract
Background
Metastatic melanoma (mM) and renal cell carcinoma (mRCC) are often treated with anti-PD-1 based therapy, however not all patients respond and further therapies are needed. High dose interleukin-2 (HD IL-2) can lead to durable responses in a subset of mM and mRCC patients. The efficacy and toxicity of HD IL-2 therapy following anti-PD-1 or anti-PD-L1 therapy have not yet been explored.
Methods
Reports on mM and mRCC patients who had received HD IL-2 after PD-1 or PD-L1 inhibition were queried from the PROCLAIMSM database. Patient characteristics, toxicity and efficacy were analyzed.
Results
A total of 57 patients (40 mM, 17 mRCC) were treated with high dose IL-2 after PD-1 or PD-L1 inhibition and had data recorded in the PROCLAIM database. The best overall response rate to HD IL-2 was 22.5% for mM (4 complete response (CR), 5 partial responses (PRs)) and 24% for mRCC (2 CRs, 2 PRs). The toxicity related to HD IL-2 observed in these patients was similar to that observed in patients treated with HD IL-2 without prior checkpoint blockade. One patient who had received prior PD-L1 blockade developed drug induced pneumonitis with HD IL-2 requiring steroid therapy.
Conclusion
In this retrospective analysis, HD IL-2 therapy displayed durable antitumor activity in mM and mRCC patients who progressed following treatment with PD-1 and PD-L1 inhibition. The toxicities were generally manageable and consistent with expectations from HD IL-2 but physicians should watch for immune related toxicities such as pneumonitis.
