    Secondary and tertiary budbreak release is enhanced by extended dormancy chilling in 'Shiraz' grapevines

    Limited information exists regarding the dormancy of secondary and tertiary buds within the compound bud of grapevines. We were interested to evaluate how extended chilling duration would affect the budbreak percentage of all three bud orders within the compound bud. Dormant potted 'Shiraz' grapevines, with thirty retained buds per vine, were placed in a cool room at 4 °C over a 20-month period to extend dormancy. Vines were then systematically removed over five dates and placed in a controlled growth environment to assess budbreak percentages and cane tissue concentrations of abscisic acid and cytokinins. Budbreak was hastened by at least 13 d with vines receiving any extra chilling compared to no initial chilling. Furthermore, the firstly observed correlative inhibition of basal buds was apparently removed with chilling. Removal of correlative inhibition within the compound bud with increased chilling duration was also observed with the increases of secondary (doubles) and tertiary (triples) buds breaking simultaneously with the primary bud at each node. This resulted in 91 % of the nodes having two developing shoots and 56 % of the nodes presenting three developing shoots by the end of the experiment. Furthermore, a sigmoidal relationship between percent secondary buds and chilling duration was observed. Possible phytohormone connections were observed with the decline of ABA as secondary and tertiary buds broke at 414 d of chilling and depletion of the CKs, which coincided with the greatest rate of primary bud break after 242 d of chilling. Other possible involvement of ABA and Cis-type cytokinin interactions with secondary and tertiary bud dormancy is discussed. The results suggest future investigations into secondary and tertiary bud release within the grapevine using similar techniques may help to better understand the biochemical mechanisms associated with dormancy

    Evaluation and histological examination of a Campylobacter fetus subsp. venerealis small animal infection model

    Bovine genital campylobacteriosis (BGC), caused by Campylobacter fetus subsp. venerealis, is associated with production losses in cattle worldwide. This study aimed to develop a reliable BGC guinea pig model to facilitate future studies of pathogenicity, abortion mechanisms and vaccine efficacy. Seven groups of five pregnant guinea pigs (1 control per group) were inoculated with one of three strains via intraperitoneal (IP) or intra-vaginal routes. Samples were examined using culture, PCR and histology. Abortions ranged from 0% to 100% and re-isolation of causative bacteria from sampled sites varied with strain, dose of bacteria and time to abortion. Histology indicated metritis and placentitis, suggesting that the bacteria induce inflammation, placental detachment and subsequent abortion. Variation of virulence between strains was observed and determined by culture and abortion rates. IP administration of C fetus subsp. venerealis to pregnant guinea pigs is a promising small animal model for the investigation of BGC abortion. (C) 2014 Elsevier Ltd. All rights reserved

    LoCuSS: The Sunyaev-Zel'dovich Effect and Weak Lensing Mass Scaling Relation

    We present the first weak-lensing-based scaling relation between galaxy cluster mass, M_wl, and integrated Compton parameter Y_sph. Observations of 18 galaxy clusters at z~0.2 were obtained with the Subaru 8.2-m telescope and the Sunyaev-Zel'dovich Array. The M_wl-Y_sph scaling relations, measured at Delta=500, 1000, and 2500 rho_c, are consistent in slope and normalization with previous results derived under the assumption of hydrostatic equilibrium (HSE). We find an intrinsic scatter in M_wl at fixed Y_sph of 20%, larger than both previous measurements of M_HSE-Y_sph scatter as well as the scatter in true mass at fixed Y_sph found in simulations. Moreover, the scatter in our lensing-based scaling relations is morphology dependent, with 30-40% larger M_wl for undisturbed compared to disturbed clusters at the same Y_sph at r_500. Further examination suggests that the segregation may be explained by the inability of our spherical lens models to faithfully describe the three-dimensional structure of the clusters, in particular, the structure along the line-of-sight. We find that the ellipticity of the brightest cluster galaxy, a proxy for halo orientation, correlates well with the offset in mass from the mean scaling relation, which supports this picture. This provides empirical evidence that line-of-sight projection effects are an important systematic uncertainty in lensing-based scaling relations.Comment: Accepted versio

    Observational Evidence of Wave Ducting and Evanescence in the Mesosphere

    A collaborative radar and imaging study of gravity waves over the Hawaiian Islands was performed during October 1993 as part of the Airborne Lidar and Observations of Hawaiian Airglow 1993/Coupling and Dynamics of Regions Equatorial (ALOHA-93/CADRE) campaign to investigate the propagation characteristics of short-period (\u3c1 hour) waves at nightglow altitudes. The horizontal wavelengths and apparent phase speeds of quasi-monochromatic wave events were measured in four separate nightglow emissions using data obtained by a high-resolution CCD imager. This information was correlated with simultaneous MF radar wind measurements over the same height interval (∼80–100 km) to infer intrinsic wave parameters in each case. Correlating the two data sets allowed the determination of the local vertical wavenumber for each event, in particular whether it be real (indicative of freely propagating waves) or imaginary (indicative of ducted or evanescent waves). The results of this study indicate a preponderance of ducted or evanescent waves at 80–100 km during the time of the observations, with up to ∼75% of the events recorded exhibiting ducted or evanescent behavior. Also noted was a tendency for ducted behavior to be more prevalent among waves with shorter horizontal wavelengths, in agreement with Doppler ducting theory. These results suggest that ducted waves are relatively common in the upper mesosphere and lower thermosphere region, at least over the mid-Pacific Ocean. As small-scale waves which are ducted have the potential to travel much longer horizontal distances than freely propagating waves, the frequency of their occurrence should be taken into account in efforts to quantify gravity wave effects at these altitudes

    A Minimal Model of Metabolism Based Chemotaxis

    Since the pioneering work by Julius Adler in the 1960's, bacterial chemotaxis has been predominantly studied as metabolism-independent. All available simulation models of bacterial chemotaxis endorse this assumption. Recent studies have shown, however, that many metabolism-dependent chemotactic patterns occur in bacteria. We hereby present the simplest artificial protocell model capable of performing metabolism-based chemotaxis. The model serves as a proof of concept to show how even the simplest metabolism can sustain chemotactic patterns of varying sophistication. It also reproduces a set of phenomena that have recently attracted attention on bacterial chemotaxis and provides insights about alternative mechanisms that could instantiate them. We conclude that relaxing the metabolism-independent assumption provides important theoretical advances, forces us to rethink some established pre-conceptions and may help us better understand unexplored and poorly understood aspects of bacterial chemotaxis

    THE ROLE OF INTERDEPENDENCE IN THE MICRO-FOUNDATIONS OF ORGANIZATION DESIGN: TASK, GOAL, AND KNOWLEDGE INTERDEPENDENCE

    Interdependence is a core concept in organization design, yet one that has remained consistently understudied. Current notions of interdependence remain rooted in seminal works, produced at a time when managers’ near-perfect understanding of the task at hand drove the organization design process. In this context, task interdependence was rightly assumed to be exogenously determined by characteristics of the work and the technology. We no longer live in that world, yet our view of interdependence has remained exceedingly task-centric and our treatment of interdependence overly deterministic. As organizations face increasingly unpredictable workstreams and workers co-design the organization alongside managers, our field requires a more comprehensive toolbox that incorporates aspects of agent-based interdependence. In this paper, we synthesize research in organization design, organizational behavior, and other related literatures to examine three types of interdependence that characterize organizations’ workflows: task, goal, and knowledge interdependence. We offer clear definitions for each construct, analyze how each arises endogenously in the design process, explore their interrelations, and pose questions to guide future research

Accurate demographic and disorder data could assist with breeding and clinical prioritisation. The VetCompassTM Programme collects clinical data on dogs under primary veterinary care in the UK.     Impacts of acoustic and gravity waves on the ionosphere

    The impact of regional-scale neutral atmospheric waves has been demonstrated to have profound effects on the ionosphere, but the circumstances under which they generate ionospheric disturbances and seed plasma instabilities are not well understood. Neutral atmospheric waves vary from infrasonic waves of &lt;20 Hz to gravity waves with periods on the order of 10 min, for simplicity, hereafter they are combined under the common term Acoustic and Gravity Waves (AGWs). There are other longer period waves like planetary waves from the lower and middle atmosphere, whose effects are important globally, but they are not considered here. The most ubiquitous and frequently observed impact of AGWs on the ionosphere are Traveling Ionospheric Disturbances (TIDs), but AGWs also affect the global ionosphere/thermosphere circulation and can trigger ionospheric instabilities (e.g., Perkins, Equatorial Spread F). The purpose of this white paper is to outline additional studies and observations that are required in the coming decade to improve our understanding of the impact of AGWs on the ionosphere.</p
