Formation of a TBX20-CASZ1 protein complex is protective against dilated cardiomyopathy and critical for cardiac homeostasis
By the age of 40, one in five adults without symptoms of cardiovascular disease are at risk for developing congestive heart failure. Within this population, dilated cardiomyopathy (DCM) remains one of the leading causes of disease and death, with nearly half of cases genetically determined. Though genetic and high throughput sequencing-based approaches have identified sporadic and inherited mutations in a multitude of genes implicated in cardiomyopathy, how combinations of asymptomatic mutations lead to cardiac failure remains a mystery. Since a number of studies have implicated mutations of the transcription factor TBX20 in congenital heart diseases, we investigated the underlying mechanisms, using an unbiased systems-based screen to identify novel, cardiac-specific binding partners. We demonstrated that TBX20 physically and genetically interacts with the essential transcription factor CASZ1. This interaction is required for survival, as mice heterozygous for both Tbx20 and Casz1 die post-natally as a result of DCM. A Tbx20 mutation associated with human familial DCM sterically interferes with the TBX20-CASZ1 interaction and provides a physical basis for how this human mutation disrupts normal cardiac function. Finally, we employed quantitative proteomic analyses to define the molecular pathways mis-regulated upon disruption of this novel complex. Collectively, our proteomic, biochemical, genetic, and structural studies suggest that the physical interaction between TBX20 and CASZ1 is required for cardiac homeostasis, and further, that reduction or loss of this critical interaction leads to DCM. This work provides strong evidence that DCM can be inherited through a digenic mechanism

Impact of lung cancer screening results on participant health-related quality of life and state anxiety in the National Lung Screening Trial
BACKGROUND Low-dose computed tomography (LDCT) lung screening has been associated with a 20% reduction in lung cancer mortality. A major barrier to the adoption of lung screening is the potential negative psychological impact of a false-positive (FP) screen, occurring in 20% to 50% of those screened. The objective of this study was to assess the impact of abnormal findings on health-related quality of life (HRQoL) and anxiety in the American College of Radiology (ACRIN)/National Lung Screening Trial (NLST). METHODS The NLST was a randomized screening trial comparing LDCT with chest X-ray screening (CXR). This study was part of the original protocol. A total of 2812 participants at 16 of 23 ACRIN sites who had baseline HRQoL assessments were asked to complete the Short Form-36 and the State Trait Anxiety Inventory (form Y-1) questionnaires to assess short-term (1 month) and long-term (6 months) effects of screening. FP were lung cancer–free at 1 year, and true-positives (TP) were not. RESULTS Of the total participants, 1024 (36.4%) participants were FP, 63 (2.2%) were TP, 344 (12.2%) had significant incidental findings (SIFs), and 1381 (49.1%) had negative screens. Participants had been randomized to LDCT (n = 1947) and CXR (n = 865). Short-term and long-term HRQoL and state anxiety did not differ across participants with FP, SIF, or negative screens. Short-term and long-term HRQoL were lower and anxiety was higher for TP participants compared to participants with FP, SIF, and negative screens. CONCLUSIONS In a large multicenter lung screening trial, participants receiving a false-positive or SIF screen result experienced no significant difference in HRQoL or state anxiety at 1 or at 6 months after screening relative to those receiving a negative result. The Intrinsic Shapes of Low Surface Brightness Galaxies (LSBGs):A Discriminant of LSBG Galaxy Formation Mechanisms
We use the low surface brightness galaxy (LSBG) samples created from the Hyper Suprime-Cam Subaru Strategic Program (781 galaxies), the Dark Energy Survey (20977 galaxies), and the Legacy Survey (selected via H I detection in the Arecibo Legacy Fast ALFA Survey, 188 galaxies) to infer the intrinsic shape distribution of the LSBG population. To take into account the effect of the surface brightness cuts employed when constructing LSBG samples, we simultaneously model both the projected ellipticity and the apparent surface brightness in our shape inference. We find that the LSBG samples are well characterized by oblate spheroids, with no significant difference between red and blue LSBGs. This inferred shape distribution is in good agreement with similar inferences made for ultra-diffuse cluster galaxy samples, indicating that environment does not play a key role in determining the intrinsic shape of LSBGs. We also find some evidence that LSBGs are more thickened than similarly massive high surface brightness dwarfs. We compare our results to intrinsic shape measures from contemporary cosmological simulations, and find that the observed LSBG intrinsic shapes place considerable constraints on the formation path of such galaxies. In particular, LSBG production via the migration of star formation to large radii produces intrinsic shapes in good agreement with our observational findings
Beyond Ultra-Diffuse Galaxies I: Mass-Size Outliers Among the Satellites of Milky Way Analogs
Large diffuse galaxies are hard to find, but understanding the environments
where they live, their numbers, and ultimately their origins, is of intense
interest and importance for galaxy formation and evolution. Using Subaru's
Hyper Suprime-Cam Strategic Survey Program, we perform a systematic search for
low surface brightness galaxies and present novel and effective methods for
detecting and modeling them. As a case study, we surveyed 922 Milky Way analogs
in the nearby Universe () and build a large sample of
satellite galaxies that are outliers in the mass-size relation. These
``ultra-puffy'' galaxies (UPGs), defined to be above the average
mass-size relation, represent the tail of the satellite size distribution. We
find that each MW analog hosts ultra-puffy
galaxies on average, which is consistent with but slightly lower than the
observed abundance at this halo mass in the Local Volume. We also construct a
sample of ultra-diffuse galaxies (UDGs) in MW analogs and find an abundance of
per host. With literature results, we confirm that
the UDG abundance scales with the host halo mass following a sublinear power
law. We argue that our definition for ultra-puffy galaxies, which is based on
the mass-size relation, is more physically-motivated than the common definition
of ultra-diffuse galaxies, which depends on surface brightness and size cuts
and thus yields different surface mass density cuts for quenched and

Tidal Features at 0.05<z<0.45 in the Hyper Suprime-Cam Subaru Strategic Program: Properties and Formation Channels
We present 1,201 galaxies at that host tidal features, detected
from the first deg of imaging from the Hyper Suprime-Cam
Subaru Strategic Program (HSC-SSP). All galaxies in the present sample have
spectroscopic observations from the Sloan Digital Sky Survey (SDSS)
spectroscopic campaigns, generating a sample of 21208 galaxies. Of these
galaxies, we identify 214 shell systems and 987 stream systems. For 575 of
these systems, we are additionally able to measure the colors of the
tidal features. We find evidence for star formation in a subset of the streams,
with the exception of streams around massive ellipticals, and find that stream
host galaxies span the full range of stellar masses in our sample. Galaxies
which host shells are predominantly red and massive: we find that observable
shells form more frequently around ellipticals than around disc galaxies of the
same stellar mass. Although the majority of the shells in our sample are
consistent with being formed by minor mergers, of shell host
galaxies have colors as red as their host galaxy, consistent with being
formed by major mergers. These "red shells" are additionally preferentially
aligned with the major axis of the host galaxy, as previously predicted from
simulations. We suggest that although the bulk of the observable shell
population originates from fairly minor mergers, which preferentially form
shells that are not aligned with the major axis of the galaxy, major mergers

A Gro/TLE-NuRD Corepressor Complex Facilitates Tbx20-Dependent Transcriptional Repression
The cardiac transcription factor Tbx20 has a critical role in the proper morphogenetic development of the vertebrate heart, and its misregulation has been implicated in human congenital heart disease. Although it is established that Tbx20 exerts its function in the embryonic heart through positive and negative regulation of distinct gene programs, it is unclear how Tbx20 mediates proper transcriptional regulation of its target genes. Here, using a combinatorial proteomic and bioinformatic approach, we present the first characterization of Tbx20 transcriptional protein complexes. We have systematically investigated Tbx20 protein-protein interactions by immunoaffinity purification of tagged Tbx20 followed by proteomic analysis using GeLC-MS/MS, gene ontology classification, and functional network analysis. We demonstrate that Tbx20 is associated with a chromatin remodeling network composed of TLE/Groucho co-repressors, members of the Nucleosome Remodeling and Deacetylase (NuRD) complex, the chromatin remodeling ATPases RUVBL1/RUVBL2, and the T-box repressor Tbx18. We determined that the interaction with TLE co-repressors is mediated via an eh1 binding motif in Tbx20. Moreover, we demonstrated that ablation of this motif results in a failure to properly assemble the repression network and disrupts Tbx20 function in vivo. Importantly, we validated Tbx20-TLE interactions in the mouse embryonic heart, and identified developmental genes regulated by Tbx20:TLE binding, thereby confirming a primary role for a Tbx20-TLE repressor complex in embryonic heart development. Together, these studies suggest a model in which Tbx20 associates with a Gro/TLE-NuRD repressor complex to prevent inappropriate gene activation within the forming heart
Home visits by neighborhood Mentor Mothers provide timely recovery from childhood malnutrition in South Africa: results from a randomized controlled trial
Abstract Background Child and infant malnourishment is a significant and growing problem in the developing world. Malnourished children are at high risk for negative health outcomes over their lifespans. Philani, a paraprofessional home visiting program, was developed to improve childhood nourishment. The objective of this study is to evaluate whether the Philani program can rehabilitate malnourished children in a timely manner. Methods Mentor Mothers were trained to conduct home visits. Mentor Mothers went from house to house in assigned neighborhoods, weighed children age 5 and younger, and recruited mother-child dyads where there was an underweight child. Participating dyads were assigned in a 2:1 random sequence to the Philani intervention condition (n = 536) or a control condition (n = 252). Mentor Mothers visited dyads in the intervention condition for one year, supporting mothers' problem-solving around nutrition. All children were weighed by Mentor Mothers at baseline and three, six, nine and twelve month follow-ups. Results By three months, children in the intervention condition were five times more likely to rehabilitate (reach a healthy weight for their ages) than children in the control condition. Throughout the course of the study, 43% (n = 233 of 536) of children in the intervention condition were rehabilitated while 31% (n = 78 of 252) of children in the control condition were rehabilitated. Conclusions Paraprofessional Mentor Mothers are an effective strategy for delivering home visiting programs by providing the knowledge and support necessary to change the behavior of families at risk
Depressed mood in pregnancy: Prevalence and correlates in two Cape Town peri-urban settlements
<p>Abstract</p> <p>Background</p> <p>The disability associated with depression and its impact on maternal and child health has important implications for public health policy. While the prevalence of postnatal depression is high, there are no prevalence data on antenatal depression in South Africa. The purpose of this study was to determine the prevalence and correlates of depressed mood in pregnancy in Cape Town peri-urban settlements.</p> <p>Methods</p> <p>This study reports on baseline data collected from the Philani Mentor Mothers Project (PMMP), a community-based, cluster-randomized controlled trial on the outskirts of Cape Town, South Africa. The PMMP aims to evaluate the effectiveness of a home-based intervention for preventing and managing illnesses related to HIV, TB, alcohol use and malnutrition in pregnant mothers and their infants. Participants were 1062 pregnant women from Khayelitsha and Mfuleni, Cape Town. Measures included the Edinburgh Postnatal Depression Scale (EPDS), the Derived AUDIT-C, indices for social support with regards to partner and parents, and questions concerning socio-demographics, intimate partner violence, and the current pregnancy. Data were analysed using bivariate analyses followed by logistic regression.</p> <p>Results</p> <p>Depressed mood in pregnancy was reported by 39% of mothers. The strongest predictors of depressed mood were lack of partner support, intimate partner violence, having a household income below R2000 per month, and younger age.</p> <p>Conclusions</p> <p>The high prevalence of depressed mood in pregnancy necessitates early screening and intervention in primary health care and antenatal settings for depression. Project Masihambisane: a cluster randomised controlled trial with peer mentors to improve outcomes for pregnant mothers living with HIV
Abstract Background Pregnant women living with HIV (WLH) face daily challenges maintaining their own and their babies' health and mental health. Standard Prevention of Maternal to Child Transmission (PMTCT) programs are not designed to address these challenges. Methods/Design As part of a cluster randomized controlled trial, WLH are invited to attend four antenatal and four postnatal small group sessions led by a peer WLH (a Peer Mentor). The WLH and their babies are assessed during pregnancy and at one week, six months, and twelve months post-birth. Mobile phones are used to collect routine information, complete questionnaires and remain in contact with participants over time. Pregnant WLH (N = 1200) are randomly assigned by clinic (N = 8 clinics) to an intervention program, called Masihambisane (n = 4 clinics, n = 600 WLH) or a standard care PMTCT control condition (n = 4 clinics; n = 600 WLH). Discussion Data collection with cellular phones are innovative and effective in low-resource settings. Standard PMTCT programs are not designed to address the daily challenges faced by WLH; Peer Mentors may be useful in supporting WLH to cope with these challenges.
