    Nano-scale analysis of titanium dioxide fingerprint-development powders

    Titanium dioxide based powders are regularly used in the development of latent fingerprints on dark surfaces. For analysis of prints on adhesive tapes, the titanium dioxide is suspended in a surfactant and used in the form of a small particle reagent (SPR). Analysis of commercially available products shows varying levels of effectiveness of print development, with some powders adhering to the background as well as the print. Scanning electron microscopy (SEM) images of prints developed with different powders show a range of levels of aggregation of particles. Analytical transmission electron microscopy (TEM) of the fingerprint powder shows TiO2 particles with a surrounding coating, tens of nanometres thick, consisting of Al and Si rich material. X ray photoelectron spectroscopy (XPS) is used to determine the composition and chemical state of the surface of the powders; with a penetration depth of approximately 10nm, this technique demonstrates differing Ti: Al: Si ratios and oxidation states between the surfaces of different powders. Levels of titanium detected with this technique demonstrate variation in the integrity of the surface coating. The thickness, integrity and composition of the Al/Si-based coating is related to the level of aggregation of TiO2 particles and efficacy of print development

    Roles and regulation of membrane-associated serine proteases

    Pericellular proteolytic activity affects many aspects of cellular behaviour, via mechanisms involving processing of the extracellular matrix, growth factors and receptors. The serine proteases have exquisitely sensitive regulatory mechanisms in this setting, involving both receptor-bound and transmembrane proteases. Receptor-bound proteases are exemplified by the uPA (urokinase plasminogen activator)/uPAR (uPAR receptor) plasminogen activation system. The mechanisms initiating the activity of this proteolytic system on the cell surface, a critical regulatory point, are poorly understood. We have found that the expression of the TTSP (type II transmembrane serine protease) matriptase is highly regulated in leucocytes, and correlates with the presence of active uPA on their surface. Using siRNA (small interfering RNA), we have demonstrated that matriptase specifically activates uPAR-associated pro-uPA. The uPA/uPAR system has been implicated in the activation of the plasminogen-related growth factor HGF (hepatocyte growth factor). However, we find no evidence for this, but instead that HGF can be activated by both matriptase and the related TTSP hepsin in purified systems. Hepsin is of particular interest, as the proteolytic cleavage sequence of HGF is an ā€˜ideal substrateā€™ for hepsin and membrane-associated hepsin activates HGF with high efficiency. Both of these TTSPs can be activated autocatalytically at the cell surface, an unusual mechanism among the serine proteases. Therefore these TTSPs have the capacity to be true upstream initiators of proteolytic activity with subsequent downstream effects on cell behaviour

    Evaluation of the HARDMAN comparability methodology for manpower, personnel and training

    The methodology evaluation and recommendation are part of an effort to improve Hardware versus Manpower (HARDMAN) methodology for projecting manpower, personnel, and training (MPT) to support new acquisition. Several different validity tests are employed to evaluate the methodology. The methodology conforms fairly well with both the MPT user needs and other accepted manpower modeling techniques. Audits of three completed HARDMAN applications reveal only a small number of potential problem areas compared to the total number of issues investigated. The reliability study results conform well with the problem areas uncovered through the audits. The results of the accuracy studies suggest that the manpower life-cycle cost component is only marginally sensitive to changes in other related cost variables. Even with some minor problems, the methodology seem sound and has good near term utility to the Army. Recommendations are provided to firm up the problem areas revealed through the evaluation
