    Development modeling of Lucilia sericata

    The relationship between insect development and temperature has been well established and has a wide range of uses, including the use of blow flies for postmortem (PMI) interval estimations in death investigations. To use insects in estimating PMI, we must be able to determine the insect age at the time of discovery and backtrack to time of oviposition. Unfortunately, existing development models of forensically important insects are only linear approximations and do not take into account the curvilinear properties experienced at extreme temperatures. A series of experiments were conducted with Lucilia sericata, a forensically important blow fly species, that met the requirements needed to create statistically valid development models. Experiments were conducted over 11 temperatures (7.5 to 32.5 °C, at 2.5 °C) with a 16:8 L:D cycle. Experimental units contained 20 eggs, 10 g beef liver, and 2.5 cm of pine shavings. Each life stage (egg to adult) had five sampling times. Each sampling time was replicated four times, for a total of 20 measurements per life stage. For each sampling time, the cups were pulled from the chambers and the stage of each maggot was documented morphologically through posterior spiracle slits and cephalopharyngeal skeletal development. Data were normally distributed with the later larval stages (L3f, L3m) having the most variation within and transitioning between stages. The biological minimum was between 7.5 °C and 10 °C, with little egg development and no egg emergence at 7.5 °C. Temperature-induced mortality was highest from 10.0 to 17.5 °C and 32.5 °C. The development data generated illustrates the advantages of large datasets in modeling Lucilia sericata development and the need for curvilinear models in describing development at environmental temperatures near the biological minima and maxima

    In the absence of cancer registry data, is it sensible to assess incidence using hospital separation records?

    BACKGROUND: Within the health literature, a major goal is to understand distribution of service utilisation by social location. Given equivalent access, differential incidence leads to an expectation of differential service utilisation. Cancer incidence is differentially distributed with respect to socioeconomic status. However, not all jurisdictions have incidence registries, and not all registries allow linkage with utilisation records. The British Columbia Linked Health Data resource allows such linkage. Consequently, we examine whether, in the absence of registry data, first hospitalisation can act as a proxy measure for incidence, and therefore as a measure of need for service. METHODS: Data are drawn from the British Columbia Linked Health Data resource, and represent 100% of Vancouver Island Health Authority cancer registry and hospital records, 1990–1999. Hospital separations (discharges) with principal diagnosis ICD-9 codes 140–208 are included, as are registry records with ICDO-2 codes C00-C97. Non-melanoma skin cancer (173/C44) is excluded. Lung, colorectal, female breast, and prostate cancers are examined separately. We compare registry and hospital annual counts and age-sex distributions, and whether the same individuals are represented in both datasets. Sensitivity, specificity and predictive values are calculated, as is the kappa statistic for agreement. The registry is designated the gold standard. RESULTS: For all cancers combined, first hospitalisation counts consistently overestimate registry incidence counts. From 1995–1999, there is no significant difference between registry and hospital counts for lung and colorectal cancer (p = 0.42 and p = 0.56, respectively). Age-sex distribution does not differ for colorectal cancer. Ten-year period sensitivity ranges from 73.0% for prostate cancer to 84.2% for colorectal cancer; ten-year positive predictive values range from 89.5% for female breast cancer to 79.35% for prostate cancer. Kappa values are consistently high. CONCLUSION: Claims and registry databases overlap with an appreciable proportion of the same individuals. First hospital separation may be considered a proxy for incidence with reference to colorectal cancer since 1995. However, to examine equity across cancer health services utilisation, it is optimal to have access to both hospital and registry files

    Evaluation of pre- and post-zygotic mating barriers, hybrid fitness and phylogenetic relationship between Cyprinodon variegatus variegatus

    The euryhaline fish Cyprinodon variegatus variegatus (Cvv) is capable of tolerating ambient salinities ranging from 0.3 to 167 g l(-1) , but incapable of long-term survival in freshwater (< 2 mM Na(+) ). However, a population of this species, now designated as a subspecies (Cyprinodon variegatus hubbsi; Cvh), has been isolated in several freshwater (0.4-1 mM Na(+) ) lakes in central Florida for the past ~150 ky. We previously demonstrated that Cvh has a significantly higher affinity for Na(+) uptake suggesting that it has adapted to its dilute freshwater environment. We here evaluate whether Cvh should be considered a separate species by characterizing pre- and post-zygotic isolation, Na(+) transport characteristics of the two populations and their hybrids, and developing a molecular phylogeny of Cvv and Cvh populations in Florida using mtDNA sequence data. We found evidence of partial prezygotic isolation with Cvv females mating almost exclusively (89%) with con-specific males in choice mating experiments. Partial post-zygotic isolation was also observed with significant (59-89%) reductions in hatching success of hybrid embryos compared with con-specific embryos. Na(+) uptake kinetics in hybrids (both Cvv x Cvh and Cvh x Cvv) bred and raised under common garden conditions were intermediate to Cvh (high affinity) and Cvv (low affinity) indicating that observed differences are genetically based. Similar observations were made with respect to short-term (96 h) survival of juveniles acutely transferred from 7 mM Na(+) to a range of more dilute (0.1-2 mM Na(+) ) freshwater. Finally, although phylogenetic analysis of Cvv and Cvh populations using mtDNA sequence for ND2 were unable to fully resolve a polytomy between Cvh and Cvv populations from northeastern Florida, these data do not falsify the hypothesis that Cvh is of monophyletic origin. Overall, the available data suggest that Cvh should be considered a separate species or at a minimum an evolutionarily significant unit
