    The practice of breastfeeding is a very important process for the infant and, up to six months, is the only food needed since it provides all the substances and nutrients required in order for an optimal growth. After this age, food introduction should begin in an appropriate way and breastfeeding could continue for up to two years of age. If these recommendations are followed, the mother and the child will enjoy health, economic and psychological benefits. The data reported for an inadequate practice of breastfeeding is alarming. The status reported by the State of the World's Children issued by the United Nations Fund for Children (UNICEF), shows that nearly two out of five children receive exclusive breastfeeding and one in four children have continued breastfeeding until they accomplish the second year of life. (UNICEF, 2007).La práctica de la lactancia materna es un proceso de vital importancia para el niño ya que hasta los seis meses es el único alimento que éste necesita porque le aporta todas las sustancias para crecer en forma óptima. A partir de esta edad se debe empezar con la introducción de alimentos de una manera adecuada, continuando con la lactancia materna hasta los dos años de vida. Si se la practica siguiendo estas recomendaciones los beneficios tanto para la madre como para el niño son múltiples. Los datos que se reportan sobre una práctica inadecuada de lactancia materna son alarmantes debido a que, en el reporte del estado mundial de la infancia emitido por el Fondo de las Naciones Unidas para la infancia (UNICEF), se muestra que aproximadamente dos de cada cinco niños reciben una lactancia materna exclusiva y uno de cada cuatro niños tienen una lactancia continua.     Phytochemical study of the Genus Salpichroa (Solanaceae), chemotaxonomic considerations, and biological evaluation in prostate and breast cancer cells

    Twelve Salpichroa taxa have been phytochemically analyzed. From the aerial parts of S. scandens, four known salpichrolides A, C, I, S, and an unreported withanolide named salpichrolide V (1), were isolated. In S. dependens, S. gayi, S. glandulosa subsp. glandulosa, S. glandulosa subps. weddellii, S. leucantha, S. micrantha, S. microloba, S. proboscidea, S. ramosissima, S. tristis var. tristis, and S. weberbauerii, no withanolides were found. The chemical content of ca. 85% of the Salpichroa taxa is in agreement with molecular studies, which suggest that Salpichroa and Jaborosa, a genus considered morphologically close to Salpichroa, are distant in the systematic of the Solanoideae subfamily. Moreover, the in vitro cytotoxic activity of a set of natural salpichrolides and derivatives was examined against two prostate carcinoma cell lines (PC3 and LNCaP) and two human breast cancer cell lines (MCF-7 and T47D).     Analgesia induced by the epigenetic drug, L-acetylcarnitine, outlasts the end of treatment in mouse models of chronic inflammatory and neuropathic pain

    Background: L-acetylcarnitine, a drug marketed for the treatment of chronic pain, causes analgesia by epigenetically up-regulating type-2 metabotropic glutamate (mGlu2) receptors in the spinal cord. Because the epigenetic mechanisms are typically long-lasting, we hypothesized that analgesia could outlast the duration of L-acetylcarnitine treatment in models of inflammatory and neuropathic pain. Results: A seven-day treatment with L-acetylcarnitine ( 100 mg/kg, once a day, i.p.) produced an antiallodynic effect in the complete Freund adjuvant mouse model of chronic inflammatory pain. L-Acetylcarnitine-induced analgesia persisted for at least 14 days after drug withdrawal. In contrast, the analgesic effect of pregabalin, amitryptiline, ceftriaxone, and N-acetylcysteine disappeared seven days after drug withdrawal. L-acetylcarnitine treatment enhanced mGlu2/3 receptor protein levels in the dorsal region of the spinal cord. This effect also persisted for two weeks after drug withdrawal and was associated with increased levels of acetylated histone H3 bound to the Grm2 gene promoter in the dorsal root ganglia. A long-lasting analgesic effect of L-acetylcarnitine was also observed in mice subjected to chronic constriction injury of the sciatic nerve. In these animals, a 14-day treatment with pregabalin, amitryptiline, tramadol, or L-acetylcarnitine produced a significant antiallodynic effect, with pregabalin displaying the greatest efficacy. In mice treated with pregabalin, tramadol or L-acetylcarnitine the analgesic effect was still visible 15 days after the end of drug treatment. However, only in mice treated with L-acetylcarnitine analgesia persisted 37 days after drug withdrawal. This effect was associated with an increase in mGlu2/3 receptor protein levels in the dorsal horns of the spinal cord. Conclusions: Our findings suggest that L-acetylcarnitine has the unique property to cause a long-lasting analgesic effect that might reduce relapses in patients suffering from chronic pain

    Aerosporas fungicas del area oriente de Santiago (1996-1997)

    Utilizando un captador Burkard se estudia la aeromicobiota presente en Santiago oriente durante 12 meses (agosto 1996 a julio 1997) identificando las aerosporas fúngicas por microscopía directa. Estas fueron agrupadas en 19 categorías taxonómicas, correspondiendo el 76.2% a Deuteromycetes, el 13.6% a Basidiomycecetes, 2% a Ascomycetes y el 8,3% a aerosporas no identificadas.En nuestro registro predominan los hongos dematiáceos debido a la gran presencia de Cladosporium, 73%. Las basidiosporas (no identificadas) constituyeron el 8,4%, carbones 4.3%, ascosporas (no identificadas)1.6%, Alternaria 1% y el resto de los géneros con frecuencias menores al 1%. Cladosporium presenta un contenido aéreo mayor en otoño (abril) como Epicoccum y Stemphylium; las basidiosporas, conidios de Aspergillus/Penicillium y ascosporas aumentan en los meses de invierno.Alternarla se dispersa con igual fecuencia durante primavera y otoño. Ganoderma y Helminthosporium aumentan durante el verano.Con el registro de tres años consecutivos se confeccionará un calendario aeromicológico que permita a los pacientes con rinitis o asma reconocer los periodos de mayor dispersión aérea de estos alérgenos

    Selective targeting of proteins by hybrid polyoxometalates: Interaction between a bis-biotinylated hybrid conjugate and avidin

    The Keggin-type polyoxometalate [\u3b3-SiW10O36]8 12 was covalently modified to obtain a bis-biotinylated conjugate able to bind avidin. Spectroscopic studies such as UV-vis, fluorimetry, circular dichroism, coupled to surface plasmon resonance technique were used to highlight the unique interplay of supramolecular interactions between the homotetrameric protein and the bis-functionalized polyanion. In particular, the dual recognition mechanism of the avidin encompasses (i) a complementary electrostatic association between the anionic surface of the polyoxotungstate and each positively charged avidin subunit and (ii) specific host-guest interactions between each biotinylated arm and a corresponding pocket on the tetramer subunits. The assembly exhibits peroxidase-like reactivity and it was used in aqueous solution for L-methionine methyl ester oxidation by H2O2. The recognition phenomenon was then exploited for the preparation of layer-by-layer films, whose structural evolution was monitored in situ by ATR-FTIR spectroscopy. Finally, cell tracking studies were performed by exploiting the specific interactions with a labeled streptavidin

    Abstract The regio- and stereoselectivity of the addition reactions of O-, C-, N-, and S-nucleophiles to l-arabinal-derived vinyl epoxide 2, the simplest non-conformationally restricted glycal-derived vinyl epoxide, has been examined and compared with the corresponding, conformationally restricted d-galactal-derived analogs 1β and 1β-Me. Results indicated that the 1,4-/1,2-regioselectivity ratio and the related syn-1,4-/anti-1,2-stereoselectivity observed in glycal-derived vinyl oxiranes is independent of the presence of substituents on the six-membered unsaturated ring, and the absence of conformational freedom: it depends only on the ability of the nucleophile to give a coordination process with the oxirane oxygen in the form of a hydrogen bond or through a coordinating cation

    A Novel t(8;14)(q24;q11) Rearranged Human Cell Line as a Model for Mechanistic and Drug Discovery Studies of NOTCH1-Independent Human T-Cell Leukemia

    MYC-translocated T-lineage acute lymphoblastic leukemia (T-ALL) is a rare subgroup of T-ALL associated with CDKN2A/B deletions, PTEN inactivation, and absence of NOTCH1 or FBXW7 mutations. This subtype of T-ALL has been associated with induction failure and aggressive disease. Identification of drug targets and mechanistic insights for this disease are still limited. Here, we established a human NOTCH1-independent MYC-translocated T-ALL cell line that maintains the genetic and phenotypic characteristics of the parental leukemic clone at diagnosis. The University of Padua T-cell acute lymphoblastic leukemia 13 (UP-ALL13) cell line has all the main features of the above described MYC-translocated T-ALL. Interestingly, UP-ALL13 was found to harbor a heterozygous R882H DNMT3A mutation typically found in myeloid leukemia. Chromatin immunoprecipitation coupled with high-throughput sequencing for histone H3 lysine 27 (H3K27) acetylation revealed numerous putative super-enhancers near key transcription factors, including MYC, MYB, and LEF1. Marked cytotoxicity was found following bromodomain-containing protein 4 (BRD4) inhibition with AZD5153, suggesting a strict dependency of this particular subtype of T-ALL on the activity of super-enhancers. Altogether, this cell line may be a useful model system for dissecting the signaling pathways implicated in NOTCH1-independent T-ALL and for the screening of targeted anti-leukemia agents specific for this T-ALL subgroup
