    First report of Cytauxzoon sp. infection in domestic cats in Switzerland: natural and transfusion-transmitted infections

    BACKGROUND: Cytauxzoonosis is an emerging tick-borne disease of domestic and wild felids. Cytauxzoon felis induces severe and often fatal disease in domestic cats. In Europe, clinical and subclinical infections caused by Cytauxzoon sp. are described. We report the first cases of Cytauxzoon sp. infection in domestic cats in Switzerland. METHODS: Clinical and laboratory data and results of PCR analyses were collected from Cytauxzoon sp. PCR-positive cats and the cats followed for up to 851 days. RESULTS: The cases were three two-month old kittens from the same litter (Cases 1-3) and two adult domestic shorthair cats (Cases 4 and 5). The cats originated from the north-west and west of Switzerland. Cases 1-3 presented with moderate to severe regenerative anaemia and intraerythrocytic inclusions. Cytauxzoon sp. was confirmed by PCR and sequencing. The kittens made a clinical and haematological recovery after blood transfusion and/or treatment with azithromycin and atovaquone, but erythroparasitaemia persisted. Case 4 presented with severe non-regenerative anaemia. Case 5 was healthy and used as a blood donor for Case 4. Following blood transfusion, Case 4 showed intraerythrocytic inclusions, and Cytauxzoon sp. was confirmed in both Cases 4 and 5 using PCR and sequencing. Case 4 achieved clinical and haematological remission after treatment with azithromycin, atovaquone and immunosuppressive drugs. Eight months later, Case 4 was presented again with anaemia but tested Cytauxzoon sp. PCR-negative. Sequencing of 1637 bp of the 18S rRNA gene of Cytauxzoon sp. revealed 100% nucleotide sequence identity among isolates of Cases 1-3 and between isolates of Cases 4 and 5, and 99% sequence identity between isolates of all cases. Phylogenetic analysis revealed the closest relationship of the Swiss isolates to Cytauxzoon sp. isolates from domestic cats and wild felids from France, Spain and Romania and to Cytauxzoon manul from a Pallas's cat. CONCLUSIONS: This is the first report of Cytauxzoon sp. infection in domestic cats in Switzerland. It is also the first report of infection in very young kittens and transmission of Cytauxzoon sp. to an adult cat by transfusion of blood from an asymptomatic cat. The cats recovered but some developed chronic asymptomatic erythroparasitaemia for up to 28 months. Domestic cats may act as reservoirs for Cytauxzoon sp. in Europe and blood donor cats should be screened for this agent by PCR

    In situ measurements of atmospheric O2 and CO2 reveal an unexpected O2 signal over the tropical Atlantic Ocean

    We present the first meridional transects of atmospheric O2 and CO2 over the Atlantic Ocean. We combine these measurements into the tracer atmospheric potential oxygen (APO), which is a measure of the oceanic contribution to atmospheric O2 variations. Our new in situ measurement system, deployed on board a commercial container ship during 2015, performs as well as or better than existing similar measurement systems. The data show small short-term variability (hours to days), a step-change corresponding to the position of the Intertropical Convergence Zone (ITCZ), and seasonal cycles that vary with latitude. In contrast to data from the Pacific Ocean and to previous modeling studies, our Atlantic Ocean APO data show no significant bulge in the tropics. This difference cannot be accounted for by interannual variability in the position of the ITCZ or the Atlantic Meridional Mode Index and appears to be a persistent feature of the Atlantic Ocean system. Modeled APO using the TM3 atmospheric transport model does exhibit a significant bulge over the Atlantic and overestimates the interhemispheric gradient in APO over the Atlantic Ocean. These results indicate that either there are inaccuracies in the oceanic flux data products in the equatorial Atlantic Ocean region, or that there are atmospheric transport inaccuracies in the model, or a combination of both. Our shipboard O2 and CO2 measurements are ongoing and will reveal the long-term nature of equatorial APO outgassing over the Atlantic as more data become available

    Telomeric Trans-Silencing in Drosophila melanogaster: Tissue Specificity, Development and Functional Interactions between Non-Homologous Telomeres

    BACKGROUND: The study of P element repression in Drosophila melanogaster led to the discovery of the telomeric Trans-Silencing Effect (TSE), a homology-dependent repression mechanism by which a P-transgene inserted in subtelomeric heterochromatin (Telomeric Associated Sequences, "TAS") has the capacity to repress in trans, in the female germline, a homologous P-lacZ transgene located in euchromatin. TSE can show variegation in ovaries, displays a maternal effect as well as an epigenetic transmission through meiosis and involves heterochromatin and RNA silencing pathways. PRINCIPAL FINDINGS: Here, we analyze phenotypic and genetic properties of TSE. We report that TSE does not occur in the soma at the adult stage, but appears restricted to the female germline. It is detectable during development at the third instar larvae where it presents the same tissue specificity and maternal effect as in adults. Transgenes located in TAS at the telomeres of the main chromosomes can be silencers which in each case show the maternal effect. Silencers located at non-homologous telomeres functionally interact since they stimulate each other via the maternally-transmitted component. All germinally-expressed euchromatic transgenes tested, located on all major chromosomes, were found to be repressed by a telomeric silencer: thus we detected no TSE escaper. The presence of the euchromatic target transgene is not necessary to establish the maternal inheritance of TSE, responsible for its epigenetic behavior. A single telomeric silencer locus can simultaneously repress two P-lacZ targets located on different chromosomal arms. CONCLUSIONS AND SIGNIFICANCE: Therefore TSE appears to be a widespread phenomenon which can involve different telomeres and work across the genome. It can explain the P cytotype establishment by telomeric P elements in natural Drosophila populations

    Variability in atmospheric O2 and CO2 concentrations in the southern Pacific Ocean and their comparison with model estimates

    We examine ship-based observations of atmospheric O-2 and CO2 in the southern Pacific Ocean made during two voyages: in February 2003 and in April 2004. We found, for the Austral late summer to autumn, evidence of a maximum in Atmospheric Potential Oxygen (APO) (an atmospheric tracer that is conservative with respect to terrestrial biological activity, APO approximate to O-2 + CO2) centered around 50 degrees S owing to biologically driven O-2 outgassing (associated with strong productivity in the Sub-Tropical Convergence Zone) and evidence in the February voyage of a decreasing APO trend from 57 degrees S south toward the Antarctic coast. The observed APO variability appears to be primarily determined by the interplay between atmospheric transport and the spatial distribution of O-2 air-sea flux resulting from regional differences in biological production. Comparisons of these observations with APO derived from coupling the TM3 atmospheric transport model with flux estimates from (1) the PISCES-T ocean biogeochemistry model, and (2) O-2 and CO2 climatologies, show that both PISCES-T and the climatology reproduce the observed gradient and curvature of APO but underestimate the magnitude of the observed APO maximum in the mid southern latitudes. Although the temporal limitation of our data does not permit us to calculate the annual mean APO gradient, the results support previous model predictions of a decreasing APO trend from the mid to high southern latitudes
