    Abstract This work presents a general procedure to characterize equivalent acoustic sources to reproduce the sound pressure field generated by the engines on an aircraft fuselage. The procedure would allow to set up ground experimental tests on aircraft components, by means of distributed loudspeakers, to obtain their vibro-acoustic performances as if they were tested in flight conditions. A FEM model of an aircraft fuselage mock-up was built up, comprising the structure, the internal acoustic cavities and the external air. The sound pressure field generated by the engines was considered as the reference solution, whereas an equivalent sound field, produced by distributed monopole sources surrounding the structure, was obtained by leveraging on the proposed Multi-Disciplinary Optimization (MDO) procedure. The MDO procedure was based on the mutual interaction between the commercial codes Siemens NX, for the CAE/FEM simulations, and Noesis Optimus, for the optimization framework

    A Novel Optimization Framework to Replicate the Vibro-Acoustics Response of an Aircraft Fuselage

    In this work, a novel optimization framework, based on a Multi-Disciplinary Optimization (MDO) procedure, applied to the vibro-acoustic Finite Element Method (FEM) model of an aircraft fuselage mock-up, is proposed. The MDO procedure, based on an Efficient Global Optimization (EGO)-like approach, is implemented to characterize acoustic sources that replicate the sound pressure field generated by the engines on the fuselage. A realistic sound pressure field, evaluated by aeroacoustic simulations, was considered as the reference acoustic load, whereas two equivalent sound fields, displayed by two different arrays of microphones and generated by the same configuration of monopoles, were calculated by the proposed vibro-acoustic FEM-MDO procedure. The proposed FEM-MDO framework enables to set up ground experimental tests on aircraft components, useful to replicate their vibro-acoustic performances as if tested in flight. More in general, such a procedure can also be used as a reference tool to design simplified tests starting from more complex ones

    Respiratory symptoms in children living near busy roads and their relationship to vehicular traffic: results of an Italian multicenter study (SIDRIA 2)

    BACKGROUND: Epidemiological studies have provided evidence that exposure to vehicular traffic increases the prevalence of respiratory symptoms and may exacerbate pre-existing asthma in children. Self-reported exposure to road traffic has been questioned as a reliable measurement of exposure to air pollutants. The aim of this study was to investigate whether there were specific effects of cars and trucks traffic on current asthma symptoms (i.e. wheezing) and cough or phlegm, and to examine the validity of self-reported traffic exposure. METHODS: The survey was conducted in 2002 in 12 centers in Northern, Center and Southern Italy, different in size, climate, latitude and level of urbanization. Standardized questionnaires filled in by parents were used to collect information on health outcomes and exposure to traffic among 33,632 6-7 and 13-14 years old children and adolescents. Three questions on traffic exposure were asked: the traffic in the zone of residence, the frequency of truck and of car traffic in the street of residence. The presence of a possible response bias for the self-reported traffic was evaluated using external validation (comparison with measurements of traffic flow in the city of Turin) and internal validations (matching by census block, in the cities of Turin, Milan and Rome). RESULTS: Overall traffic density was weakly associated with asthma symptoms but there was a stronger association with cough or phlegm (high traffic density OR = 1.24; 95% CI: 1.04, 1.49). Car and truck traffic were independently associated with cough or phlegm. The results of the external validation did not support the existence of a reporting bias for the observed associations, for all the self-reported traffic indicators examined. The internal validations showed that the observed association between traffic density in the zone of residence and respiratory symptoms did not appear to be explained by an over reporting of traffic by parents of symptomatic subjects. CONCLUSION: Children living in zones with intense traffic are at higher risk for respiratory effects. Since population characteristics are specific, the results of validation of studies on self-reported traffic exposure can not be generalized

    New combinatorial strategies to improve the PARP inhibitors efficacy in the urothelial bladder Cancer treatment

    Abstract Background Novel therapeutic strategies are urgently needed for the treatment of metastatic Urothelial Bladder Cancer. DNA damaging repair (DDR) targeting has been introduced in cinical trials for bladder cancer patients that carry alterations in homologous DNA repair genes, letting to envisage susceptibility to the Poly (adenosine diphosphate [ADP]) ribose polymerase (PARP) inhibitors. Main body PARP inhibition, by amplifying the DNA damage, augments the mutational burden and promotes the immune priming of the tumor by increasing the neoantigen exposure and determining upregulation of programmed death ligand 1 (PD-L1) expression. Thus, the combination of PARP-inhibition and the PD/PD-L1 targeting may represent a compelling strategy to treat bladder cancer and has been introduced in recent clinical trials. The targeting of DDR has been also used in combination with epigenetic drugs able to modulate the expression of genes involved in DDR, and also able to act as immunomodulator agents suggesting their use in combination with immune-checkpoint inhibitors. Conclusion In conclusion, it may be envisaged the combination of three classes of drugs to treat bladder cancer, by targeting the DDR process in a tumor context of DDR defect, together with epigenetic agents and immune-checkpoint inhibitors, whose association may amplify the effects and reduce the doses and the toxicity of each single drug

    The muscle invasive form of urothelial bladder cancer (UBC) is a deadly disease. Currently, the therapeutic approach of UBC is mostly based on surgery and standard chemotherapy. Biomarkers to establish appropriate drugs usage are missing. Deficiency of the tumor suppressor CCDC6 determines PARP-inhibitor sensitivity. The CCDC6 levels are modulated by the deubiquitinase USP7. In this work we scored CCDC6 and USP7 expression levels in primary UBC and we evaluated the expression levels of CCDC6 in correlation with the effects of the PARP-inhibitors combined with the USP7 inhibitor, P5091, in vitro. Since PARP-inhibitors could be enhanced by conventional chemotherapy or DNA damage inducers, we tested the new agent RRx-001, able to induce DNA damage, to prove the benefit of combined treatments in bladder cancer cells
