    Membrane processing of grape must for control of the alcohol content in fermented beverages

    The great demand of beverages, both alcohol-free and with low alcohol content, is a great challenge for the production of beverages with controlled alcohol content through the use of sustainable enological practices. The present work addresses this challenge with the processing of grape must by reverse osmosis (RO) for must reconstitution with different sugar contents prior to the alcoholic fermentation. The original must came from grapes grown in Quinta do Quinto, in Santarém, collected after destemming and mechanic crushing, and preserved in a refrigerated chamber at -1.6 °C until processing by RO. The RO processing was carried out in Escola Superior Agrária de Santarém, with a pilot plant equipped with RO spiral wound modules, M38RO, from Alfa Laval, Denmark. The total membrane permeation area is 15 m2. The work pressure was 55 bar. The original must had 23.7 oBrix, a density of 1108 g.L-1, 15.2% (v/v) of probable alcohol, and a conductivity of 2.01 mS.cm-1. The must reconstitution was carried out, by mixing the concentrated grape must with the vegetal water produced by RO (permeate) to obtain beverages with a nominal alcohol content of 5%, 7%, 10% and 13% (v/v). The fermentation average temperature was between 18.2 and 19.7 °C, and the final density rounded about 993 g.cm-3. The beverages were analysed by different parameters, including total polyphenols, total anthocyanins, colour intensity and hue, the coordinates CIELab, alcohol content, total acidity, volatile acidity, pH, free SO2 and total SO2. The attributes of the beverage, corresponding to the visual appearance, aroma and taste senses, as well as the overall judgment were evaluated by the tasters. The proposed method can produce beverages with controlled low alcohol content.     Peri-operative anaphylaxis in Coimbra: the experience of our drug allergy clinic

    Background: The incidence of peri-operative anaphylactic reactions ranges from 1:10,000 to 1:20,000. It is crucial to identify the risk factors, the trigger agent and to fi nd adequate alternatives. Aims: To characterise patients with peri-operative anaphylactic reactions. Methods: We performed a retrospective study of patients with suspected adverse peri-operative reactions to anaesthetic drugs, observed in a Drug Allergy Outpatients Clinic from 1994 to 2005. Anaphylaxis was diagnosed based on the clinical history and skin tests to latex, neuromuscular blocking agents (NMBA), hypnotics, opioids and antibiotics. Specifi c IgE to latex was performed in patients with positive skin-prick-tests. Results: We evaluated the clinical charts of 45 patients with adverse peri-operative reactions, 39 of whom were female. Anaphylaxis was diagnosed in 16 patients (35.6%), 15 females, mean age 36.3 years. The remaining patients had non-anaphylactic (28.8%) or not well-characterised reactions (35.6%). In terms of severity, 50% presented grade III and 50% grade IV anaphylaxis. Skin-prick-tests were positive to latex in three patients and negative to all anaesthetic drugs. Intradermal skin tests were positive in 10 out of 16 patients with anaphylactic reactions: 10 to NMBA [atracurium (n = 8), vecuronium (n = 4), suxamethonium (n = 4) and pancuronium (n = 2)], one to fentanyl, one to thiopental and one to cefuroxime. In three cases, the culprit agent was not identifi ed. In four patients, at least one alternative NMBA, with no skin test reactivity, was found. Of these four patients, two underwent further surgical procedures without adverse reactions. Five patients had previously had two or more anaesthetic procedures and four had presented adverse reactions in prior surgeries. Conclusions: In our study the most common causes of anaphylaxis were NMBAs (62.5%) and latex (18.7%), in line with that described in the literature. In 81% of patients with anaphylactic reactions, skin tests were positive to at least one anaesthetic drug or to latex. A high incidence of severe anaphylaxis was found, probably because cases with less severe clinical signs were not referred to our clinic. The results emphasise the need for systematic evaluation of anaphylactic reactions during anaesthesia. This can be achieved by the creation of a specialised Allergo-Anaesthesia Clinic, as has been the case in our Department

    A historical overview, although the existing literature default the management consulting as a extraordinary and a unique phenomenon in the corporate context, in truth these statements do not come accompanied by a number of academic studies that emphasize the importance of effective work of management consultants. To combat the lack of studies in this emerging area, this article aims to understand whether it may be in fact the importance of knowledge produced in this sector is associated with the type of relationships that are built between the parties and, noting that the successful relationships are the basis of the knowledgeproduced in the sector who will then be responsible for the balance of powers and who should take control of projects.     Breeding Madeiran Storm-petrels Oceanodroma castro on Farilhão Grande islet was first described in the early 80s. Despite being an important breeding colony at nationally and global level for the species distribution, few studies regarding monitoring and/or research have been developed. In order to update the knowledge about this population, SPEA, under the FAME project, made 7 visits to the islet between 2011 and 2012. We used mist-nets, nocturnal vocalizations and ground search to assess the population size. The population was estimated to be 473 individuals, resulting in 102-201 breeding pairs, which may indicate a high population decreasing. Comparison with previous studies emphasizes the need to conduct a directed census in order to obtain more robust estimate of this population. For the first time it was used a tracking device (GLS) in Madeiran Storm-petrels to assess movements and distribution of the species during the breeding season. The only GLS recovered showed that the individual made a 5 days trip over open sea to the west coast of Morocco. The walls built in 2002, during an action performed previously by SPEA, which host some Madeiran Storm-petrel nests, were rebuilt. We also implemented 36 new artificial nests that will enable future work with this population. A census of Yellow-legged Gull Larus michahellis that breeds on the islet took place, and 170-180 pairs were estimated. During the several visits to the island, it was detected gulls like signs of predation on Madeiran Storm-petrel. This effect however will be better reported later

    Nanofiltration of Cork Wastewaters and Their Possible Use in Leather Industry as Tanning Agents

    Cork processing wastewater is a very complex mixture of vegetal extracts and has, among other natural compounds, a very high content of phenolic/tannic colloidal matter that is responsible for severe environmental problems. In the present work, the concentration of this wastewater by nanofiltration was investigated with the aim of producing a cork tannin concentrate to be utilized in tanning. Permeation results showed that the permeate fluxes are controlled by both osmotic pressure and fouling/gel layer phenomena, leading to a rapid decrease of permeate fluxes with the concentration factor. The rejection coefficients to organic matter were higher than 95%, indicating that nanofiltration has a very good ability to concentrate the tannins and produce a permeate stream depleted from organic matter. The cork tannin concentrate obtained by nanofiltration and evaporation had total solids concentration of 34.8 g/l. The skins tanned by this concentrate were effectively converted to leather with a shrinking temperature of 7 degrees C

    Association Between Memory B-Cells and Clinical and Immunological Features of Primary Sjögren’s Syndrome and Sicca Patients

    B-cells play a pivotal role in primary Sjögren's syndrome (pSS) pathogenesis. We aim to (1) evaluate the distribution of B-lymphocyte subpopulations in pSS and Sicca patients, (2) establish cut-off points that discriminate pSS from controls, (3) evaluate the association between memory B-cells and phenotypic features in pSS. We included 57 pSS patients, 68 Sicca and 24 healthy controls. Circulating B-cells were characterized by flow cytometry as naïve and memory subsets and classified from Bm1 to Bm5. Compared to controls, pSS patients had lower percentages (29.5 vs 44.4%) and absolute numbers (47 vs 106 cells/µl) of memory B-cells. Through ROC curves, a cut-off of ≤ 58 total memory B-cells/µl yielded a specificity of 0.88 and a sensitivity of 0.60 for pSS, and was met by 59.6% of pSS patients, 38.8% of Sicca and 12.5% of controls. A cut-off of < 23.5 Switched-memory B-cells/µl yielded a specificity of 0.88 and a sensitivity of 0.54 and was met by 54.4% of pSS patients, 37.3% of Sicca and 12.5% of controls. In pSS, lower total memory B-cells count was associated with longer disease duration (14.3 vs 8.1 years, p = 0.006) and more active disease profile, as evaluated by the European League Against Rheumatism (EULAR) Sjögren's Syndrome Disease Activity Index (ESSDAI) (3.1 vs 1.4, p = 0.043). Decreased numbers of memory B-cells clearly discriminated pSS from controls and can also have prognostic value. It remains to be clarified whether Sicca patients with decreased memory B-cells represent pSS and if B-cell profiling could help in the diagnosis of pSS.info:eu-repo/semantics/publishedVersio

    Pioglitazone Prevents Capillary Rarefaction in Streptozotocin-Diabetic Rats Independently of Glucose Control and Vascular Endothelial Growth Factor Expression

    Background/Aims: Reduction of capillary network density occurs early in the development of metabolic syndrome and may be relevant for the precipitation of diabetes. Agonists of the peroxisome proliferator-activated receptor (PPAR)-gamma transcription factor are vasculoprotective, but their capacity for structural preservation of the microcirculation is unclear. Methods: Male Wistar rats were rendered diabetic by streptozotocin and treated with pioglitazone in chow for up to 12 weeks. Capillary density was determined in heart and skeletal muscle after platelet endothelial cell adhesion molecule-1 (PECAM-1) immunostaining. Hallmarks of apoptosis and angiogenesis were determined. Results: Capillary density deteriorated progressively in the presence of hyperglycemia (from 971/mm(2) to 475/mm(2) in quadriceps muscle during 13 weeks). Pioglitazone did not influence plasma glucose, left ventricular weight, or body weight but nearly doubled absolute and relative capillary densities compared to untreated controls (1.2 vs. 0.6 capillaries/myocyte in heart and 1.5 vs. 0.9 capillaries/myocyte in quadriceps muscle) after 13 weeks of diabetes. No antiapoptotic or angiogenic influence of pioglitazone was detected while a reduced expression of hypoxia-inducible factor-3 alpha and PPAR coactivator-1 alpha (PGC-1 alpha) mRNA as well as vascular endothelial growth factor (VEGF) protein possibly occurred as a consequence of improved vascularization. Conclusion: Pioglitazone preserves microvascular structure in diabetes independently of improvements in glycemic control and by a mechanism unrelated to VEGF-mediated angiogenesis.
