Toward Building Self-Sustaining Groups in PCR-based Tasks through Implicit Coordination: The Case of Heuristic Evaluation
Usability flaws found in the later stages of the software development process can be extremely costly to resolve. Accordingly, usability evaluation (UE) is an important, albeit usually expensive, part of development. We report on how the inexpensive UE method of heuristic evaluation (HE) can benefit from collaborative software (CSW), implicit coordination, and principles from collaboration engineering. In our study, 439 novice participants were trained in HE methods and then performed HE. Our results show that traditional nominal HE groups can experience implicit coordination through the collaborative software features of group memory and group awareness. One of the key results is that CSW groups had less duplication of effort than traditional nominal groups; these differences were magnified as group size increased from three to six members. Furthermore, because they coordinated less, traditional nominal groups performed more work in the overall process of HE. We attribute the reduction in duplication for CSW-supported groups to the implicit coordination available to them; CSW-supported groups could see violations input by other group members, but could not directly discuss the violations. These findings not only show the power of implicit coordination in groups, but should dramatically change how HE is conducted. These results may also extend to other evaluation tasks, such as software inspection and usability assessment tasks

Ultradian glucocorticoid exposure directs gene-dependent and tissue-specific mRNA expression patterns in vivo
In this paper we report differential decoding of the ultradian corticosterone signal by glucocorticoid target tissues. Pulsatile corticosterone replacement in adrenalectomised rats resulted in different dynamics of Sgk1 mRNA production, with a distinct pulsatile mRNA induction profile observed in the pituitary in contrast to a non-pulsatile induction in the prefrontal cortex (PFC). We further report the first evidence for pulsatile transcriptional repression of a glucocorticoid-target gene in vivo, with pulsatile regulation of Pomc transcription in pituitary. We have explored a potential mechanism for differences in the induction dynamics of the same transcript (Sgk1) between the PFC and pituitary. Glucocorticoid receptor (GR) activation profiles were strikingly different in pituitary and prefrontal cortex, with a significantly greater dynamic range and shorter duration of GR activity detected in the pituitary, consistent with the more pronounced gene pulsing effect observed. In the prefrontal cortex, expression of Gilz mRNA was also non-pulsatile and exhibited a significantly delayed timecourse of increase and decrease when compared to Sgk1, additionally highlighting gene-specific regulatory dynamics during ultradian glucocorticoid treatment

Building a collaborative culture in cardiothoracic operating rooms: Pre and postintervention study protocol for evaluation of the implementation of teamSTEPPS training and the impact on perceived psychological safety
IntroductionThe importance of effective communication, a key component of teamwork, is well recognised in the healthcare setting. Establishing a culture that encourages and empowers team members to speak openly in the cardiothoracic (CT) operating room (OR) is necessary to improve patient safety in this high-risk environment.Methods and analysisThis study will take place at Barnes-Jewish Hospital, an academic hospital in affiliation with Washington University School of Medicine located in the USA. All team members participating in cardiac and thoracic OR cases during this 17-month study period will be identified by the primary surgical staff attending on the OR schedule.TeamSTEPPS (Team Strategies and Tools to Enhance Performance and Patient Safety) training course will be taught to all CT OR staff. Before TeamSTEPPS training, staff will respond to a 39-item questionnaire that includes constructs from the Agency for Healthcare Research and Quality Hospital Survey on Patient Safety Culture, Edmondson’s ‘Measure of psychological safety’ questionnaire, and questionnaires on turnover intentions, job satisfaction and ‘burnout’. The questionnaires will be readministered at 6 and 12 months.The primary outcomes to be assessed include the perceived psychological safety of CT OR team members, the overall effect of TeamSTEPPS on burnout and job satisfaction, and observed turnover rate among the OR nurses. As secondary outcomes, we will be assessing self-reported rates of medical error and near misses in the ORs with a questionnaire at the end of each case.Ethics and disseminationEthics approval is not indicated as this project does not meet the federal definitions of research requiring the oversight of the Institutional Review Board (IRB). Patient health information (PHI) will not be generated during the implementation of this project. Results of the trial will be made accessible to the public when published in a peer-reviewed journal following the completion of the study.</jats:sec
The Rise and Fall of Debris Disks: MIPS Observations of h and chi Persei and the Evolution of Mid-IR Emission from Planet Formation
We describe Spitzer/MIPS observations of the double cluster, h and
Persei, covering a 0.6 square-degree area surrounding the cores of both
clusters. The data are combined with IRAC and 2MASS data to investigate
616 sources from 1.25-24 . We use the long-baseline -[24] color
to identify two populations with IR excess indicative of circumstellar
material: Be stars with 24 excess from optically-thin free free
emission and 17 fainter sources (J 14-15) with [24] excess consistent
with a circumstellar disk. The frequency of IR excess for the fainter sources
increases from 4.5 through 24 . The IR excess is likely due to
debris from the planet formation process. The wavelength-dependent behavior is
consistent with an inside-out clearing of circumstellar disks. A comparison of
the 24 excess population in h and Per sources with results for
other clusters shows that 24 emission from debris disks 'rises' from 5
to 10 Myr, peaks at 10-15 Myr, and then 'falls' from 15/20 Myr to

Violation of a Leggett-Garg inequality with ideal non-invasive measurements
The quantum superposition principle states that an entity can exist in two
different states simultaneously, counter to our 'classical' intuition. Is it
possible to understand a given system's behaviour without such a concept? A
test designed by Leggett and Garg can rule out this possibility. The test,
originally intended for macroscopic objects, has been implemented in various
systems. However to-date no experiment has employed the 'ideal negative result'
measurements that are required for the most robust test. Here we introduce a
general protocol for these special measurements using an ancillary system which
acts as a local measuring device but which need not be perfectly prepared. We
report an experimental realisation using spin-bearing phosphorus impurities in
silicon. The results demonstrate the necessity of a non-classical picture for
this class of microscopic system. Our procedure can be applied to systems of

Achieving Acetylcholine Receptor Clustering in Tissue-Engineered Skeletal Muscle Constructs In vitro through a Materials-Directed Agrin Delivery Approach
Volumetric muscle loss (VML) can result from trauma, infection, congenital anomalies, or surgery, and produce permanent functional and cosmetic deficits. There are no effective treatment options for VML injuries, and recent advances toward development of muscle constructs lack the ability to achieve innervation necessary for long-term function. We sought to develop a proof-of-concept biomaterial construct that could achieve acetylcholine receptor (AChR) clustering on muscle-derived cells (MDCs) in vitro. The approach consisted of the presentation of neural (Z+) agrin from the surface of microspheres embedded with a fibrin hydrogel to muscle cells (C2C12 cell line or primary rat MDCs). AChR clustering was spatially restricted to areas of cell (C2C12)-microsphere contact when the microspheres were delivered in suspension or when they were incorporated into a thin (2D) fibrin hydrogel. AChR clusters were observed from 16 to 72 h after treatment when Z+ agrin was adsorbed to the microspheres, and for greater than 120 h when agrin was covalently coupled to the microspheres. Little to no AChR clustering was observed when agrin-coated microspheres were delivered from specially designed 3D fibrin constructs. However, cyclic stretch in combination with agrin-presenting microspheres led to dramatic enhancement of AChR clustering in cells cultured on these 3D fibrin constructs, suggesting a synergistic effect between mechanical strain and agrin stimulation of AChR clustering in vitro. These studies highlight a strategy for maintaining a physiological phenotype characterized by motor endplates of muscle cells used in tissue engineering strategies for muscle regeneration. As such, these observations may provide an important first step toward improving function of tissue-engineered constructs for treatment of VML injuries
