    Sojourner expectations: Are they met and does it matter if they’re not?

    This research examines the association between cross-cultural travelers’ well-being and the discrepancy between expected and experienced adaptation, in a longitudinal study of intercultural exchange students (N = 1762; Mage = 17 years; 70 % female). Specifically, two competing hypotheses were tested. Whereas the accuracy hypothesis suggests that unmet expectations lead to poorer outcomes, the directional hypothesis proposes that the outcome of unmet expectations will depend on whether expectations are overmet or undermet. Both sojourners’ adaptation expectations (pre-travel) and adaptation experiences (during the sojourn) were measured alongside general well-being. Controlling for baseline, well-being during the sojourn was regressed on the direction (undermet or overmet) and the magnitude (extent of discrepancy) of the adaptation expectation-experience discrepancies. These analyses were conducted across time (at entry or 5 months into the sojourn), different types of adaptation (psychological and sociocultural adaptation), and different measures of well-being (stress and satisfaction with life). Across analyses, results indicate that the direction of mismatch appears to matter little for small mismatches. However, for larger mismatches a positive effect emerged for sojourner well-being when the experience was better than expected, but a negative effect when the experience was worse than expected. Overall, the results support a directional hypothesis

    Choose your (international) contacts wisely: A multilevel analysis on the impact of intergroup contact while living abroad

    A two year longitudinal study examined the influence of social contact abroad on student sojourners? cultural adjustment and intergroup affect. Social contact was measured with regard to the three people that sojourners (exchange students) spent most time with. Both the quality of the relationship with each contact and the cultural background (host national, co-national) of each contact were measured, that is contact quality and contact source, respectively. The data were analysed through repeated measures multilevel modelling. Results indicate that good quality contact (independent of source) is associated with higher cultural adjustment in sojourners and lower levels of stress. Contact quality however, was not associated with intergroup affect. Source of contact was found to matter, especially over time. Specifically, in the later stages of the sojourn, having more co-national contacts among ones three most frequent contacts, was associated with higher levels of stress, reduced cultural adjustment and higher secondary outgroup derogation. These findings suggest that who sojourners have contact with, matters for both cultural adjustment and intergroup affect

    A tight spot: How personality moderates the impact of social norms on sojourner adaptation

    When living abroad, how do you navigate the norms of your new culture? Taking an interactionalist perspective, we examine how contextual factors and personality traits jointly affect sojourner's adaptation. We hypothesized that tightness (strong, rigidly imposed norms) of the host culture would be associated with lower levels of adaptation, and that tightness of the home culture would be associated with higher levels of adaptation. Further, we proposed that the impact of tightness should be dependent on personality traits associated with navigating social norms (agreeableness, conscientiousness, and honesty-humility). Longitudinal data were analyzed from intercultural exchange students (N = 889) travelling from and to 23 different countries. Multilevel Modeling showed that sojourners living in a tighter culture had poorer adaptation than those in a looser culture. In contrast, sojourners originating from a tighter culture showed better adaptation. The negative effect of cultural tightness was moderated by agreeableness and honesty-humility, but not conscientiousness

    Advancing our understanding of the EU sports policy: the socio-cultural model of sports regulation and players’ agents

    This paper explains the development of the socio-cultural dimension of the European Union (EU) sports policy over the course of the decade from 2005 to 2015. By adopting the theoretical lenses of the Advocacy Coalition Framework (ACF), the paper firstly offers a novel perspective on the belief systems of the members of the socio-cultural advocacy coalition that operates within the EU sports policy subsystem. A set of empirical policy core beliefs of the coalition actors reflecting their concerns regarding to sport’s integrity and economic welfare that affect its healthy development is presented and explained as the basic causes of the policy problem, i.e., threats that undermine sport’s specific characteristics. In this light, the paper then illustrates that the coalition members have perceived the issue of players’ agents, in particular problems in the activities of agents in European football, to be detrimental to the integrity of sport. As a result, the issue has been the subject of a considerable policy activity by the coalition actors with a view to establishing an alternative regulatory framework to effectively govern players’ agents. Consequently, the paper elucidates the evolving nature of the socio-cultural model of sports regulation that not only promotes the social role of sport in Europe but now also addresses the problems that affect its well-being and clear image. The paper’s principal method of investigation is based on the content analysis of official policy documents and statements of the coalition actors

    He Was not Alone: Bosman in Context

    This chapter analyses the social, political, historical and economic context of the Bosman case. The chapter argues that Bosman needs to be understood as yet another stage of a continued process of commercialisation and transformation in European football. Football players fought to transform their employment conditions since the 1960s. Clubs questioned the legitimacy of UEFA to regulate European football and organise club competitions as they wanted a larger share of the commercial profits of the game. Finally, political institutions in Brussels started to exert pressure on football governing bodies to modify the international transfer system. The chapter argues that, taking these into account, major transformations in the governance and regulation structures of football were needed, and they would have happened even without the Bosman ruling. Consequently, Bosman cannot be seen, on its own, as the only cause of the transformation of modern football in Europe. Finally, the chapter dedicates some space to Jean Marc Bosman’s legal team composed by Luc Misson and Jean Louis Dupont. It was their legal expertise what made a challenge before the Belgian and European courts also possible

    EU sports law: a uniform algorithm for regulatory rules

In applying the EU economic provisions to the regulatory rules in sport, four different categories of “sporting exception” can be discerned in the jurisprudence of the Court. They include sporting rules that do not produce any economic effect, ‘purely sporting’ rules, inherent rules, and objectively justified rules. Based on the existing parameters of the EU sports law and policy, this article advances arguments in support of discarding the nuances in the Court’s analytical approach to sporting exception. Ordinary EU law, coupled by the concept of specificity of sport that is now included in Article 165(1) TFEU, already contains the all-encompassing, uniform analytical structure apt to accommodate all categories of regulatory rules in sports. In addition, the proposed uniform framework can be often be utilised to justify the challenged sporting rules in both internal market law and competition law, thus avoiding duplication of analysis. This is enabled by the high degree of convergence in their application to the rules of private regulatory bodies

Utilizing the hepatitis B virus (HBV) transgenic mouse model of chronic infection, we demonstrate that the nuclear receptor, hepatocyte nuclear factor 4α (HNF4α, NR2A1), is essential for viral biosynthesis in the liver. The dependency of HBV transcription on HNF4α links viral biosynthesis and persistence to a developmentally regulated transcription factor essential for host viability

    A Novel Function of Apolipoprotein E: Upregulation of ATP-Binding Cassette Transporter A1 Expression

    Despite the well known importance of apolipoprotein (Apo) E in cholesterol efflux, the effect of ApoE on the expression of ATP-binding cassette transporter A1 (ABCA1) has never been investigated. The objective of this study was to determine the effect of ApoE on ApoB-carrying lipoprotein-induced expression of ABCA1, a protein that mediates cholesterol efflux. Our data demonstrate that ApoB-carrying lipoproteins obtained from both wild-type and ApoE knockout mice induced ApoAI-mediated cholesterol efflux in mouse macrophages, which was associated with an enhanced ABCA1 promoter activity, and an increased ABCA1 mRNA and protein expression. In addition, these lipoproteins increased the level of phosphorylated specificity protein 1 (Sp1) and the amount of Sp1 bound to the ABCA1 promoter. However, all these inductions were significantly diminished in cells treated with ApoE-free lipoproteins, when compared to those treated with wild-type lipoproteins. Enrichment with human ApoE3 reversed the reduced inducibility of ApoE-free lipoproteins. Moreover, we observed that inhibition of Sp1 DNA-binding by mithramycin A diminished ABCA1 expression and ApoAI-mediated cholesterol efflux induced by ApoB-carrying lipoproteins, and that mutation of the Sp1-binding motif in the ABCA1 promoter region diminished ApoB-carrying lipoprotein-induced ABCA1 promoter activity. Collectively, these data suggest that ApoE associated with ApoB-carrying lipoproteins has an upregulatory role on ABCA1 expression, and that induction of Sp1 phosphorylation is a mechanism by which ApoE upregulates ABCA1 expression

    Cell line-dependent variability in HIV activation employing DNMT inhibitors

    Long-lived reservoirs of Human Immunodeficiency Virus (HIV) latently infected cells present the main barrier to a cure for HIV infection. Much interest has focused on identifying strategies to activate HIV, which would be used together with antiretrovirals to attack reservoirs. Several HIV activating agents, including Tumor Necrosis Factor alpha (TNFα) and other agents that activate via NF-kB are not fully effective in all latent infection models due to epigenetic restrictions, such as DNA methylation and the state of histone acetylation. DNA methyltransferases (DNMT) inhibitors like 5-aza-2'deoxycytidine (Aza-CdR) and histone deacetylase (HDAC) inhibitors like Trichostatin A (TSA) have been proposed as agents to enhance reactivation and have shown activity in model systems. However, it is not clear how the activities of DNMT and HDAC inhibitors range across different latently infected cell lines, potential models for the many different latently infected cells within an HIV patient. We determined HIV activation following treatment with TNFα, TSA and Aza-CdR across a range of well known latently infected cell lines. We assessed the activity of these compounds in four different Jurkat T cell-derived J-Lat cell lines (6.3, 8.4, 9.2 and 10.6), which have a latent HIV provirus in which GFP replaces Nef coding sequence, and ACH-2 and J1.1 (T cell-derived), and U1 (promonocyte-derived) cell lines with full-length provirus. We found that Aza-CdR plus TNFα activated HIV at least twice as well as TNFα alone for almost all J-Lat cells, as previously described, but not for J-Lat 10.6, in which TNFα plus Aza-CdR moderately decreased activation compared to TNFα alone. Surprisingly, a much greater reduction of TNFα-stimulated activation with Aza-CdR was detected for ACH-2, J1.1 and U1 cells. Reaching the highest reduction in U1 cells with a 75% reduction. Interestingly, Aza-CdR not only decreased TNFα induction of HIV expression in certain cell lines, but also decreased activation by TSA. Since DNMT inhibitors reduce the activity of provirus activators in some HIV latently infected cell lines the use of epigenetic modifying agents may need to be carefully optimized if they are to find clinical utility in therapies aimed at attacking latent HIV reservoirs
