    Origins and structure of social and political attitudes: insights from personality system theory and behavioural genetics

    People differ, often strikingly, in their views on desired social structures and processes. For example, while some value ethnic diversity in their society, others believe non -indigenous individuals (whatever that might mean) should be repatriated to their land of origin. Similarly, whereas some believe religion should play no role in determining social policy, others strongly advocate the importance of living according to religious scripture, including at a social level. This variation in attitudes, and its implication for societal cohesion, has made research on the origins of social and political attitudes of enduring interest to psychologists, sociologists, political scientists, among many others.The goal of the current thesis was to extend work in this literature in two key ways: Firstly, I examined whether political attitudes can be understood within a personality system model. This work addresses previous mixed results on the links of basic personality traits to political conservatism. In Chapter 3, I test predictions from this model; namely, that direct influences on political behaviour flow from moral values, with personality mostly acting indirectly via these moral values, rather than directly affecting political attitudes. Findings from two studies (published as Lewis & Bates, 2011a) supported these predictions suggesting that the new model helps explain inconsistencies in previous research attempting to link personality to political orientation that have not included the intermediary level of values.Secondly, I examined the genetic architecture of social attitudes constructs in three separate studies. Chapter 4 addressed whether in -group favouritism reflects heritable effects, and, secondly, whether race -favouritism was accounted for broad or specific genetic effects. Results indicated that a common biological mechanism exists facilitating generalised favouritism, with evidence for additional genetic effects specific to each form of group favouritism. These findings (published as Lewis & Bates, 2010) suggest that (at least) at the genetic level, race favouritism is multiply determined.In Chapter 5, I examined whether prosocial obligations across the domains of welfare, work, and civic obligation share a common genetic basis, or reflect specific heritable components (published as Lewis & Bates, 2011b). In females, results indicated the existence of a common heritable factor underlying each of these prosocial obligations. In males, a prosocial factor was also observed; familial effects (genetic and shared -environment effects were indistinguishable) influenced this general mechanism. At the domain -specific level, modest genetic effects were observed in females for civic and work obligations, with shared - environment effects influencing welfare obligations. In males, genetic influences were observed for welfare obligation, with unique -environments affecting work and civic duty.Finally, in Chapter 6, I present work examining the genetic architecture of religious belief. Although genetic factors are known to influence strength of religious belief, the psychological mechanism(s) through which this biological influence is manifest are presently unknown. Two non -theological constructs - 1) need for community integration and 2) need for existential certainty - were hypothesised to account for the genetic effects on religiosity. The results supported this hypothesis, with genetic influences on these traits wholly accounting for the heritable basis of religiosity, suggesting that religion "re- uses" systems involved in meeting both social and existential needs

    PUBLIC CONSERVATION LAND AND EMPLOYMENT GROWTH IN THE NORTHERN FOREST REGION

    As with many environmental issues, debates about increasing public conservation lands in the Northern Forest region frequently center on a perceived tradeoff between jobs and the environment. In particular, opponents of conservation lands often argue that employment will decline significantly when land is diverted from commodity-oriented uses such as wood products production. To evaluate this claim, we estimate a model of simultaneous employment and net migration growth using data on the 92 non-metropolitan counties comprising the region. Growth in employment and net migration are measured over the period 1990 to 1997 and the set of exogenous variables includes the 1990 share of the county land base in public conservation uses. We find that net migration rates were systematically higher in counties with more conservation lands, but the effects are relatively small. Public conservation lands were found to have no systematic effect on employment growth over the 1990 to 1997 period. Two extensions are considered. We examine the separate effects of preservationist and multiple-use lands. We also identify a "natural experiment" involving changes in national forest management that allows us to estimate the effects of diverting private forestland to public conservation uses. Our central conclusions are that existing public conservation lands have a positive, but small, effect on employment and migration in the Northern Forest region and that, over the range of our data, employment and migration are unlikely to be affected by timber harvest reductions resulting from the establishment of new conservation lands.Labor and Human Capital, Land Economics/Use,

    In-plane effects on segmented-mirror control

    Extremely large optical telescopes are being designed with primary mirrors composed of hundreds of segments. The “out-of-plane” piston, tip, and tilt degrees of freedom of each segment are actively controlled using feedback from relative height measurements between neighboring segments. The “in-plane” segment translations and clocking (rotation) are not actively controlled; however, in-plane motions affect the active control problem in several important ways, and thus need to be considered. We extend earlier analyses by constructing the “full” interaction matrix that relates the height, gap, and shear motion at sensor locations to all six degrees of freedom of segment motion, and use this to consider three effects. First, in-plane segment clocking results in height discontinuities between neighboring segments that can lead to a global control system response. Second, knowledge of the in-plane motion is required both to compensate for this effect and to compensate for sensor installation errors, and thus, we next consider the estimation of in-plane motion and the associated noise propagation characteristics. In-plane motion can be accurately estimated using measurements of the gap between segments, but with one unobservable mode in which every segment clocks by an equal amount. Finally, we examine whether in-plane measurements (gap and/or shear) can be used to estimate out-of-plane segment motion; these measurements can improve the noise multiplier for the “focus-mode” of the segmented-mirror array, which involves pure dihedral angle changes between segments and is not observable with only height measurements

    Rapid E2-E3 Assembly and Disassembly Enable Processive Ubiquitylation of Cullin-RING Ubiquitin Ligase Substrates

    Degradation by the ubiquitin-proteasome system requires assembly of a polyubiquitin chain upon substrate. However, the structural and mechanistic features that enable template-independent processive chain synthesis are unknown. We show that chain assembly by ubiquitin ligase SCF and ubiquitin-conjugating enzyme Cdc34 is facilitated by the unusual nature of Cdc34-SCF transactions: Cdc34 binds SCF with nanomolar affinity, nevertheless the complex is extremely dynamic. These properties are enabled by rapid association driven by electrostatic interactions between the acidic tail of Cdc34 and a basic ‘canyon’ in the Cul1 subunit of SCF. Ab initio docking between Cdc34 and Cul1 predicts intimate contact between the tail and the basic canyon, an arrangement confirmed by crosslinking and kinetic analysis of mutants. Basic canyon residues are conserved in both Cul1 paralogs and orthologs, suggesting that the same mechanism underlies processivity for all cullin-RING ubiquitin ligases.     Hypoxia as a target for drug combination therapy of liver cancer

    Hepatocellular carcinoma (HCC) is the third most frequentcause of cancer deaths worldwide. The standard of care for intermediate HCC is transarterial chemoembolization, which combines tumour embolization with locoregional delivery of the chemotherapeutic doxorubicin. Embolization therapies induce hypoxia, leading to the escape and proliferation of hypoxia-adapted cancer cells. The transcription factor that orchestrates responses to hypoxia is hypoxia-inducible factor 1 (HIF-1). The aim of this work is to show that targeting HIF-1 with combined drug therapy presents an opportunity for improving outcomes for HCC treatment. HepG2 cells were cultured under normoxic and hypoxic conditions exposed to doxorubicin, rapamycin and combinations thereof, and analyzed for viability and the expression of hypoxia-induced HIF-1α in response to these treatments. A pilot study was carried out to evaluate the antitumour effects of these drug combinations delivered from drug-eluting beads in vivo using an ectopic xenograft murine model of HCC. A therapeutic doxorubicin concentration that inhibits the viability of normoxic and hypoxic HepG2 cells and above which hypoxic cells are chemoresistant was identified, together with the lowest effective dose of rapamycin against normoxic and hypoxicHepG2 cells. It was shown that combinations of rapamycinand doxorubicin are more effective than doxorubicin alone. Western Blotting indicated that both doxorubicin and rapamycin inhibit hypoxia-induced accumulation of HIF-1α.     Systems approach to the improvement of engine warm-up behaviour

    Modifications to the coolant and oil circuits of a modern production 2.4 l diesel engine have been made in an attempt to promote oil warm-up to reduce fuel consumption. The new system used oil to cool exhaust gas recirculation (EGR) gases and incorporates a number of coolant flow control valves to reduce heat loss during warm-up. The engine was run over cold-start New European Drive Cycles with various flow strategies as a screening exercise to understand the behaviour of the system. Fuel consumption benefits of up to 4 per cent were observed, but these were accompanied by 3 per cent increases in nitrogen oxide (NO x) emissions. Detailed analysis of the coolant flows and temperatures showed that, when throttling the flow, the mass of coolant in the degas bottle and radiator could be isolated from the system during warm-up, essentially reducing the thermal inertia. Heat transfer directly to the oil from the EGR gases rather than via the coolant allowed more heat to be put into the oil, with engine oil supply temperatures up to 6 °C hotter; however, it was not possible to verify that the oil was hotter at the bearings, valve train, and cylinder liner. The engine strategy was seen to react to the faster warm-up and to retard injection timing, reducing NO x but also compromising overall fuel consumption benefits. Further tests were conducted with various injection timings to establish a NO x—fuel consumption trade-off to demonstrate further benefits when the engine strategy is included in the operation of novel thermal management systems. </jats:p
