    Supraphysiological Oxygen Levels in Mammalian Cell Culture: Current State and Future Perspectives

    Most conventional incubators used in cell culture do not regulate O2 levels, making the headspace O2 concentration ~18%. In contrast, most human tissues are exposed to 2-6% O2 (physioxia) in vivo. Accumulating evidence has shown that such hyperoxic conditions in standard cell culture practices affect a variety of biological processes. In this review, we discuss how supraphysiological O2 levels affect reactive oxygen species (ROS) metabolism and redox homeostasis, gene expression, replicative lifespan, cellular respiration, and mitochondrial dynamics. Furthermore, we present evidence demonstrating how hyperoxic cell culture conditions fail to recapitulate the physiological and pathological behavior of tissues in vivo, including cases of how O2 alters the cellular response to drugs, hormones, and toxicants. We conclude that maintaining physioxia in cell culture is imperative in order to better replicate in vivo-like tissue physiology and pathology, and to avoid artifacts in research involving cell culture

    The conservation status of mammals and avifauna in the Montagne des Français massif, Madagascar

    The Montagne des Français is a limestone massif in northern Madagascar, which is characterised by a wide range of biotopes including xerophytic karst, gallery forest, dry western forest, grassland and caves. It is situated only 12 km from the regional capital, Antsiranana, and few, if any areas of primary forest remain. In the first comprehensive study to have been carried out at this location we report the presence of 12 mammal species. We also report the presence of 63 bird species. We use data derived from both structured and semi-structured interviews to assess the conservation status of the mammals and birds within the massif. Our study shows that local beliefs are dominated by taboos or fady and that these vary within families and communities. Current anthropogenic pressures on biodiversity include zebu grazing, charcoal production, hunting and rice cultivation. The massif was afforded Temporary Protected Area Status in 2006 and our results suggest that this protection should be made permanent. We propose opportunities for further research and sustainable development initiatives that could contribute to the conservation of the biological resources within the massif. Success in conserving this area will only be achieved if the local communities are fully engaged. RÉSUMÉ La Montagne des Français est un massif calcaire au nord de Madagascar, caractĂ©risĂ© par une vaste gamme de biotopes, y compris une formation calcaire connue localement sous le nom de ‘tsingy’ avec une vĂ©gĂ©tation xĂ©rophyte, une forĂȘt riveraine, une forĂȘt sĂšche de l’ouest, des zones herbeuses et des grottes. Elle se trouve Ă  12 km seulement de la plus grande ville du nord, Antsiranana, et prĂ©sente une couverture de forĂȘts intactes extrĂȘmement rĂ©duite. Le travail sur le terrain a Ă©tĂ© rĂ©alisĂ© par des bĂ©nĂ©voles de Frontier et des chercheurs de Frontier et de l’UniversitĂ© d’Antsiranana. Des inventaires on Ă©tĂ© effectuĂ©s pendant une annĂ©e, au cours de quatre pĂ©riodes d’essais qui s’étalaient chacune sur une durĂ©e de l’ordre de neuf semaines. Les inventaires sur les mammifĂšres ont fait appel Ă  trois mĂ©thodes, dont les lignes de trous-piĂšges, l’emploi de piĂšges Sherman et des recherches nocturnes alĂ©atoires. Un inventaire sur les oiseaux a Ă©tĂ© compilĂ©, utilisant la technique de la liste de recensement McKinnon. Au cours de cette premiĂšre Ă©tude dĂ©taillĂ©e portant sur cette localitĂ©, nous avons relevĂ© la prĂ©sence de 12 espĂšces de mammifĂšres ainsi que la prĂ©sence de 63 espĂšces d’oiseaux. Neuf des espĂšces de mammifĂšres recensĂ©s sont endĂ©miques Ă  Madagascar et la plupart de ces espĂšces semblaient ĂȘtre reprĂ©sentĂ©es par des effectifs rĂ©duits sur la Montagne des Français au cours de la pĂ©riode d’étude. De toutes les espĂšces d’oiseaux reportĂ©es, 26 (41% ) sont endĂ©miques de Madagascar. Afin d’évaluer l’état de conservation des mammifĂšres et des oiseaux rencontrĂ©s Ă  la Montagne des Français, nous avons utilisĂ© des donnĂ©es recueillies au cours d’entrevues formelles et semi-formelles. Notre Ă©tude montre que les croyances locales sont dominĂ©es par des tabous ou fady et que ceux-ci varient selon les familles et communautĂ©s. Les pressions anthropogĂ©niques actuelles qui pĂšsent sur la biodiversitĂ© sont reprĂ©sentĂ©es par le pĂąturage des zĂ©bus, la production de charbon de bois, la chasse et la culture de riz. En 2006 le massif a bĂ©nĂ©ficiĂ© d’un statut d’Aire ProtĂ©gĂ©e Temporaire mais nos rĂ©sultats suggĂšrent qu’un statut de protection permanente serait justifiĂ©. Nous proposons de poursuivre les efforts en matiĂšre de recherche et encourageons toute entreprise de dĂ©veloppement durable qui pourrait contribuer Ă  la conservation des ressources biologiques Ă  l’intĂ©rieur du massif. La rĂ©ussite de la protection de cette rĂ©gion ne pourra se faire sans la totale adhĂ©sion de l’ensemble des communautĂ©s locales

    Adipose tissue from metabolic syndrome mice induces an aberrant miRNA signature highly relevant in prostate cancer development

    Prostate cancer (PCa) remains an important public health concern in Western countries. Metabolic syndrome (MeS) is a cluster of pathophysiological disorders with increasing prevalence in the general population that is a risk factor for PCa. Several studies have determined that a crosstalk between white adipose tissue (WAT) and solid tumors favors cancer aggressiveness. In this work, our main goal was to investigate the interaction between WAT and PCa cells through microRNAs (miRNAs), in MeS mice. We developed a MeS-like disease model using C57BL/6J mice chronically fed with high-fat diet (HFD) that were inoculated with TRAMP-C1 PCa cells. A group of five miRNAs (mmu-miR-221-3p, 27a-3p, 34a-5p, 138-5p, and 146a-5p) were increased in gonadal WAT (gWAT), tumors, and plasma of MeS mice compared to control animals. Three of these five miRNAs were detected in the media from gWAT and TRAMP-C1 cell cocultures, and significantly increased in MeS context. More importantly, hsa-miR-221-3p, 146a-5p, and 27a-3p were increased in bloodstream of PCa patients compared to healthy donors. Using miRNA microarrays, we found that 121 miRNAs were differentially released to the coculture media between HFD-gWAT and tumor cells compared to control diet-gWAT and tumor cells. Target genes for the 66 most deregulated miRNAs were involved in common pathways, mainly related to fatty acid metabolism, ER protein processing, amino acid degradation, PI3K AKT signaling, and PCa. Our findings show for the first time a signature of five miRNAs as important players involved in the interaction between WAT and PCa in MeS mice. Further research will be necessary to track these miRNAs in the interaction between these tissues as well as their role in PCa patients with MeS.Fil: Massillo, Cintia Lorena.     CTBP1 and metabolic syndrome induce an mRNA and miRNA expression profile critical for breast cancer progression and metastasis

    Metastatic breast cancer (BrCa) is still one of the main causes of cancer death in women. Metabolic syndrome (MeS), a risk factor for BrCa, is associated to high grade tumors, increased metastasis and recurrence of this disease. C-terminal binding protein 1 (CTBP1) is a co-repressor of tumor suppressor genes that is activated by low NAD+/NADH ratio. Previously, we demonstrated that CTBP1 hyperactivation by MeS increased tumor growth in MDA-MB-231-derived xenografts regulating several genes and miRNAs. In this work, our aim was to elucidate the role of CTBP1 and MeS in BrCa metastasis. We found that CTBP1 protein diminished adhesion while increased migration of triple negative BrCa cells. CTBP1 and MeS modulated the expression of multiple genes (ITGB4, ITGB6, PRSS2, COL17A1 and FABP4) and miRNAs (miR-378a-3p, miR-146a-5p, let-7e-3p, miR-381-5p, miR-194-5p, miR-494- 3p) involved in BrCa progression of MDA-MB-231-derived xenografts. Furthermore, we demonstrated that MeS increased lung micrometastasis and liver neoplastic disease in mice. CTBP1 hyperactivation seems to be critical for MeS effect on BrCa metastasis since CTBP1 depletion completely impaired the detection of circulating tumor cells.     CTBP1/CYP19A1/Estradiol axis together with adipose tissue impacts over prostate cancer growth associated to metabolic syndrome

    Metabolic syndrome (MeS) increases prostate cancer (PCa) risk and aggressiveness. Cterminal binding protein 1 (CTBP1) is a transcriptional co-repressor of tumor suppressor genes that is activated by low NAD+/NADH ratio. Previously, our group established a MeS and PCa mice model that identified CTBP1 as a novel link associating both diseases. We found that CTBP1 controls the transcription of aromatase (CYP19A1), a key enzyme that converts androgens to estrogens. The aim of this work was to investigate the mechanism that explains CTBP1 as a link between MeS and PCa based on CYP19A1 and estrogen synthesis regulation using PCa cell lines, MeS/PCa mice and adipose co-culture systems. We found that CTBP1 and E1A binding protein p300 (EP300) bind to CYP19A1 promoter and downregulate its expression in PC3 cells. Estradiol, through the estrogen receptor beta, released CTBP1 from CYP19A1 promoter triggering its transcription and modulating PCa cell proliferation. We generated NSG and C57BL/6J MeS mice by chronically feeding animals with high fat diet. In the NSG model, CTBP1 depleted PCa xenografts showed an increase in the CYP19A1 expression with the subsequent increment in intratumor estradiol concentrations. Additionally, in C57BL/6J mice, MeS induces hypertrophy, hyperplasia and inflammation of the white adipose tissue, which leads to a proinflammatory phenotype and increases serum estradiol concentration. Thus, MeS increased PCa growth and Ctbp1, Fabp4 and IL-6 expression levels. These results describe, for the first time, a novel CTBP1/CYP19A1/Estradiol axis that explains, in part, the mechanism for prostate tumor growth increase by MeS.Fil: Massillo, Cintia Lorena.     Social determinants of health and the double burden of disease in Nepal: a secondary analysis.

    BACKGROUND: As the global burden of disease evolves, lower-resource countries like Nepal face a double burden of non-communicable and infectious disease. Rapid adaptation is required for Nepal's health system to provide life-long, person-centred care while simultaneously improving quality of infectious disease services. Social determinants of health be key in addressing health disparities and could direct policy decisions to promote health and manage the disease burden. Thus, we explore the association of social determinants with the double burden of disease in Nepal. METHODS: This is a retrospective, ecological, cross-sectional analysis of infectious and non-communicable disease outcome data (2017 to 2019) and data on social determinants of health (2011 to 2013) for 753 municipalities in Nepal. Multinomial logistic regression was conducted to evaluate the associations between social determinants and disease burden. RESULTS: The 'high-burden' combined double burden (non-communicable and infectious disease) outcome was associated with more accessible municipalities, (adjOR3.94[95%CI2.94-5.28]), municipalities with higher proportions of vaccine coverage (adjOR12.49[95%CI3.05-51.09]) and malnutrition (adjOR9.19E103[95%CI19.68E42-8.72E164]), lower average number of people per household (adjOR0.32[95%CI0.22-0.47]) and lower indigenous population (adjOR0.20[95%CI0.06-0.65]) compared to the 'low-burden' category on multivariable analysis. 'High-burden' of non-communicable disease was associated with more accessible municipalities (adjOR1.93[95%CI1.45-2.57]), higher female proportion within the municipality (adjOR1.69E8[95%CI3227.74-8.82E12]), nutritional deficiency (adjOR1.39E17[95%CI11799.83-1.64E30]) and malnutrition (adjOR2.17E131[95%CI4.41E79-1.07E183]) and lower proportions of population under five years (adjOR1.05E-10[95%CI9.95E-18-0.001]), indigenous population (adjOR0.32[95%CI0.11-0.91]), average people per household (adjOR0.44[95%CI0.26-0.73]) and households with no piped water (adjOR0.21[95%CI0.09-0.49]), compared to the 'low-burden' category on adjusted analysis. 'High burden' of infectious disease was also associated with more accessible municipalities (adjOR4.29[95%CI3.05-6.05]), higher proportions of population under five years (adjOR3.78E9[95%CI9418.25-1.51E15]), vaccine coverage (adjOR25.42[95%CI7.85-82.29]) and malnutrition (adjOR4.29E41[95%CI12408.29-1.48E79]) and lower proportions of households using firewood as fuel (adjOR0.39[95%CI0.20-0.79]) ('moderate-burden' category only) compared to 'low-burden'. CONCLUSIONS: While this study produced imprecise estimates and cannot be interpreted for individual risk, more accessible municipalities were consistently associated with higher disease burden than remote areas. Female sex, lower average number per household, non-indigenous population and poor nutrition were also associated with higher burden of disease and offer targets to direct interventions to reduce the burden of infectious and non-communicable disease and manage the double burden of disease in Nepal

    CtBP1 associates metabolic syndrome and breast carcinogenesis targeting multiple miRNAs

    Metabolic syndrome (MeS) has been identified as a risk factor for breast cancer. C-terminal binding protein 1 (CtBP1) is a co-repressor of tumor suppressor genes that is activated by low NAD+/NADH ratio. High fat diet (HFD) increases intracellular NADH. We investigated the effect of CtBP1 hyperactivation by HFD intake on mouse breast carcinogenesis. We generated a MeS-like disease in female mice by chronically feeding animals with HFD. MeS increased postnatal mammary gland development and generated prominent duct patterns with markedly increased CtBP1 and Cyclin D1 expression. CtBP1 induced breast cancer cells proliferation. Serum from animals with MeS enriched the stem-like/progenitor cell population from breast cancer cells. CtBP1 increased breast tumor growth in MeS mice modulating multiple genes and miRNA expression implicated in cell proliferation, progenitor cells phenotype, epithelial to mesenchymal transition, mammary development and cell communication in the xenografts.     The Development of Videoconference-Based Support for People Living With Rare Dementias and Their Carers: Protocol for a 3-Phase Support Group Evaluation

    BACKGROUND: People living with rarer dementias face considerable difficulty accessing tailored information, advice, and peer and professional support. Web-based meeting platforms offer a critical opportunity to connect with others through shared lived experiences, even if they are geographically dispersed, particularly during the COVID-19 pandemic. OBJECTIVE: We aim to develop facilitated videoconferencing support groups (VSGs) tailored to people living with or caring for someone with familial or sporadic frontotemporal dementia or young-onset Alzheimer disease, primary progressive aphasia, posterior cortical atrophy, or Lewy body dementia. This paper describes the development, coproduction, field testing, and evaluation plan for these groups. METHODS: We describe a 3-phase approach to development. First, information and knowledge were gathered as part of a coproduction process with members of the Rare Dementia Support service. This information, together with literature searches and consultation with experts by experience, clinicians, and academics, shaped the design of the VSGs and session themes. Second, field testing involved 154 Rare Dementia Support members (people living with dementia and carers) participating in 2 rounds of facilitated sessions across 7 themes (health and social care professionals, advance care planning, independence and identity, grief and loss, empowering your identity, couples, and hope and dementia). Third, a detailed evaluation plan for future rounds of VSGs was developed. RESULTS: The development of the small groups program yielded content and structure for 9 themed VSGs (the 7 piloted themes plus a later stages program and creativity club for implementation in rounds 3 and beyond) to be delivered over 4 to 8 sessions. The evaluation plan incorporated a range of quantitative (attendance, demographics, and geography; pre-post well-being ratings and surveys; psycholinguistic analysis of conversation; facial emotion recognition; facilitator ratings; and economic analysis of program delivery) and qualitative (content and thematic analysis) approaches. Pilot data from round 2 groups on the pre-post 3-word surveys indicated an increase in the emotional valence of words selected after the sessions. CONCLUSIONS: The involvement of people with lived experience of a rare dementia was critical to the design, development, and delivery of the small virtual support group program, and evaluation of this program will yield convergent data about the impact of tailored support delivered to geographically dispersed communities. This is the first study to design and plan an evaluation of VSGs specifically for people affected by rare dementias, including both people living with a rare dementia and their carers, and the outcome of the evaluation will be hugely beneficial in shaping specific and targeted support, which is often lacking in this population
