Get PDF

Get PDF

Get PDF
Motivation: Ebola virus (EBOV) is considered as the prototypical pathogen of viral hemorrhagic fever, causing severe disease and mortality rates as high as 90%. The implementation of different High-Throughput Screening systems to avoid the use of BSL-4 facilities will promote the generation of lead molecules and so the development of new therapeutics for EBOV within the next few years. The goals of the present project are directed to the discovery of new antiviral drugs against EBOV by using HTS of different libraries of small compounds, some of which have previously been an excellent source of potent antiviral compounds, by the use of a retroviral pseudotype particle (pEBOV-GFP) carrying the EBOV glycoprotein (GP). Methods: Retroviral pseudotypes were generated by the transfection with the HIV gag-pol plasmid, pCMV-Δ8.91, the EBOV GP expression construct, pCAGGS-EBOV, and the green fluorescence protein (GFP) reporter construct, pCSGW at a ratio of 1.1:0.5:0.75 (core:envelope:reporter). At we carryed out at the IBiS the screening of different libraries of small compounds of different nature. The libraries of small compounds to be screened were synthesized by: i) the Medicinal Chemistry Institute of Madrid (IQM-CSIC), ii) the Unit of Molecular Chemistry and Pharmacology of the Institute for Research in Biomedicine of Barcelona (IRB), and iii) the Group of Organic Chemistry at the Faculty of Pharmacy of the University of Seville. Antiviral activivity was evaluated by pEBOV-mediated GFP expression. Results: After the evaluation of 60 compounds, only compounds 4, 10, 17, 23, 33, 34, 125, 126, 279, 284, 298, 372, 385, 414 and 42 showed a significant (> 50%) inhibition of pEBOV-GFP entry (50 µM), presenting values of cytotoxicity (CC50) greater than 100 mM, therefore considered safe. Compounds 21, 371, 82, despite showing optimal inhibition percentages at 50 µM, presented CC50 values lower than our 100 µM threshold and because of that they were excluded from further evaluations. Conclusions: Although the antiviral activity of molecules identified by these system has to be confirmed against the wild-type EBOV, it is a useful tool to identify anti-EBOV candidates that also solves the necessity of work in BSL-4 facilities. Compounds identified in this work could potentially represent strong hit compounds for the development of a treatment for the EBOV disease

Estrategias Docentes Para la Internacionalización del Alumnado Universitario

Get PDF
Effectiveness of the Ponseti method in the treatment of clubfoot: a systematic review

Get PDF
Clubfoot is a common congenital deformity of the lower limbs. It should be treated as soon as possible so that its correction is more easily achieved. The objective of this systematic review was to assess the effectiveness of the Ponseti method in the treatment of clubfoot. A bibliographic search was carried out in different databases, including PubMed and SciELO. Filters such as full text and randomized controlled trial were selected to find those articles that best matched our search. Among the results, we selected the ones that interested us, and the rest were discarded, either because they did not meet the requirements for our work or because they were repeated. In total, we collected 19 articles, but after using the critical evaluation instrument CASPe, 7 of them were eliminated, leaving us with a total of 12 articles for our systematic review. After analyzing the results obtained in the selected articles, we concluded that the Ponseti method is effective in the treatment of clubfoot, presenting a high success rate.Partial funding for open access charge: Universidad de Málag

© 2021 The Author(s).New therapeutic targets are revolutionizing colorectal cancer clinical management, opening new horizons in metastatic patients’ outcome. Polo Like Kinase1 (PLK1) inhibitors have high potential as antitumoral agents, however, the emergence of drug resistance is a major challenge for their use in clinical practice. Overcoming this challenge represents a hot topic in current drug discovery research. BI2536-resistant colorectal cancer cell lines HT29R, RKOR, SW837R and HCT116R, were generated in vitro and validated by IG50 assays and xenografts models by the T/C ratio. Exons 1 and 2 of PLK1 gene were sequenced by Sanger method. AXL pathway, Epithelial-to-Mesenchymal transition (EMT) and Multidrug Resistance (MDR1) were studied by qPCR and western blot in resistant cells. Simvastatin as a re-sensitizer drug was tested in vitro and the drug combination strategies were validated in vitro and in vivo. PLK1 gene mutation R136G was found for RKOR. AXL pathway trough TWIST1 transcription factor was identified as one of the mechanisms involved in HT29R, SW837R and HCT116R lines, inducing EMT and upregulation of MDR1. Simvastatin was able to impair the mechanisms activated by adaptive resistance and its combination with BI2536 re-sensitized resistant cells in vitro and in vivo. Targeting the mevalonate pathway contributes to re-sensitizing BI2536-resistant cells in vitro and in vivo, raising as a new strategy for the clinical management of PLK1 inhibitors. This study has been funded by Instituto de Salud Carlos III (ISCIII) -Fondos FEDER proyects PI16/01468 and PI19/01231

[Abstract] The Podiatric Health Questionnaire (PHQ) is a foot health psicometric tool consisting of six items for measuring foot health status. Currently, PHQ has been adapted into Spanish. Thus, this research focused on develop the Persian translation and adaptation of the PHQ. The translation into Persian and test-retest reliability methods were applied from english version questionnarie. As regards to the total mark for each sub-scale, internal consistency and reliability were determined by the Cronbach α and intraclass correlation coefficient (ICC) with a confidence interval (CI) of 95%. High internal consistency was shown for the six sub-scales: a) walk with a Cronbach α of 0.775; b) hygiene and c) nail care with 0.789 and 0.796 respectively; d) foot on pain with 0.779; e) concern domain with 0.788; and f) quality of life with 0.798; and g) visual analogic scale with 0.803. Excellent test-retest reliability (ICC = 0.912 [95% CI = 0.845–0.950]) was shown for the total score. Conclusions: The persian version of the PHQ was shown to be a valid and reliable tool for an acceptable use in the Iran population

Dark Matter Annual Modulation Results from the ANAIS-112 Experiment

Get PDF
An annual modulation in the interaction rate of galactic dark matter particles is foreseen due to Earth’s movement around the Sun; the DAMA/LIBRA observation of a modulation signal compatible with expectations has intrigued the community for twenty years. The ANAIS-112 experiment, with a target of 112.5 kg of NaI(Tl), has been running smoothly at the Canfranc Underground Laboratory (Spain) since 2017. It aims to test this observation using the same detection technique and target. Results on the modulation search from two years of data (220.7 kg·y) have been presented and the analysis of three years (313.6 kg·y) is underway. Under the hypothesis of modulation, the deduced amplitudes from best fits are in all cases compatible with zero for the two energy regions at [2–6] and [1–6] keV; the results agree with the expected sensitivity for the considered exposure and fully support the goal of achieving a 3σ sensitivity to explore the DAMA/LIBRA result for a five-year operation. Here, the ANAIS-112 set-up and performance will be briefly recapped and the annual modulation results and prospects will be discussed
