    Determinación de neofitadieno en grasa subcutánea de cerdo Ibérico con diferentes sistemas de alimentación

    This study was carried out in order to determine the relationship between the content of a branched hydrocarbon (neophytadiene) in the subcutaneous fat and the feeding system of Iberian pigs during the fattening period previous to slaughter. Thirty-six batches of Iberian pigs, belonging to the four categories of feeding systems described in the Spanish legislation (Bellota, Recebo, Campo and Cebo) were studied. These categories differ in their degree of utilization of the natural resources (acorn and pasture) of the extensive rearing system or in their level of replacement by concentrated feed. SPE combined with GC-MS techniques were carried out to isolate and to identify hydrocarbons. Recebo and Bellota batches showed the highest levels of neophytadiene content, followed by Campo batches, and finally, batches of the Cebo category. The results showed a great variability between batches from the same feeding category, mainly from Bellota, Recebo and Campo categories, which makes the classification of the animals in these three categories difficult. However, the animals from the Cebo category were correctly classified when reared in intensive conditions.El objetivo del trabajo se centró en determinar el contenido de un hidrocarburo ramificado (neofitadieno) en la grasa subcutánea del cerdo ibérico, y su relación con el tipo de alimentación recibida durante la etapa de cebo previa al sacrificio (en la fase final de cebo). Para ello, se estudiaron treinta y seis lotes de cerdos Ibéricos, pertenecientes a las cuatro categorías de calidad en base a la alimentación de los animales establecidas en la Norma de Calidad (Bellota, Recebo, Campo y Cebo). Estas categorías se diferencian en el grado de aprovechamiento de los recursos naturales de la dehesa (bellotas y hierba) y en el nivel de sustitución de los mismos por piensos concentrados. Para el aislamiento e identificación de los hidrocarburos se utilizaron técnicas de SPE combinadas con GC-MS. Los lotes de Recebo y Bellota presentaron los mayores niveles de neofitadieno, seguidos por los lotes de Campo, y por último los lotes de Cebo. Se observó una gran variabilidad en los niveles de neofitadieno entre los lotes pertenecientes a una misma categoría de alimentación, sobre todo en los lotes de Bellota, Recebo y Campo, por lo que es difícil una correcta clasificación de los animales en las tres categorías en base al contenido en neofitadieno. Sin embargo, la clasificación dentro de la categoría de cebo se hace de forma correcta cuando los cerdos son producidos en condiciones intensivas

    Iberian pig fat characteristics depend on the type of feeding at the end of its finish-fattening period. The routine analysis to differentiate among the qualities of the feeding types given to the pigs in the fattening stage has been the use of fatty acid profiles by gas chromatography. Due to de doubts about the effectiveness of this analysis in the montanera period, the aim of this global study was to test the validity of various analytical methods to determine the feeding type of Iberian pigs, focusing on the fatty acid profile. Three montanera periods with a total of 749 samples from 38 batches have been studied; using a total of 144 dry-cured shoulder shanks, 99 of which are of known pig origin. Results showed that the determination of the fatty acid profile using gas chromatography is not a consistent method to classify the animals according to diet in the recebo category, although it provided good percentages of success for classifying the bellota and cebo categories.Las características de la grasa de cerdo Ibérico dependen del tipo de alimentación recibida en el último estadío de engorde. El análisis que se ha utilizado hasta ahora para diferenciar las diferentes calidades de alimentación de los cerdos en este período ha sido el análisis de los perfiles de ácidos grasos de la grasa por técnicas de cromatografía de gases. Debido a las dudas sobre la efectividad de esta técnica en la montanera, el objetivo del proyecto global (RTA2008-0026) fue probar la validez de varios métodos analíticos para determinar el tipo de alimentación del cerdo ibérico, centrándonos en este trabajo en el estudio de los perfiles de ácidos grasos. Para el desarrollo de este estudio se utilizaron tres campañas de montanera con un total de 749 muestras de 38 partidas, y con 144 paletas de las cuales 99 tenían una trazabilidad completa. Los resultados mostraron que la determinación de la alimentación de los cerdos ibéricos usando el análisis del perfil de ácidos grasos no es un método consistente para clasificar los animales de acuerdo a la categoría de recebo, mientras que para las categorías de bellota y cebo, los resultados encontrados mostraron unos buenos porcentajes de éxito

    The geology of Cuba: A brief overview and synthesis

    Cuba is the largest island in the Greater Antilles, and its geology records three important episodes: (1) the Jurassic breakup of North and South America (Pangea) and associated passive margin and oceanic sedimentary and magmatic evolution; (2) the sedimentary, magmatic, and metamorphic evolution of an intra-oceanic Cretaceous-Paleogene ophiolite-arc complex; and (3) the Paleogene 'soft collision' and transfer of the NW Caribbean plate (and Cuba) to the North American plate. Thick sequences of Jurassic-Cretaceous strata (conglomerates, sandstones, limestones, dolo­stones, shales) and interlayered basaltic rocks characterize passive margin sequences preserved in the Guaniguanico terrane (western Cuba, related to the Mayan passive margin and the Gulf of Mexico) and the Bahamas Platform borderlands (north of Cuba). Passive margin deposition ceased in latest Cretaceous time, when increasing relief of accreted (overriding) oceanic arc and ophiolite complexes shed coarse sediments (olistostrome and flysch), followed by carbonate deposition. Fragments of the intervening oceanic lithosphere (Proto-Caribbean, connected to the Central Atlantic) and fore- and back-arc oceanic lithosphere (Caribbean, of Pacific origin) occur as tectonic fragments detached from the ophiolitic units, including serpentinized harzburgites and dunites, banded and isotropic gabbros, basalts (tholeiitic and fore-arc basalts, locally with boninites) and Late Jurassic (Tithonian) through Late Cretaceous (Coniacian and younger) oceanic sediments. Arc activity in the Cuban segment of the Greater Antilles produced sedimentary, volcanic, and plutonic rocks during Cretaceous times (ca. 135-70 Ma). A new arc developed in eastern Cuba during Paleocene-middle Eocene times. Cuban arc sequences include island-arc tholeiitic, calcalkaline, and alkaline bimodal suites of volcanic and plutonic rocks. Remnants of Proto-Caribbean oceanic lithosphere occur as exhumed mélange-bearing eclogite-, blueschist-, and garnet-amphibolite-facies tectonic blocks (oldest age ca. 120 Ma) within a serpentinite matrix intercalated with, or at the base of, the overthrusted ophiolitic bodies. Cuban Cretaceous arc magmatic activity ended due to the subduction of Proto-Caribbean passive margin sequences of the Caribeana terrane, an offshore protuberance of Yucatan. This event formed strongly deformed high-pressure meta­sedimentary and metaigneous rocks at ca. 70 Ma, when the Caribbean plate began to collide with North America. The collision, which included overriding of the ophiolitic and arc units over both subducted and unsubducted passive margin sequences, also produced synorogenic basins and filled them, a process that continued until ca. 40 Ma. This foldbelt was succeeded by local uplift and subsidence to form late Eocene-Recent unconformable post-orogenic continental basins

    Trace-elemental and multi-isotopic (Sr-Nd-Pb) discrimination of jade in the circum-Caribbean: Implications for pre-colonial inter-island exchange networks

    Dense and strong, hydrothermal-metasomatic jadeitite and jadeite-omphacite rocks were used as tools and adornments throughout the wider Caribbean since initial inhabitation. Regionally, rich sources of jadeitite and jadeite-omphacite jade are known only in Guatemala (north and south of the Motagua Fault Zone), eastern Cuba and the northern Dominican Republic, establishing that humans transported jadeitic material over vast distances. This study validates that geochemical fingerprinting is a viable provenance method for Caribbean pre-colonial jadeitic lithologies. An assemblage of 101 source rocks has been characterised for trace element and combined Sr-Nd-Pb isotope compositions. Four statistical approaches (Principal Component Analysis, t-Distributed Stochastic Neighbour Embedding, Decision Tree, and Multiclass Regression) were assessed, employing source-distinct trace element ratios. A multiclass regression technique based on trace element ratios of immobile high field strength, light to medium rare earth and fluid-mobile, large-ion-lithophile elements is shown to be most effective in discriminating the four source regions. Ninety-one % of the Guatemalan samples can be discriminated from the Dominican and Cuban sources using La/Th, Zr/Hf and Y/Th ratios. Jadeitic rocks cropping out in the Dominican Republic can be distinguished from Cuban jades employing Er/Yb, Nb/Ta and Ba/Rb ratios with 71% certainty. Furthermore, the two Guatemala sources, north and south of the Motagua Fault Zone, can be discriminated by using (among others) Zr/Hf, Ta/Th, La/Sm and Dy/Y ratios with an 89% success rate. This raises the possibility of determining, in detail, former trading and mobility networks between different islands and the Meso- and Central American mainland within the Greater Caribbean. The provenance technique was applied to 19 pre-colonial jade celts excavated from the Late Ceramic Age Playa Grande archaeological site in the northern Dominican Republic. Three artefacts are discriminated as derived from the Guatemalan source, indicating that, despite a source of jade within 25 km, material was traded from Guatemala.     Diversity across major and candidate genes in European local pig breeds

    The aim of this work was to analyse the distribution of causal and candidate mutations associated to relevant productive traits in twenty local European pig breeds. Also, the potential of the SNP panel employed for elucidating the genetic structure and relationships among breeds was evaluated. Most relevant genes and mutations associated with pig morphological, productive, meat quality, reproductive and disease resistance traits were prioritized and analyzed in a maximum of 47 blood samples from each of the breeds (Alentejana, Apulo-Calabrese, Basque, Bísara, Majorcan Black, Black Slavonian (Crna slavonska), Casertana, Cinta Senese, Gascon, Iberian, Krskopolje (Krskopoljski), Lithuanian indigenous wattle, Lithuanian White Old Type, Mora Romagnola, Moravka, Nero Siciliano, Sarda, Schwa-bisch-Hallisches Schwein (Swabian Hall pig), Swallow-Bellied Mangalitsa and Turopolje). We successfully analyzed allelic variation in 39 polymorphisms, located in 33 candidate genes. Results provide relevant information regarding genetic diversity and segregation of SNPs associated to production and quality traits. Coat color and morphological trait-genes that show low level of segregation, and fixed SNPs may be useful for traceability. On the other hand, we detected SNPs which may be useful for association studies as well as breeding programs. For instance, we observed predominance of alleles that might be unfavorable for disease resistance and boar taint in most breeds and segregation of many alleles involved in meat quality, fatness and growth traits. Overall, these findings provide a detailed catalogue of segregating candidate SNPs in 20 European local pig breeds that may be useful for traceability purposes, for association studies and for breeding schemes. Population genetic analyses based on these candidate genes are able to uncover some clues regarding the hidden genetic substructure of these populations, as the extreme genetic closeness between Iberian and Alentejana breeds and an uneven admixture of the breeds studied. The results are in agreement with available knowledge regarding breed history and management, although largest panels of neutral markers should be employed to get a deeper understanding of the population’s structure and relationships

    Predicting Acorn-Grass Weight Gain Index using non-destructive Near Infrared Spectroscopy in order to classify Iberian pig carcasses according to feeding regime

    The classification of Iberian pig carcasses into different commercial categories according to feeding regime was evaluated by means of a non-destructive analysis of the subcutaneous adipose tissue using Near Infrared Spectroscopy (NIRS). A quantitative approach was used to predict the Acorn-Grass Weight Gain Index (AGWGI), and a set of criteria was established for commercial classification purposes. A total of 719 animals belonging to various batches, reflecting a wide range of feeding regimes, production systems and years, were analyzed with a view to developing and evaluating quantitative NIRS models. Results for the external validation of these models indicate that NIRS made clear differentiation of batches as a function of three feeding regimes possible with high accuracy (<i>Acorn, Recebo</i> and <i>Feed</i>), on the basis of the mean representative spectra of each batch. Moreover, individual analysis of the animals showed a broad consensus between field inspection information and the classification based on the AGWGI NIRS prediction, especially for extreme categories (<i>Acorn</i> and <i>Feed</i>).<br><br>La clasificación en distintas categorías comerciales según régimen alimenticio de canales de cerdo Ibérico fue evaluada mediante el análisis no destructivo de muestras de tejido adiposo subcutáneo por Espectroscopía del Infrarrojo Cercano (NIRS). Partiendo de una aproximación cuantitativa para predecir el Índice de Reposición en Montanera (IRM) se establecieron una serie de criterios para proceder a su clasificación comercial. Se analizaron un total de 719 animales pertenecientes a diversas partidas, que recogen una amplia variabilidad de muestras de distintos regímenes alimenticios, campañas y sistemas productivos, para el desarrollo y evaluación de los modelos NIRS cuantitativos. Los resultados de validación externa de los modelos indicaron que es posible discriminar con una gran exactitud entre partidas de distintos categorías (<i>Bellota, Recebo</i> y <i>Cebo</i>), en base al espectro medio representativo de cada partida. Además, el análisis individualizado de los animales mostró un amplio consenso entre la información recibida de campo y la clasificación en base a la predicción del parámetro IRM por NIRS, sobre todo para categorías con características extremas (<i>Bellota</i> y <i>Cebo</i>)

    Genome‐wide detection of copy number variants in European autochthonous and commercial pig breeds by whole‐genome sequencing of DNA pools identified breed‐characterising copy number states

    In this study, we identified copy number variants (CNVs) in 19 European autochthonous pig breeds and in two commercial breeds (Italian Large White and Italian Duroc) that represent important genetic resources for this species. The genome of 725 pigs was sequenced using a breed‐specific DNA pooling approach (30–35 animals per pool) obtaining an average depth per pool of 42×. This approach maximised CNV discovery as well as the related copy number states characterising, on average, the analysed breeds. By mining more than 17.5 billion reads, we identified a total of 9592 CNVs (~683 CNVs per breed) and 3710 CNV regions (CNVRs; 1.15% of the reference pig genome), with an average of 77 CNVRs per breed that were considered as private. A few CNVRs were analysed in more detail, together with other information derived from sequencing data. For example, the CNVR encompassing the KIT gene was associated with coat colour phenotypes in the analysed breeds, confirming the role of the multiple copies in determining breed‐specific coat colours. The CNVR covering the MSRB3 gene was associated with ear size in most breeds. The CNVRs affecting the ELOVL6 and ZNF622 genes were private features observed in the Lithuanian Indigenous Wattle and in the Turopolje pig breeds respectively. Overall, the genome variability unravelled here can explain part of the genetic diversity among breeds and might contribute to explain their origin, history and adaptation to a variety of production systems.info:eu-repo/semantics/acceptedVersio
