The silent information regulator protein (Sir2) and its homologs are NAD+-dependent deacetylase enzymes that play important roles in a variety of physiological processes. However, the functions of the Sir2 family in plants are poorly understood. Here, we report that Arabidopsis AtSRT2, a homolog of yeast Sir2, negatively regulates plant basal defense against the pathogen Pseudomonas syringae pv. tomato DC3000 (PstDC3000). In response to PstDC3000 infection, the expression of AtSRT2 was down-regulated in a salicylic acid (SA)-independent manner. In addition, knock-out of AtSRT2 (srt2) enhanced resistance against PstDC3000 and increased expression of pathogenesis-related gene 1 (PR1). Conversely, overexpression of AtSRT2 resulted in hypersusceptibility to PstDC3000 and impaired PR1 induction. Consistent with this phenotype, expression of PAD4, EDS5 and SID2, three essential genes in the SA biosynthesis pathway, were increased in the srt2 mutant and decreased in AtSRT2-overexpressing plants. Taken together, these results demonstrate that AtSRT2 is a negative regulator of basal defense, possibly by suppressing SA biosynthesis

Sevoflurane Pre-conditioning Ameliorates Diabetic Myocardial Ischemia/Reperfusion Injury Via Differential Regulation of p38 and ERK.

Diabetes mellitus (DM) significantly increases myocardial ischemia/reperfusion (MI/R) injury. During DM, cardioprotection induced by conventional pre-conditioning (PreCon) is decreased due to impaired AMP-activated protein kinase (AMPK) signaling. The current study investigated whether PreCon with inhaled anesthetic sevoflurane (SF-PreCon) remains cardioprotective during DM, and identified the involved mechanisms. Normal diet (ND) and high-fat diet (HFD)-induced DM mice were randomized into control and SF-PreCon (3 cycles of 15-minute period exposures to 2% sevoflurane) groups before MI/R. SF-PreCon markedly reduced MI/R injury in DM mice, as evidenced by improved cardiac function (increased LVEF and Ā±Dp/dt), decreased infarct size, and decreased apoptosis. To determine the relevant role of AMPK, the effect of SF-PreCon was determined in cardiac-specific AMPKĪ±2 dominant negative expressing mice (AMPK-DN). SF-PreCon decreased MI/R injury in AMPK-DN mice. To explore the molecular mechanisms responsible for SF-PreCon mediated cardioprotection in DM mice, cell survival molecules were screened. Interestingly, in ND mice, SF-PreCon significantly reduced MI/R-induced activation of p38, a pro-death MAPK, without altering ERK and JNK. In DM and AMPK-DN mice, the inhibitory effect of SF-PreCon upon p38 activation was significantly blunted. However, SF-PreCon significantly increased phosphorylation of ERK1/2, a pro-survival MAPK in DM and AMPK-DN mice. We demonstrate that SF-PreCon protects the heart via AMPK-dependent inhibition of pro-death MAPK in ND mice. However, SF-PreCon exerts cardioprotective action via AMPK-independent activation of a pro-survival MAPK member in DM mice. SF-PreCon may be beneficial compared to conventional PreCon in diabetes or clinical scenarios in which AMPK signaling is impaired

The incidence of ovarian cancer in the UK has increased by almost twenty percent since the 1970ā€™s and the majority of cases are not diagnosed until the late stages, when metastasis is more likely to have occurred. Focal Adhesion Kinase (FAK) is one of the key protein complexes which is integral to cell migration and has been linked to a variety of solid tumours. ShenLingLan (SLDM) is a traditional herbal medicine which has been formulated for the treatment of solid tumours. This study aimed to establish the impact of SLDM on FAK in ovarian cancer cells in vitro and transcript levels of FAK in an ovarian cancer cohort. FAK and paxillin phosphorylation events stimulated by SLDM treatment were identified using a Kinexusā„¢ antibody based protein array. The impact of SLDM on cell attachment and migration was evaluated using Electric cell-substrate impedance sensing (ECIS), whilst the changes in focal adhesion complex localisation were assessed using immunofluorescence. In an ovarian cancer cohort, differences in FAK and paxillin transcript levels were assessed against key clinical parameters such as differentiation, stage and survival outcome. SLDM treatment of ovarian cancer cells in vitro resulted in the suppression of FAK and paxillin phosphorylation at several sites, which appeared to manifest as decreased cellular attachment and migration in a range of immortalised ovarian cancer cells. Increased FAK and paxillin transcript copies were observed in high grade and poorly differentiated ovarian tumours as well as in tumours from patients with ovarian cancer related incidence. SLDM has a profound effect on the migratory and adhesive properties of ovarian cancer cells, potentially via inhibitory effects on the activation of the FAK pathway, which is aberrant in clinical ovarian cancers

Impact of E-Commerce and Digital Marketing Adoption on the Financial and Sustainability Performance of MSMEs during the COVID-19 Pandemic

    Get PDF
The COVID-19 pandemic has remarkably affected the business processes and performance of micro-, small-, and medium-sized enterprises (MSMEs) across the world. MSMEs have had to adopt and implement numerous strategies to sustain their businesses, and their financial and sustainability performance has been impacted by their choice of e-commerce (EC) platforms and digital marketing (DM) strategies. The objective of this research was to explore the effects of EC and DM platforms and strategies on facilitating MSMEsā€™ financial and sustainability performance amid the devastating COVID-19 pandemic. This study gathered data from 212 MSMEs from three districts of Bangladesh. A partial least squares structural equation modeling (PLS-SEM) approach was undertaken, to test the hypothesized model. The findings revealed that e-commerce had a significant association with MSMEsā€™ financial performance and sustainability amid the pandemic. It was also observed that digital marketing strategies had a substantial impact on MSMEsā€™ financial performance. However, the linkage between DM strategies and MSMEsā€™ sustainability was found to be insignificant. Furthermore, it was found that the financial performance of MSMEs mediated the relationship between e-commerce adoption and their sustainability performance. These findings contribute to the extant technology adoption literature, by exploring the role of e-commerce and digital marketing on firmsā€™ financial outcomes amid a global pandemic. Managers and policymakers of small businesses can learn several things from this study, and understand how crucial digital commerce and digital marketing are to their success and long-term survival

Background: Fresh pine needles, which evergreen, renewable and can be collected at any time, have abundant essential oil. The pine needle essential oil possess various biological activities, including antimicrobial, antioxidant, anti-inflammatory, cardioprotective properties etc., and can be widely used as medicine or medical raw materials, fragrances etc. So in order to make full use of pine needle plant resources, especially essential oil, the identification and quantification of essential oil were investigated. Materials and Methods: GC-MS was used to identified compounds in pine needle essential oil of Pinus massoniana Lamb. from Zhejiang, and GC method was developed for determining 5 compounds (namely Ī±-pinene, Ī²-pinene, limonene, bornyl acetate, Ī²-caryophyllene) in pine needle essential oil from different habitats of Pinus massoniana Lamb. and different species (Pinus koraiensis Sieb. et Zucc., Pinus sylvestris var. mongolica Litv., Abies holophylla Maxim.). Hierarchical clustering analysis (HCA) was used to evaluate homogeneity of pine needles in China. Results: 42 compounds(monoterpenes and sesquiterpenes) were identified by GC-MS, which accounted for 99.62% of total essential oil, particularly contained Ī±-and Ī²-pinene (45.23%). Quantification results showed content of bornyl acetate were the lowest, while content of Ī±-pinene, Ī²-pinene were all high except Sichuan, among them Ī±-pinene were higher than Ī²-pinene except for Shandong and Jiangsu, and total content of essential oil in Guangxi is the highest, Jiangsu was the lowest within Pinus massoniana from different habitats. Within pine needle from different species, the highest total content was Pinus koraiensis, the lowest was Pinus sylvestris, Interestingly, the highest content of bornyl acetate was Abies holophylla. By HCA, samples were sorted into two clusters, Abies holophylla and the other, that is Abies holophylla is different from Pinus massoniana, Pinus koraiensis and Pinus sylvestris. Pinus massoniana from Sichuan is different from other Pinus, in which limonene was the highest in all samples. Conclusions: Compounds of pine needle essential oil vary greatly between genus, while smaller between species, so composition differences among pine needle essential oil was related with genus and species. Abies holophylla is different from other pinus species, and Pinus massoniana from Sichuan is different from other habitats. Pine needle essential oil contains abundant Ī±-and Ī²-pinene, which can be used as an alternative raw materials source of pinene. And Abies holophylla and Pinus massoniana from Sichuan can become bornyl acetate and limonene alternative source of raw materials

BackgroundThe measurement of the coping difficulties of parents of premature infants after discharge provides objective data for nurses to prepare infants for discharge. However, no Chinese scale has been developed to measure parentsā€™ coping difficulties after their premature infants are discharged.AimTo translate the parent version of the Post-Discharge Coping Difficulty Scale (Ped-PDCDS) from English to Chinese and test the reliability and validity of the Chinese version in parents of premature infants.MethodsA multicenter cross-sectional study of 356 parents of premature infants was conducted. The scale was symmetrically translated. Validity was evaluated in terms of content, construct, discriminant, and convergent validities. Reliability was assessed in terms of internal consistency, split-half reliability, and testā€“retest reliability.ResultsThe Chinese Ped-PDCDS finally contained 11 items. Exploratory and confirmatory factor analyses results showed that the Chinese Ped-PDCDS had three dimensions, and the convergent and discriminant validities of the scale was satisfactory. The overall reliability, split-half reliability, and testā€“retest reliability of the scale was 0.85, 0.92, and 0.84, respectively.ConclusionThe Chinese Ped-PDCDS has adequate psychometric properties, and is an easy and appropriate instrument for measuring parentsā€™ difficulty in coping with premature infants
