    Benefits of host genetic diversity for resistance to infection depend on parasite diversity

    Host populations with high genetic diversity are predicted to have lower levels of infection prevalence. This theory assumes that host genetic diversity results in variation in susceptibility and that parasites exhibit variation in infectivity. Empirical studies on the effects of host heterogeneity typically neglect the role of parasite diversity. We conducted three laboratory experiments designed to test if genetic variation in Daphnia magna populations and genetic variation in its parasites together influence the course of parasite spread after introduction. We found that a natural D. magna population exhibited variation in susceptibility to infection by three parasite species and had strong host clone–parasite species interactions. There was no effect of host heterogeneity in experimental host populations (polycultures and monocultures) separately exposed to single strains of three parasite species. When we manipulated the genetic diversity of a single parasite species and exposed them to host monocultures and polycultures, we found that parasite prevalence increased with the number of parasite strains. Host monocultures exposed to several parasite strains had higher mean parasite prevalence and higher variance than polycultures. These results indicate that effect of host genetic diversity on the spread of infection depends on the level of genetic diversity in the parasite population

    Effects of preservation method on canine (Canis lupus familiaris) fecal microbiota.

    Studies involving gut microbiome analysis play an increasing role in the evaluation of health and disease in humans and animals alike. Fecal sampling methods for DNA preservation in laboratory, clinical, and field settings can greatly influence inferences of microbial composition and diversity, but are often inconsistent and under-investigated between studies. Many laboratories have utilized either temperature control or preservation buffers for optimization of DNA preservation, but few studies have evaluated the effects of combining both methods to preserve fecal microbiota. To determine the optimal method for fecal DNA preservation, we collected fecal samples from one canine donor and stored aliquots in RNAlater, 70% ethanol, 50:50 glycerol:PBS, or without buffer at 25 °C, 4 °C, and -80 °C. Fecal DNA was extracted, quantified, and 16S rRNA gene analysis performed on Days 0, 7, 14, and 56 to evaluate changes in DNA concentration, purity, and bacterial diversity and composition over time. We detected overall effects on bacterial community of storage buffer (F-value = 6.87, DF = 3, P < 0.001), storage temperature (F-value=1.77, DF = 3, P = 0.037), and duration of sample storage (F-value = 3.68, DF = 3, P < 0.001). Changes in bacterial composition were observed in samples stored in -80 °C without buffer, a commonly used method for fecal DNA storage, suggesting that simply freezing samples may be suboptimal for bacterial analysis. Fecal preservation with 70% ethanol and RNAlater closely resembled that of fresh samples, though RNAlater yielded significantly lower DNA concentrations (DF = 8.57, P < 0.001). Although bacterial composition varied with temperature and buffer storage, 70% ethanol was the best method for preserving bacterial DNA in canine feces, yielding the highest DNA concentration and minimal changes in bacterial diversity and composition. The differences observed between samples highlight the need to consider optimized post-collection methods in microbiome research

    Reproductive Differences among Delmarva Grass Shrimp (Palaemonetes pugio and P. vulgaris) Populations

    Populations of female grass shrimps (Palaemonetes pugio and P. vulgaris) were sampled from five coastal embayments in Delaware, Maryland, and Virginia (Delmarva) and compared with respect to reproductive and life history attributes. We observed interspecific differences in timing of reproduction, carapace length, ratio of carapace length to total body length, body weight, clutch weight, clutch size, and egg volume. Onset of reproduction in P. vulgaris lagged behind P. pugio. Although there was no difference in the relationship between clutch size and carapace length for the two species, carapace length/total body length in P. pugio was greater than that in P. vulgaris. A multivariate analysis of variance indicated significant differences in carapace length, clutch weight, body weight, clutch size, and egg volume attributable to effects of species, population, and interactions between them. At all sites, P. pugio produced larger eggs than P. vulgaris. Although the two species did not differ in reproductive effort, both species exhibited increases in reproductive effort with latitude. Clutch size also tended to increase with latitude for both species. In populations where both species were abundant, adult females of P. pugio were longer and heavier and produced heavier egg masses comprised of fewer, larger eggs

    Protocol for serious fall injury adjudication in the Strategies to Reduce Injuries and Develop Confidence in Elders (STRIDE) study

    Background: This paper describes a protocol for determining the incidence of serious fall injuries for Strategies to Reduce Injuries and Develop Confidence in Elders (STRIDE), a large, multicenter pragmatic clinical trial with limited resources for event adjudication. We describe how administrative data (from participating health systems and Medicare claims) can be used to confirm participant-reported events, with more time- and resource-intensive full-text medical record data used only on an as-needed basis. Methods: STRIDE is a pragmatic cluster-randomized controlled trial involving 5451 participants age \u3e /= 70 and at increased risk for falls, served by 86 primary care practices in 10 US health systems. The STRIDE intervention involves a nurse falls care manager who assesses a participant\u27s underlying risks for falls, suggests interventions using motivational interviewing, and then creates, implements and longitudinally follows up on an individualized care plan with the participant (and caregiver when appropriate), in partnership with the participant\u27s primary care provider. STRIDE\u27s primary outcome is serious fall injuries, defined as a fall resulting in: (1) medical attention billable according to Medicare guidelines with a) fracture (excluding isolated thoracic vertebral and/or lumbar vertebral fracture), b) joint dislocation, or c) cut requiring closure; OR (2) overnight hospitalization with a) head injury, b) sprain or strain, c) bruising or swelling, or d) other injury determined to be serious (i.e., burn, rhabdomyolysis, or internal injury). Two sources of data are required to confirm a serious fall injury. The primary data source is the participant\u27s self-report of a fall leading to medical attention, identified during telephone interview every 4 months, with the confirmatory source being (1) administrative data capturing encounters at the participating health systems or Medicare claims and/or (2) the full text of medical records requested only as needed. Discussion: Adjudication is ongoing, with over 1000 potentially qualifying events adjudicated to date. Administrative data can be successfully used for adjudication, as part of a hybrid approach that retrieves full-text medical records only when needed. With the continued refinement and availability of administrative data sources, future studies may be able to use administrative data completely in lieu of medical record review to maximize the quality of adjudication with finite resources.     In social animals, scent secretions and marking behaviors play critical roles in communication, including intraspecific signals, such as identifying individuals and group membership, as well as interspecific signaling. Anal sacs are an important odor producing organ found across the carnivorans (species in the mammalian Order Carnivora). Secretions from the anal sac may be used as chemical signals by animals for behaviors ranging from defense to species recognition to signaling reproductive status. In addition, a recent study suggests that domestic cats utilize short-chain free fatty acids in anal sac secretions for individual recognition. The fermentation hypothesis is the idea that symbiotic microorganisms living in association with animals contribute to odor profiles used in chemical communication and that variation in these chemical signals reflects variation in the microbial community. Here we examine the fermentation hypothesis by characterizing volatile organic compounds (VOC) and bacteria isolated from anal sac secretions collected from a male Bengal cat (Felis catus × Prionailurus bengalensis), a cross between the domestic cat and the leopard cat. Both left and right anal sacs of a male Bengal cat were manually expressed (emptied) and collected. Half of the material was used to culture bacteria or to extract bacterial DNA and the other half was used for VOC analysis. DNA was extracted from the anal sac secretions and used for a 16S rRNA gene PCR amplification and sequencing based characterization of the microbial community. Additionally, some of the material was plated out in order to isolate bacterial colonies. Three taxa (Bacteroides fragilis, Tessaracoccus, and Finegoldia magna) were relatively abundant in the 16S rRNA gene sequence data and also isolated by culturing. Using Solid Phase Microextraction (SPME) gas chromatography-mass spectrometry (GC-MS), we tentatively identified 52 compounds from the Bengal cat anal sac secretions and 67 compounds from cultures of the three bacterial isolates chosen for further analysis. Among 67 compounds tentatively identified from bacterial isolates, 51 were also found in the anal sac secretion. We show that the bacterial community in the anal sac consists primarily of only a few abundant taxa and that isolates of these taxa produce numerous volatiles that are found in the combined anal sac volatile profile. Several of these volatiles are found in anal sac secretions from other carnivorans, and are also associated with known bacterial biosynthesis pathways. This is consistent with the fermentation hypothesis and the possibility that the anal sac is maintained at least in part to house bacteria that produce volatiles for the host

    936-86 Plasma Fibrinogen Level Predicts Severity of Intimal Thickening After Cardiac Transplantation

    Diffuse atherosclerosis is the primary reason for late graft failure after cardiac transplantation. Because there is increasing evidence that imbalances in the hemostatic and fibrinolytic pathways are associated with allogeneic rejection, we hypothesized that atherothrombotic risk factors may contribute to accelerated atherosclerosis. We therefore prospectively evaluated the burden of coronary atherosclerosis by intravascular ultrasound (IVUS) in 20 patients and measured plasma fibrinogen (FGN). lipoprotein (a) (Lp(a)) and net fibrinolytic activity of plasma using a standard fibrin plate assay. Intimal thickening was quantified using IVUS by measuring the intimal index (li=intimal area/[intimal area+luminal area]) in 2–5 segments of the LAD using planimetry. The maximal Ii per patient was calculated and indexed to the time post-transplant (Mxli/Yr). FGN predicted severity of Mxli/Yr (r2=0.41, p=0.008). In patients with decreased plasma fibrinolytic activity (lytic zone <100 mm2). Mxli/Yr was increased ten-fold (0.21±0.17 vs. 0.02±0.02, p=0.002). Because Lp(a) colocalizes with fibrinogen in the vessel wall and inhibits fibrinolysis, we correlated plasma Lp(a) levels with the degree of intimal thickening. Lp(a) did not predict Mxli/Yr (p=NS). In conclusion, these data suggest that plasma FGN and net fibrinolytic activity predict the degree of intimal thickening and that fibrin deposition may play an integral role in diffuse coronary atherosclerosis after cardiac transplantation
