Hydrogels of enzymatically modified dextrin
Dextrin is a glucose-containing saccharide polymer linked by α-(1→4) D-glucose units and is produced by partial
hydrolysis of starch. Hydrolysis can be accomplished by the use of acid, enzymes, or by a combination of both. In
this work, the Bacillus subtilis protease Proleather FG-F catalyzed the transesterification of the soluble
polysaccharide with vinyl acrylate (VA), using anhydrous dimethylsulfoxide (DMSO) as reaction medium.
Different degrees of substitution (DS, defined as the amount of acrylate groups per 100 glucopyranose residues)
ranging from 11 to 19 % were achieved by controlling the molar ratio of VA to dextrin. Dextrin derivatized with
VA (dextrin-VA) was characterized by gel permeation chromatography. Gels were obtained by free radical
polymerization of aqueous solutions of dextrin-VA with different degrees of substitution and monomer
concentration. Swelling experiments have been performed in order to determine the gel pore sizes. Specific
physical properties, as degradation behaviour were also evaluated. These hydrogels are being developed as scaffold
materials for bioactive molecule and cell delivery, tissue engineering and a variety of other biomedical applications

A criação da Política Europeia de Vizinhança (PEV) em 2003 visou promover segurança
e prosperidade nas novas fronteiras da União Europeia (UE) após o Alargamento de 2004.
Dado o sucesso desta ronda de Alargamento em reformar a periferia da UE, a PEV foi
concebida para replicar o poder transformador da Política de Alargamento, centrado na
exportação de normas da UE através de incentivos. De facto, o acervo comunitário da UE
tem consagrado progressivamente uma obrigação legal e um compromisso político de
promoção das suas normas na ação externa. Esta assunção tem alimentado o debate
académico sobre a conceptualização da UE enquanto potência normativa. Com efeito,
esta dissertação propõe um estudo sobre o poder normativo da UE no quadro da PEV.
Expõe, portanto, uma análise sobre a ambição da UE em promover as suas normas nos
vizinhos de Este e Sul como meio para alcançar uma nova ordenação das relações com a
periferia, que permita uma maior integração política e económica, bem como a
estabilidade regional. Concretamente, a investigação procurou, primeiro, conceptualizar
a PEV como uma política de expressão do poder normativo da UE. Segundo, analisou a
evolução da expressão do poder normativo da UE na implementação da PEV (2003-2022)
ao longo do tempo e entre as duas regiões da PEV. Terceiro, procurou concluir se o poder
normativo da UE contribuiu para fomentar a integração política e económica com os
vizinhos, bem como a estabilidade regional. Os resultados da investigação permitiram
confirmar que a PEV constitui um quadro político que visa exportar as normas da UE.
Todavia, verificou-se que o exercício do poder normativo da UE teve um sucesso
marginal para fomentar a integração política e económica, visto que a adoção de normas
da UE permaneceu limitada e circunscrita a um número reduzido de vizinhos. Além disso,
a PEV não contribuiu para reforçar a estabilidade regional na periferia da UE – pelo
contrário, há um agravamento da instabilidade após a invasão russa da Ucrânia em 2022.
Dada esta avaliação, a investigação identificou três razões que explicam o insucesso da
expressão do poder normativo da UE no quadro da PEV. Primeiro, a PEV constitui uma
réplica excessiva do modelo institucional e instrumentos da Política de Alargamento.
Segundo, a implementação da PEV não permitiu definir uma abordagem consistente para
a exportação das normas da UE. Terceiro, a agenda da UE enfrenta crescente competição
por parte de potências globais e regionais na vizinhança.The creation of the European Neighbourhood Policy (ENP) in 2003 aimed to foster
security and prosperity in the soon-to-be new borders of the European Union (EU)
following the 2004 Enlargement. Given the success of this Enlargement round to
(re)shape EU’s periphery, the ENP was envisaged to emulate the transformative power
of the Enlargement Policy, centred on the export of EU norms attached to incentives. In
fact, the acquis communautaire of the EU has progressively enshrined a legal obligation
and political commitment to promote EU norms in its external action, leading to a vivid
academic debate on the conceptualisation of the EU as a normative power. Accordingly,
this dissertation proposes a study on the normative power of the EU in the framework of
the ENP. It offers an analysis on EU’s ambition to export its norms to Eastern and
Southern neighbours as a means to attain a new ordering of relations with its periphery
that allows for stronger political and economic integration as well as regional stability. In
specific, this research sought, first, to conceptualise the ENP as a policy framework for
the EU to exert its normative power. Second, it analysed the evolution of the exertion of
EU’s normative power throughout the implementation of the ENP (2003-2022) over time
and between the two ENP regions. Third, it aimed to assert whether EU’s normative
power has contributed to foster political and economic integration with neighbours as
well as regional stability. The research findings confirmed that the ENP constitutes a
policy framework which seeks to export EU norms. However, it was found that the
exertion of EU’s normative power has had a marginal success to foster political and
economic integration, considering that the adoption of EU norms has been minimal and
circumscribed to a handful of partners. Besides, the ENP has failed to enhance regional
stability in EU’s periphery – on the contrary, there is an aggravation of instability
following the 2022 Russian invasion of Ukraine. Given this assessment, this research
identified three reasons that account for the unsuccess of EU’s exertion of its normative
power in the framework of the ENP. First, the ENP constitutes an overstretching of the
institutional model and policy instruments of the Enlargement Policy. Second, ENP’s
implementation has failed to produce a consistent approach for exporting EU norms.

“Seeing a country with your own eyes”: The impact of immersion experiences in language travel
This study approaches immersion experiences in the context of language tourism. Immersion is here understood as tourists’ extensive engagement and interaction with the members of the host culture. Previous studies have pointed out that language tourism implies a deeper contact with locals; however, other studies have observed that many language programmes abroad are not always as immersive as expected. These contradictory findings suggest that there may be a tremendous variation in contact with locals in the context of language tourism.
Hence, the present mixed methods study uses interviews with 22 language travellers, and 1,011 survey answers to address the question of how language tourists who had an intense contact with local residents differ – in terms of profile and travel experiences – from those language travellers who did not have such an intense contact. There are significant differences between these groups. Those who had a more intense contact with locals are more likely to be intrinsically motivated by an interest for the local culture, to travel alone, to report higher levels of fluency, more frequent accommodation at homestays, and greater engagement in a wider range of activities at the destination.info:eu-repo/semantics/publishedVersio
Preservação consciente da Amazônia: transversalidade entre ciências naturais e meio ambiente na educação infantil
Projeto de investigaçãoNa atual realidade de um mundo cada vez mais globalizado, a formação de um cidadão consciente crítico, participativo, e com idéias novas para mudar o caos em que nos metemos, deve acontece desde os primeiros anos de vida. Tal formação se insere presentemente numa sociedade em que o conhecimento científico e a intervenção da tecnologia no dia-a-dia das pessoas são cada vez mais valorizados. Esse cidadão precisa compreender o mundo e suas transformações, situando-se como um indivíduo integrante desse fenômeno de globalização, e proativos na construção de uma sociedade mais solidária, fraterna, igualitária, justa e libertária.
Como parte inicial da construção de uma futura proposta, que minimize essa real desagregação socialambiental em que vivemos, tem este trabalho como meta reelaborar e renovar o conteúdo programático entre os temas transversais das ciências naturais e do meio ambiente. Num segundo momento, deve ser desenvolvido um novo método de didático pedagógico, com o apóio dos conhecimentos da Neurociência e da Psicologia, para trabalhar a consciência das crianças de até cinco anos de idade, no sentido de mudar o modelo educacional atual, quase ou totalmente falido.
A Amazônia brasileira e a região norte de Portugal são os cenários perfeitos para se realizar essa experiência, de um novo modelo educacional aqui proposto. O qual irá trabalhar o conteúdo programático das ciências naturais e do meio ambiente, em prol da preservação da natureza, e o desenvolvimento da consciência humana, visando uma harmonia socioambiental com qualidade de vida.
Os espaços educacionais escolhidos para realização dessa experiência serão algumas escolas públicas da rede de ensino fundamental do estado do Pará, Brasil, mais precisamente no município de Castanhal, e outras da rede de ensino da região do Minho, Portugal.Fundação para a Ciência e a Tecnologia (FCT) - unidade de investigação 31
The effect of an RGD-Human Chitin-Binding Domain fusion protein on the adhesion of fibroblasts to reacetylated chitosan films
Biomaterials used for tissue engineering applications must provide a structural support
for the tissue development and also actively interact with cells, promoting adhesion,
proliferation and differentiation. To achieve this goal, adhesion molecules may be used,
such as the tripeptide Arg-Gly-Asp (RGD). RGD was found to be the major functional
amino acid sequence responsible for cellular adhesion. This sequence can be used to elicit
specific cellular responses and it has been extensively demonstrated that RGD sequence
improves cell adhesion, spreading and proliferation in different materials. Chitosan and
chitin represent a family of biopolymers, made up of b(14)-linked N-acetyl-Dglucosamine
and D-glucosamine subunits. Due to their biodegradability and
biocompatibility, chitin and chitosan, are widely studied for biomedical applications.
A method based on the use of a human Carbohydrate-Binding Module, with affinity for
chitin, was tested as an alternative approach to the chemical grafting of bioactive
peptides. This approach would simultaneously allow the production of recombinant
peptides (alternatively to peptide synthesis) and provide a simple way for the specific and
strong adsorption of the peptides to the biomaterial. A fusion recombinant protein,
containing the RGD sequence fused to a human chitin-binding module (ChBM), was
expressed in E. coli. The adhesion of fibroblasts to reacetylated chitosan (RC) films was
the model system selected to analyse the properties of the obtained proteins. Thus, the
evaluation of cell attachment and proliferation on polystyrene surfaces and reacetylated
chitosan films, coated with the recombinant proteins, was performed using mouse
embryo fibroblasts 3T3. The results show that the recombinant proteins affect negatively
fibroblasts anchorage to the materials surface, inhibiting its adhesion and proliferation.
We also conclude that this negative effect is fundamentally due to the human chitinbinding
domain
Ensino fundamental brasileiro : manual escolar e formação do professor
INTRODUÇÃO: Na alargada concepção de ensino e de aprendizagem da formação de professores brasileiros prevalece o modelo dominantemente transmissivo, esquecendo a possibilidade de intervensão dos educandos na solução dos problemas estudados, ou seja, privilegia a emissão do saber através do conteúdo formatado e limitado no Currículo Nacional e no Livro Didático, o que é pouco apropriado dentro do atual contexto social. A análise crítica, reflexiva e construtiva do conteúdo da unidade Ciência, do Livro Didático brasileiro, é um caminho viável de aperfeiçoamento dessa ultrapassada concepção de docência, concebida na Graduação do Ensino Superior no Brasil.
MÉTODOS: Foram selecionados e comparadas duas coleções de Livros Didáticos adotadas para o nível do Ensino Fundamental brasileiro, referentes ao 1º, 2º, 3º e 4º Ano escolar. Estas obras pedagógicas fazem parte das editoras com melhor aceitação no Estado do Pará. Foi elaborada uma grelha de análise de conteúdo, baseada na construída no âmbito do projeto Europeu FP6 BIOHEAD-CITIZEN (Carvalho & Clément, 2007).
RESULTADOS: O Livro Didático LDa (Passos e Passos, 2009) segue uma linha de ensino-aprendizagem tradicional, valorizando o campo afetivo, emocional, estético e lúdico. Embora, também enfatize, denuncie e critique a degradação da zona Rural, através da atividade agropecuária e da exploração indiscriminada da sua floresta. Além disso, mostram a real e grave situação encontrada no Meio Urbano, assumindo uma postura crítica e de cobrança na solução dos graves problemas sociais vividos, hoje, no Brasil. A obra equivalente LDb (Trivellato e Lico, 2010) segue uma linha de abordagem mais realista, apoiada em dados científicos, enfatizando com imagem fotográfica uma mensagem da vida e dos problemas ambientais que acontecem no país, em especial na Amazônia. Em ambas as coleções dos Livros Didáticos percebe-se que o conteúdo induz a um método de ensino e aprendizagem voltado para a simples transmissão do conhecimento, sem considerar a ótica de educar para proceder com uma intervenção de maneira construtiva, na sociedade e no seu Meio, objetivando minimizar os graves problemas que afetam a Natureza local, regional e planetária.
CONCLUSÕES: Considerando a necessidade urgente de se preservar a Natureza do planeta Terra, em especial da região Amazônica, pode-se afirmar que os Livros Didáticos analisados cumprem com a sua competência de educar o aluno a conhecer e refletir sobre o seu habitat. Porém, não ensinam como proceder e intervir na solução dos problemas ambientais e sociais que acontecem nesse Meio, o que é particularmente crítico no Estado do Pará e da região Amazônica. Este estudo mostra Será portanto necessário promover uma formação de professores que não só (i) tenha em atenção a atualização e a promoção de práticas educativas participativas e intervenientes, que também complementem os livros didáticos em vigor nas escolas, (ii) estimulando uma intervenção pró-ativa do aluno na solução dos problemas estudados e, ainda, (iii) que os professores adquiram competências para participar e contribuir na seleção dessas obras pedagógicas, aplicadas nas suas respetivas escolas, quiçá também contribuindo para a concepção e formulação das mesmas.CIEC - Centro de Investigação em Estudos da Criança, UM (UI 317 da FCT
A Model to Improve the Accuracy of WSN Simulations
Simulation studies have been extensively adopted in the networking research community. Nevertheless, the performance of the software components running within the network devices is often not modeled by generic network simulators. This aspect is particularly important in wireless sensor networks (WSN). As motes present very limited computing resources, the overhead of the software components cannot be ignored. Consequently, WSN simulation results may diverge significantly from the reality. After showing experimentally the validity of this assumption, the paper proposes a set of generic equations to model the performance of WSN software components. Validation tests using contention and multiplexing-based MAC protocols show that the inclusion of the proposed model in a WSN simulator improves the confidence degree in the simulation results significantly
Planet earth as a living being in mutation : which contributions present the portuguese school manuals of Elementary School?
No momento em que a Natureza do Planeta Terra é um dos temas mais debatido em quase todos fóruns de pesquisa e educação, tornou-se pertinente proceder à análise dos conteúdos desta temática nos manuais escolares portugueses, do 1º Ciclo do Ensino Básico (1ºCEB). Foi construida e analisada uma grelha baseada na de Ecologia e Educação Ambiental do Projeto Europeu FP6 BIOHEAD-CITIZEN. Foram selecionados três manuais por cada ano escolar do 1º CEB, tendo sido escolhidos de entre as editoras com maior difusão no distrito de Braga. Em todos os manuais consultados tornou-se evidente a valorização dos saberes de vida previamente adquirido pelo aluno, antes de iniciar a sua vida escolar. O meio comum é principalmente o Rural, com identificação e classificação das espécies animais e vegetais. Valoriza-se o cuidar dessas espécies, como por exemplo: saber criar e tratar os animais domésticos, ter conhecimento básico sobre a Sementeira, o Plantio, o Cultivo e a Colheita de hortaliças e frutas. As imagens são em grande parte figurativas, com uma mensagem puramente estética. Torna-se necessário incluir exemplos reais de espécies animais e vegetais ameaçadas de extinção, devido à caça excessiva, ao cultivo intensivo, ou à destruição pelo uso insensato do Fogo. Também seria interessante ensinar sobre os elementos vitais da vida (Ar, Água e Fogo), destacando os seus benefícios e malefícios. Verificou-se a necessidade de se propor determinadas recomendações, com vista à sua melhoria, na perspetiva de proporcionar à criança conhecer e desenvolver competências no sentido de preservar e recompor o seu habitat, contribuindo também para o meio ambiente global, ensinado como um Ser vivo em mutação.Nowadays that the Nature of the Planet Earth is one of the most debated subjects in almost all Forums of research and education, it is pertinent to examine the contents of this theme in Portuguese textbooks of the 1st Cycle of Basic Education (1º CBE). A grid was constructed and analyzed based on a grid of Ecology and Environmental Education of the European Project FP6 BIOHEAD-CITIZEN. We selected three textbooks for each school year of the 1st CBE, having been chosen among the publishers with more diffusion in the district of Braga. In all consulted textbooks, it became evident the emphasis on the pupils’ previous knowledge before starting their school life. The Rural environment is the most common, presenting identification and classification of animal and plant species. The care for the species is valorised, as for example: to know how to grow and handle domestic animals, to have basic knowledge about seeding, planting, growing and harvesting fruits and vegetables. The images are mainly figurative, with a message purely aesthetic. It becomes necessary to include actual examples of animal and plant species threatened with extinction due to overhunting, to intensive cultivation or the senseless destruction by fire. It would also be interesting to teach about the vital elements of life (Air, Water and Fire) highlighting its benefits and harms. Transcriptomic screen for DIS3 and DIS3L1 exosome subunits associated functional networks in colorectal cancer
The final step of cytoplasmic mRNA degradation proceeds in either a 5’-3’ direction,
catalyzed by XRN1, or in a 3’-5’ direction catalyzed by the exosome. In yeast, DIS3/Rrp44
protein is the catalytic subunit of the exosome. In humans, there are three known paralogues of
this enzyme: DIS3, DIS3L1, and DIS3L2. Important findings over the last years have shed a new
light onto the.mechanistic details of RNA degradation by these exoribonucleases. In addition, it
has been shown that they are involved in growth, mitotic control and important human diseases,
including cancer. For example, DIS3L2 inactivation was associated with mitotic abnormalities
and altered expression of mitotic checkpoint proteins (Astuti et al., 2012). In another study, DIS3
was found to be highly expressed in colorectal cancer (CRC), suggesting an oncogenic function
(Camps et al., 2013).
A major challenge in systems biology is to reveal the cellular networks that give rise to
specific phenotypes (Lan et al., 2013). In this project, we aim to analyze how DIS3 and DIS3L1
regulate the human transcriptome, and how their functional interactions modulate the
transcriptional reprogramming of colorectal cancer cells. We will perform an extensive
characterization of the DIS3 and DIS3L1 mRNA targets, using DIS3 and DIS3L1 knockdown and
microarray analysis, in normal colorectal cells, and in different CRC cell lines, in the presence
and absence of stress stimuli, such as hypoxia.
Differential expression and gene set enrichment analyses of collected data will elucidate
new cellular pathways regulated by DIS3 and DIS3L1 and/or by their targets, as well as how
they can be involved in CRC. In addition, this analysis may reveal novel functional networks
through which the RNA exosome modulates the eukaryotic transcriptome
