    Implantation of subcutaneous heart rate data loggers in southern elephant seals (Mirounga leonina)

    Unlike most phocid species (Phocidae), Mirounga leonina (southern elephant seals) experience a catastrophic moult where they not only replace their hair but also their epidermis when ashore for approximately 1 month. Few studies have investigated behavioural and physiological adaptations of southern elephant seals during the moult fast, a particularly energetically costly life cycle’s phase. Recording heart rate is a reliable technique for estimating energy expenditure in the field. For the first time, subcutaneous heart rate data loggers were successfully implanted during the moult in two free-ranging southern elephant seals over 3–6 days. No substantial postoperative complications were encountered and consistent heart rate data were obtained. This promising surgical technique opens new opportunities for monitoring heart rate in phocid seals

    Pyrotechnic Actuator for Retracting Tubes Between MSL Subsystems

    An apparatus, denoted the "retractuator" (a contraction of "retracting actuator"), was designed to help ensure clean separation between the cruise stage and the entry-vehicle subsystem of the Mars Science Laboratory (MSL) mission. The retractuator or an equivalent mechanism is needed because of tubes that (1) transport a heat-transfer fluid between the stages during flight and (2) are cut immediately prior to separation of the stages retractuator. The role of the retractuator is to retract the tubes, after they are cut and before separation of the subsystem, so that cut ends of the tubes do not damage thermal-protection coats on the entry vehicle and do not contribute to uncertainty of drag and consequent uncertainty in separation velocity

    Lexicality and frequency in specific language impairment: accuracy and error data from two nonword repetition tests

    Purpose: Deficits in phonological working memory and deficits in phonological processing have both been considered potential explanatory factors in Specific Language Impairment (SLI). Manipulations of the lexicality and phonotactic frequency of nonwords enable contrasting predictions to be derived from these hypotheses. Method: 18 typically developing (TD) children and 18 children with SLI completed an assessment battery that included tests of language ability, non-verbal intelligence, and two nonword repetition tests that varied in lexicality and frequency. Results: Repetition accuracy showed that children with SLI were unimpaired for short and simple high lexicality nonwords, whereas clear impairments were shown for all low lexicality nonwords. For low lexicality nonwords, greater repetition accuracy was seen for nonwords constructed from high over low frequency phoneme sequences. Children with SLI made the same proportion of errors that substituted a nonsense syllable for a lexical item as TD children, and this was stable across nonword length. Conclusions: The data show support for a phonological processing deficit in children with SLI, where long-term lexical and sub-lexical phonological knowledge mediate the interpretation of nonwords. However, the data also suggest that while phonological processing may provide a key explanation of SLI, a full account is likely to be multi-faceted

    Caracterização e ocorrência do distúrbio do amolecimento precoce em mamões 'Golden'

    The occurrence of green skin and soft pulp in 'Golden' papaya fruit during certain seasons has been reported by farmers in the northern of the state of Espirito Santo, Brazil. The objective of this study was to characterize and determine the occurrence of this disorder, which was referred as "early softening disorder". Fruits were harvested weekly for 11 months (from September to July). The fruits were stored at 10°C, and then fruit flesh firmness and skin color were analyzed. The results of the firmness test were submitted to regression analysis assuming a linear trendline. The slope of the curve was called the 'softening index' (SI). Fruits with early softening are characterized by a loss of firmness in less than 10 days, even when stored under refrigeration. Although softened, the skin of the fruit remains partially green. Fruits with the disorder occurred more frequently from mid-summer to mid-autumn (February to May). It is not possible to distinguish early softening disorder fruits from those without the disorder by skin color and flesh firmness analysis at the time of the harvest.Tem sido relatado por produtores da região norte do Espírito Santo a ocorrência de mamões 'Golden' com casca verde e polpa mole, em determinadas épocas do ano. O objetivo deste trabalho foi caracterizar e determinar a ocorrência deste distúrbio denominado de amolecimento precoce. Foram realizadas coletas semanais durante 11 meses (período de setembro a julho). Os frutos foram armazenados a 10°C e analisados quanto à firmeza da polpa e à cor da casca. Os resultados de firmeza da polpa foram submetidos à análise de regressão, assumindo-se que a equação é do tipo linear, e o ângulo de inclinação da curva foi chamado Índice de Amolecimento (IA). Frutos com o distúrbio caracterizaram-se pela perda da firmeza em menos de 10 dias, mesmo quando armazenados sob refrigeração. Embora amolecidos, a coloração da casca manteve-se parcialmente verde. A maior frequência de frutos com o distúrbio ocorreu de meados de verão a meados de outono (fevereiro a maio). Não é possível distinguir frutos com o distúrbio do amolecimento precoce daqueles normais pela análise da cor da casca e da firmeza da polpa, no momento da colheita

    GATA3 and MDM2 are synthetic lethal in estrogen receptor-positive breast cancers.

    Synthetic lethal interactions, where the simultaneous but not individual inactivation of two genes is lethal to the cell, have been successfully exploited to treat cancer. GATA3 is frequently mutated in estrogen receptor (ER)-positive breast cancers and its deficiency defines a subset of patients with poor response to hormonal therapy and poor prognosis. However, GATA3 is not yet targetable. Here we show that GATA3 and MDM2 are synthetically lethal in ER-positive breast cancer. Depletion and pharmacological inhibition of MDM2 significantly impaired tumor growth in GATA3-deficient models in vitro, in vivo and in patient-derived organoids/xenograft (PDOs/PDX) harboring GATA3 somatic mutations. The synthetic lethality requires p53 and acts via the PI3K/Akt/mTOR pathway. Our results present MDM2 as a therapeutic target in the substantial cohort of ER-positive, GATA3-mutant breast cancer patients. With MDM2 inhibitors widely available, our findings can be rapidly translated into clinical trials to evaluate in-patient efficacy

    The relationship between seminal leukocytes, oxidative status in the ejaculate, and apoptotic markers in human spermatozoa

    The aim of this study was to investigate the relationship between seminal leukocytes, reactive oxygen species (ROS) production in the ejaculate, and markers of apoptosis in human spermatozoa. Semen samples were collected from 60 patients attending fertility clinics at the Reproductive Biology Unit at Tygerberg Academic Hospital and Vincent Pallotti Hospital, Cape Town, South Africa. The concentration of seminal leukocytes was determined and was correlated with ROS production in the ejaculate, the percentage of superoxide (·O2 )- and hydrogen peroxide (H2O2)-positive spermatozoa, glutathione activation in the ejaculate, and with markers of apoptosis in spermatozoa, namely cysteine-dependent aspartate-directed proteases (caspase)-3/7 activation, mitochondrial membrane potential (ΔΨm), and the percentage of terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL)-positive sperm. Significant correlations with the concentration of seminal leukocytes were found for ROS production in the ejaculate, the percentage of ·O2 -positive spermatozoa, and caspase-3/7 activation in the ejaculate. Leukocytospermic samples showed significantly higher ROS production, percentage of ·O2 -positive sperm, GSH activation, and caspase-3/7 activation compared to non-leukocytospermic samples. The percentage of ·O2 -positive sperm was significantly correlated with sperm ΔΨm and caspase-3/7 activation in the ejaculate. Sperm ΔΨm and TUNEL-positive sperm did not correlate with seminal leukocyte concentration. Data demonstrate that high seminal leukocyte concentrations that leads to increased seminal ROS production, and is also associated with caspase activation in the male germ cell and increased mitochondrial ROS production. The latter could possibly be a result of disturbed ΔΨm. The activation of caspase-3/7 could then follow the increased intrinsic superoxide levels due to depleted intrinsic glutathione (GSH). These cellular events might not directly and immediately lead to DNA fragmentation as an endpoint of apoptosis because of topological hindrances.Web of Scienc
