    Strain stiffening of peripheral nerves subjected to longitudinal extensions in vitro

    The mechanical response of peripheral nerves is crucial to understand their physiological and pathological conditions. However, their response to external mechanical solicitations is still partially unclear, since peripheral nerves could behave in a quite complex way. In particular, nerves react to longitudinal strains increasing their stiffness to keep axons integrity and to preserve endoneural structures from overstretch. In this work, the strain stiffening of peripheral nerves was investigated in vitro through a recently introduced computational framework, which is able to theoretically reproduce the experimental behaviour of excised tibial and sciatic nerves. The evolution and the variation of the tangent modulus of tibial and sciatic nerve specimens were quantitatively investigated and compared to explore how stretched peripheral nerves change their instantaneous stiffness

    INVOLVEMENT OF BASIC FIBROBLAST GROWTH-FACTOR IN SURAMIN-INDUCED INHIBITION OF V79/AP4 FIBROBLAST CELL-PROLIFERATION

    The V79/AP4 Chinese hamster fibroblasts were densely stained with the anti-basic fibroblast growth factor (bFGF) antibody demonstrating an endogenous production of the peptide. The in vitro proliferation of these cells was stimulated by exogenous bFGF and the maximum growth (259% increase in H-3-thymidine incorporation into DNA) was reached with bFGF 10 ng ml-1. Inhibition of bFGF-mediated mitogenic pathway was obtained with a 15-mer antisense oligodeoxynucleotide targeted against bFGF mRNA and with suramin, a drug which blocks the biological activity of heparin-binding growth factors. bFGF antisense oligomer reduced the synthesis of DNA by 79.5 and 89.5% at 20 and 60 muM, respectively; this effect was reversed by the addition of exogenous bFGF to the culture medium. A short-term exposure to suramin 300 mug ml-1 produced a modest reduction in H-3-thymidine incorporation but suppressed the mitogenic effect of bFGF on V79/AP4 cells. In cells treated with suramin 300 mug ml-1 the drug concentration increased linearly over 3 days, reaching 13.15 mug mg-1 of protein; cell proliferation was inhibited in a dose-related manner as evaluated by the colony formation assay (IC50: 344.22 mug ml-1) and by the number of mitoses observed in culture. Furthermore, the drug induced ultrastructural alterations, consisting of perinuclear cisternae swelling, chromatin condensation, nucleolar segregation and cytoplasmic vacuolations. These findings demonstrated that the endogenous production of bFGF plays an important role in V79/AP4 fibroblasts proliferation, and the inhibition of bFGF-mediated mitogenic signalling with bFGF antisense oligomer or suramin is an effective mean of reducing cell growth

    Two-colour generation in a chirped seeded Free-Electron Laser

    We present the experimental demonstration of a method for generating two spectrally and temporally separated pulses by an externally seeded, single-pass free-electron laser operating in the extreme-ultraviolet spectral range. Our results, collected on the FERMI@Elettra facility and confirmed by numerical simulations, demonstrate the possibility of controlling both the spectral and temporal features of the generated pulses. A free-electron laser operated in this mode becomes a suitable light source for jitter-free, two-colour pump-probe experiments

    Prevention of incisional hernia post emergency laparotomy: A time to change? A case series.

    Introduction: Complicated acute diverticulitis (CAD) is a surgical challenge in which a mini-invasive approach may be offered. Laparoscopic peritoneal lavage (LPL) was introduced as an alternative to sigmoid resection. However, the role of LPL is still under debate. Aim of this study was to evaluate which surgical strategy between LPL and Laparoscopic Sigmoidectomy (LS) could give better outcomes in CAD. Materials and methods: This prospective, observational, multicenter study lasted from 2015 until to 2018. Inclusion criteria: left colonic or sigmoid CAD (modified Hinchey’’s classification: grade II not responder to conservative treatment and grade III). Exclusion criteria: septic shock, immunodepression, previous multiple surgical operations, modified Hinchey’’s grade I and IV,\15 and[85 years. Comparisons were made between LPL and LS groups. Results: 66 patients were enrolled: 28 (42%) had LPL and 38 (58%) LS. Following sigmoidectomy, 24 pts (63%) had a primary anastomosis and 14 pts (37%) an end-colostomy (Hartmann’’s procedure). There were no significant differences regarding age, male gender rate and mean BMI (p = 0.314, p = 0.07, p = 0.129, respectively). ASA score [2 was significantly higher in LPL (p = 0.05). The number of previous episodes of diverticulitis and the mean C-Reactive Protein dosage were similar (p = 0.756 and 0.846). Mannheim Peritonitis Index was significantly higher in LPL (0.004). No differences were found regarding to the distribution of Hinchey’’s grades II and III (p = 0.727). 1 (4%) patient in LPL and 5 pts (13%) in LS needed a conversion to open surgery (p = 0.181). Overall, the morbidity rates were 33% in LPL and 18% in LS (p = 0.169). Organ space infection (30% vs 3%, p = 0.002) and the re-operation rates (18.5% vs 0; p = 0.006) resulted significantly higher in the LPL group. Mortality was nihil. Mean post-op length of stay was 11.4 days in LPL and 8.23 days in LS (p = 0.088). Diverticular recurrence was significantly increased in LPL (p = 0.003). Conclusions: Compared to LS, LPL is associated with increased ongoing sepsis, emergency re-intervention and recurrence of acute diverticulitis.     Set-optimization meets variational inequalities

    We study necessary and sufficient conditions to attain solutions of set-optimization problems in therms of variational inequalities of Stampacchia and Minty type. The notion of a solution we deal with has been introduced Heyde and Loehne, for convex set-valued objective functions. To define the set-valued variational inequality, we introduce a set-valued directional derivative and we relate it to the Dini derivatives of a family of linearly scalarized problems. The optimality conditions are given by Stampacchia and Minty type Variational inequalities, defined both by the set valued directional derivative and by the Dini derivatives of the scalarizations. The main results allow to obtain known variational characterizations for vector valued optimization problems

    Single-shot transverse coherence in seeded and unseeded free-electron lasers: A comparison

    The advent of x-ray free-electron lasers (FELs) drastically enhanced the capabilities of several analytical techniques, for which the degree of transverse (spatial) coherence of the source is essential. FELs can be operated in self-amplified spontaneous emission (SASE) or seeded configurations, which rely on a qualitatively different initialization of the amplification process leading to light emission. The degree of transverse coherence of SASE and seeded FELs has been characterized in the past, both experimentally and theoretically. However, a direct experimental comparison between the two regimes in similar operating conditions is missing, as well as an accurate study of the sensitivity of transverse coherence to key working parameters. In this paper, we carry out such a comparison, focusing in particular on the evolution of coherence during the light amplification process

    Characterization of soft x-ray echo-enabled harmonic generation free-electron laser pulses in the presence of incoherent electron beam energy modulations

    Echo-enabled harmonic generation free-electron lasers (EEHG FELs) are promising candidates to produce fully coherent soft x-ray pulses by virtue of efficient high-harmonic frequency up-conversion from ultraviolet lasers. The ultimate spectral limit of EEHG, however, remains unclear, because of the broadening and distortions induced in the output spectrum by residual broadband energy modulations in the electron beam. We present a mathematical description of the impact of incoherent (broadband) energy modulations on the bunching spectrum produced by the microbunching instability through both the accelerator and the EEHG line. The model is in agreement with a systematic experimental characterization of the FERMI EEHG FEL in the photon energy range 130\u2013210 eV. We find that amplification of electron beam energy distortions primarily in the EEHG dispersive sections explains an observed reduction of the FEL spectral brightness proportional to the EEHG harmonic number. Local maxima of the FEL spectral brightness and of the spectral stability are found for a suitable balance of the dispersive sections\u2019 strength and the first seed laser pulse energy. Such characterization provides a benchmark for user experiments and future EEHG implementations designed to reach shorter wavelengths
