    How Habitat Change and Rainfall Affect Dung Beetle Diversity in Caatinga, a Brazilian Semi-Arid Ecosystem

    The aim of the present study was to evaluate how dung beetle communities respond to both environment and rainfall in the Caatinga, a semi-arid ecosystem in northeastern Brazil. The communities were sampled monthly from May 2006 to April 2007 using pitfall traps baited with human feces in two environments denominated “land use area” and “undisturbed area.” Abundance and species richness were compared between the two environments and two seasons (dry and wet season) using a generalized linear model with a Poisson error distribution. Diversity was compared between the two environments (land use area and undisturbed area) and seasons (dry and wet) using the Two-Way ANOVA test. Non-metric multidimensional scaling was performed on the resemblance matrix of Bray-Curtis distances (with 1000 random restarts) to determine whether disturbance affected the abundance and species composition of the dung beetle communities. Spearman's correlation coefficient was used to determine whether rainfall was correlated with abundance and species richness. A total of 1097 specimens belonging to 13 species were collected. The most abundant and frequent species was Dichotomius geminatus Arrow (Coleoptera: Scarabaeidae). The environment exerted an influence over abundance. Abundance and diversity were affected by season, with an increase in abundance at the beginning of the wet season. The correlation coefficient values were high and significant for abundance and species richness, which were both correlated to rainfall. In conclusion, the restriction of species to some environments demonstrates the need to preserve these areas in order to avoid possible local extinction. Therefore, in extremely seasonable environments, such as the Caatinga, seasonal variation strongly affects dung beetle communities

    A New Approach for Heparin Standardization: Combination of Scanning UV Spectroscopy, Nuclear Magnetic Resonance and Principal Component Analysis

    The year 2007 was marked by widespread adverse clinical responses to heparin use, leading to a global recall of potentially affected heparin batches in 2008. Several analytical methods have since been developed to detect impurities in heparin preparations; however, many are costly and dependent on instrumentation with only limited accessibility. A method based on a simple UV-scanning assay, combined with principal component analysis (PCA), was developed to detect impurities, such as glycosaminoglycans, other complex polysaccharides and aromatic compounds, in heparin preparations. Results were confirmed by NMR spectroscopy. This approach provides an additional, sensitive tool to determine heparin purity and safety, even when NMR spectroscopy failed, requiring only standard laboratory equipment and computing facilities

    Overweight and constipation in adolescents

    Background: the association between overweight and gastrointestinal symptoms has been recently studied in the literature; however, few studies have evaluated the association between overweight and constipation in adolescents in a community-based sample. the aim of this study was to analyze the prevalence of constipation and its association with being overweight in a community-based survey with adolescents.Methods: This cross-sectional study included 1,077 adolescents who were enrolled in five schools in the city of Sao Jose dos Campos, Brazil. Constipation was defined according to modified and combined Rome III criteria for adolescents and adults. Being overweight was defined as a body mass index (BMI) that was equal to or greater than that of the 85(th) percentile for age and gender.Results: Constipation was diagnosed in 18.2% (196/1077) of the included adolescents. There was no significant difference in the prevalence of constipation in males and females who were both younger and older than 14 years. Fecal incontinence was observed in 25 adolescents, 22 (88.0%) of whom were diagnosed as being constipated. the prevalence of being overweight was found in 13.5% (145/1077) of the study population. the prevalence of constipation was observed to be similar in adolescents who were (19.4%; 28/144) and were not (18.0%; 168/933) overweight (p = 0.764; OR = 1.10). Fecal incontinence that was associated with constipation was more frequent in adolescents who were overweight (37.0%; 8/28) than in adolescents who were not overweight (8.5%; 14/168; p = 0.005; OR = 4.40).Conclusions: the prevalence of constipation was high among the investigated adolescents.     Comparative proteome and peptidome analysis of the cephalic fluid secreted by Arapaima gigas (Teleostei: Osteoglossidae) during and outside parental care

    Parental investment in Arapaima gigas includes nest building and guarding, followed by a care provision when a cephalic fluid is released from the parents’ head to the offspring. This fluid has presumably important functions for the offspring but so far its composition has not been characterised. In this study the proteome and peptidome of the cephalic secretion was studied in parental and non-parental fish using capillary electrophoresis coupled to mass spectrometry (CE-MS) and GeLC-MS/MS analyses. Multiple comparisons revealed 28 peptides were significantly different between males and parental males (PC-males), 126 between females and parental females (PC-females), 51 between males and females and 9 between PC-males and PC-females. Identification revealed peptides were produced in the inner ear (pcdh15b), eyes (tetraspanin and ppp2r3a), central nervous system (otud4, ribeye a, tjp1b and syn1) among others. A total of 422 proteins were also identified and gene ontology analysis revealed 28 secreted extracellular proteins. From these, 2 hormones (prolactin and stanniocalcin) and 12 proteins associated to immunological processes (serotransferrin, α-1-antitrypsin homolog, apolipoprotein A-I, and others) were identified. This study provides novel biochemical data on the lateral line fluid which will enable future hypotheses-driven experiments to better understand the physiological roles of the lateral line in chemical communication
