    Heterozygous Yeast Deletion Collection Screens Reveal Essential Targets of Hsp90

    Hsp90 is an essential eukaryotic chaperone with a role in folding specific “client” proteins such as kinases and hormone receptors. Previously performed homozygous diploid yeast deletion collection screens uncovered broad requirements for Hsp90 in cellular transport and cell cycle progression. These screens also revealed that the requisite cellular functions of Hsp90 change with growth temperature. We present here for the first time the results of heterozygous deletion collection screens conducted at the hypothermic stress temperature of 15°C. Extensive bioinformatic analyses were performed on the resulting data in combination with data from homozygous and heterozygous screens previously conducted at normal (30°C) and hyperthermic stress (37°C) growth temperatures. Our resulting meta-analysis uncovered extensive connections between Hsp90 and (1) general transcription, (2) ribosome biogenesis and (3) GTP binding proteins. Predictions from bioinformatic analyses were tested experimentally, supporting a role for Hsp90 in ribosome stability. Importantly, the integrated analysis of the 15°C heterozygous deletion pool screen with previously conducted 30°C and 37°C screens allows for essential genetic targets of Hsp90 to emerge. Altogether, these novel contributions enable a more complete picture of essential Hsp90 functions

    Inequitable walking conditions among older people: examining the interrelationship of neighbourhood socio-economic status and urban form using a comparative case study

    <p>Abstract</p> <p>Background</p> <p>Supportive neighbourhood walking conditions are particularly important for older people as they age and who, as a group, prefer walking as a form of physical activity. Urban form and socio-economic status (SES) can influence neighbourhood walking behaviour. The objectives of this study were: a) to examine how urban form and neighbourhood SES inter-relate to affect the experiences of older people who walk in their neighbourhoods; b) to examine differences among neighbourhood stakeholder key informant perspectives on socio-political processes that shape the walkability of neighbourhood environments.</p> <p>Methods</p> <p>An embedded comparative case study examined differences among four Ottawa neighbourhoods that were purposefully selected to provide contrasts on urban form (inner-urban versus suburban) and SES (higher versus lower). Qualitative data collected from 75 older walkers and 19 neighbourhood key informants, as well as quantitative indicators were compared on the two axes of urban form and SES among the four neighbourhoods.</p> <p>Results and discussion</p> <p>Examining the inter-relationship of neighbourhood SES and urban form characteristics on older people's walking experiences indicated that urban form differences were accentuated positively in higher SES neighbourhoods and negatively in lower SES neighbourhoods. Older people in lower SES neighbourhoods were more affected by traffic hazards and more reliant on public transit compared to their higher SES counterparts. In higher SES neighbourhoods the disadvantages of traffic in the inner-urban neighbourhood and lack of commercial destinations in the suburban neighbourhood were partially offset by other factors including neighbourhood aesthetics. Key informant descriptions of the socio-political process highlighted how lower SES neighbourhoods may face greater challenges in creating walkable places. These differences pertained to the size of neighbourhood associations, relationships with political representatives, accessing information and salient neighbourhood association issues. Findings provide evidence of inequitable walking environments.</p> <p>Conclusion</p> <p>Future research on walking must consider urban form-SES inter-relationships and further examine the equitable distribution of walking conditions as well as the socio-political processes driving these conditions. There is a need for municipal governments to monitor differences in walking conditions among higher and lower SES neighbourhoods, to be receptive to the needs of lower SES neighbourhood and to ensure that policy decisions are taken to address inequitable walking conditions.</p

    The stability of blood Eosinophils in chronic obstructive pulmonary disease

    Blood eosinophils are a predictive biomarker of inhaled corticosteroid response in chronic obstructive pulmonary disease (COPD). We investigated blood eosinophil stability over 1 year using the Global Initiative for Chronic Obstructive Lung Disease (GOLD) 2019 thresholds of < 100, 100- < 300 and ≥ 300 eosinophils/μL in 225 patients from the COPDMAP cohort. Blood eosinophils showed good stability (rho: 0.71, p < 0.001, ICC 0.84), and 69.3% of patients remained in the same eosinophil category at 1 year. 85.3% of patients with eosinophils < 100 cells/μL had stable counts. The majority of blood eosinophil counts remain stable over 1 year using the GOLD 2019 thresholds

    Oscillating red giants in eclipsing binary systems: empirical reference value for asteroseismic scaling relation

The internal structures and properties of oscillating red-giant stars can be accurately inferred through their global oscillation modes (asteroseismology). Based on 1460 d of Kepler observations we perform a thorough asteroseismic study to probe the stellar parameters and evolutionary stages of three red giants in eclipsing binary systems. We present the first detailed analysis of individual oscillation modes of the red-giant components of KIC 8410637, KIC 5640750, and KIC 9540226. We obtain estimates of their asteroseismic masses, radii, mean densities, and logarithmic surface gravities by using the asteroseismic scaling relations as well as grid-based modelling. As these red giants are in double-lined eclipsing binaries, it is possible to derive their independent dynamical masses and radii from the orbital solution and compare it with the seismically inferred values. For KIC 5640750 we compute the first spectroscopic orbit based on both components of this system. We use high-resolution spectroscopic data and light curves of the three systems to determine up-to-date values of the dynamical stellar parameters. With our comprehensive set of stellar parameters we explore consistencies between binary analysis and asteroseismic methods, and test the reliability of the well-known scaling relations. For the three red giants under study, we find agreement between dynamical and asteroseismic stellar parameters in cases where the asteroseismic methods account for metallicity, temperature, and mass dependence as well as surface effects. We are able to attain agreement from the scaling laws in all three systems if we use Δ ref, emp = 130.8 ± 0.9 μHz instead of the usual solar reference value.status: publishe

    Xenodiagnoses were performed every 3 hours using 10 Triatoma infestans 3rd instar for 24 to 72 hours, in 18 chronic chagasics with positive serology and/or xenodiagnosis. There was no statistically significant difference in the positivity of assays performed during the day (9:00 to 18:00h) compared to those performed at night (21:00 to 6:00h), (chi² = 0.1526 p = 0.696). Xenodiagnosis was performed in ten of the patients for 13 successive days but there was no periodicity detected in the positive assays.<br>Realizamos xenodiagnósticos de 3 em 3 horas com 10 ninfas do 3° estágio de Triatoma infestans por um período de 24 a 72 horas, em 18 chagásicos crônicos que tinham sorologia e/ou xenodiagnóstico positivos.     Exploring the distribution of park availability, features, and quality across Kansas City, Missouri by income and race/ethnicity: an environmental justice investigation

    Background: Parks are key community assets for physical activity, but some evidence suggests these resources are not equitably distributed. Purpose: This study examined disparities by income and race/ethnicity in the availability, features, and quality of parks across Kansas City, Missouri. Methods: All parks and census tracts (CTs) were mapped using GIS and park features and quality were determined via audits. MANCOVAs analyzed differences in park availability, features, and quality across low, medium and high income and race/ethnicity CT tertiles. Results: Low income CTs contained significantly more parks, but also had fewer parks with playgrounds and more quality concerns per park. High minority CTs had more parks with basketball courts, but fewer parks with trails. Medium income CTs contained more aesthetic features per park. Conclusions: Future research should examine policies that contribute to and that might rectify disparities in park features and quality, especially in low income and high minority areas
