    Evaluation of a toxoid fusion protein vaccine produced in plants to protect poultry against necrotic enteritis

    Background Necrotic enteritis (NE) is caused by type A strains of the bacterium Clostridium perfringens. Total global economic losses to the poultry industry due to NE is estimated to be over two billion dollars annually. Traditionally, NE has been effectively controlled by inclusion of antibiotics in the diet of poultry. However, recent concerns regarding the impact of this practice on increasing antibiotic resistance in human pathogens have led us to consider alternative approaches, such as vaccination, for controlling this disease. NE strains of C. perfringens produce two major toxins, a-toxin and NetB. Immune responses against either toxin can provide partial protection against NE. Methods We have developed a fusion protein combining a non-toxic carboxyl-terminal domain of a-toxin (PlcC) and an attenuated, mutant form of NetB (NetB-W262A) for use as a vaccine antigen to immunize poultry against NE. We utilized a DNA sequence that was codon-optimized for Nicotiana benthamiana to enable high levels of expression. The 6-His tagged PlcC-NetB fusion protein was synthesized in N. benthamiana using a geminiviral replicon transient expression system, purified by metal affinity chromatography, and used to immunize broiler birds. Results Immunized birds produced a strong serum IgY response against both the plant produced PlcC-NetB protein and against bacterially produced His-PlcC and His-NetB. Immunized birds were significantly protected against a subsequent in-feed challenge with virulent C. perfringens when treated with the fusion protein. These results indicate that a plant-produced PlcC-NetB toxoid is a promising vaccine candidate for controlling NE in poultry

    Can forest management based on natural disturbances maintain ecological resilience?

    Given the increasingly global stresses on forests, many ecologists argue that managers must maintain ecological resilience: the capacity of ecosystems to absorb disturbances without undergoing fundamental change. In this review we ask: Can the emerging paradigm of natural-disturbance-based management (NDBM) maintain ecological resilience in managed forests? Applying resilience theory requires careful articulation of the ecosystem state under consideration, the disturbances and stresses that affect the persistence of possible alternative states, and the spatial and temporal scales of management relevance. Implementing NDBM while maintaining resilience means recognizing that (i) biodiversity is important for long-term ecosystem persistence, (ii) natural disturbances play a critical role as a generator of structural and compositional heterogeneity at multiple scales, and (iii) traditional management tends to produce forests more homogeneous than those disturbed naturally and increases the likelihood of unexpected catastrophic change by constraining variation of key environmental processes. NDBM may maintain resilience if silvicultural strategies retain the structures and processes that perpetuate desired states while reducing those that enhance resilience of undesirable states. Such strategies require an understanding of harvesting impacts on slow ecosystem processes, such as seed-bank or nutrient dynamics, which in the long term can lead to ecological surprises by altering the forest's capacity to reorganize after disturbance

    Indirect signals from light neutralinos in supersymmetric models without gaugino mass unification

    We examine indirect signals produced by neutralino self-annihilations, in the galactic halo or inside celestial bodies, in the frame of an effective MSSM model without gaugino-mass unification at a grand unification scale. We compare our theoretical predictions with current experimental data of gamma-rays and antiprotons in space and of upgoing muons at neutrino telescopes. Results are presented for a wide range of the neutralino mass, though our discussions are focused on light neutralinos. We find that only the antiproton signal is potentially able to set constraints on very low-mass neutralinos, below 20 GeV. The gamma-ray signal, both from the galactic center and from high galactic latitudes, requires significantly steep profiles or substantial clumpiness in order to reach detectable levels. The up-going muon signal is largely below experimental sensitivities for the neutrino flux coming from the Sun; for the flux from the Earth an improvement of about one order of magnitude in experimental sensitivities (with a low energy threshold) can make accessible neutralino masses close to O, Si and Mg nuclei masses, for which resonant capture is operative.Comment: 17 pages, 1 tables and 5 figures, typeset with ReVTeX4.     A poda verde é uma prática cultural utilizada para melhorar as condições do dossel vegetativo dos vinhedos, visando a favorecer a qualidade da uva e do vinho. Nesse sentido, realizou-se este experimento entre as safras de 1993/1994 e 1996/1997, com diferentes modalidades de poda verde, num vinhedo do cv. Merlot conduzido em latada. Houve 12 tratamentos e três repetições, sendo o delineamento experimental em blocos casualizados. Os tratamentos constituíram-se da testemunha e de 11 diferentes modalidades de poda verde, ou seja, desbrota, desponta e desfolha, algumas delas em diferentes épocas do ciclo vegetativo da videira. O componente principal 1, da análise de componentes principais (ACP) feita em cada ano, separadamente, mostra que o tratamento 10 (desbrota + desponta + desfolha realizada no início da floração, eliminando-se as folhas abaixo dos cachos) discriminou-se nos quatro anos, e os tratamentos 7 (desfolha realizada 21 dias antes da colheita, eliminando-se metade das folhas abaixo dos cachos) e 6 (desfolha realizada 21 dias antes da colheita, eliminando-se as folhas abaixo dos cachos), em três deles; a ACP da média dos quatro anos também evidencia essa discriminação entre eles. Constata-se que o tratamento 10 foi um dos que tiveram intensidade de poda verde mais intensa, caracterizando-se por variáveis indicativas de plantas com vigor e produtividade mais baixos que os demais

    Non-local rheology in dense granular flows -- Revisiting the concept of fluidity

    Granular materials belong to the class of amorphous athermal systems, like foams, emulsion or suspension they can resist shear like a solid, but flow like a liquid under a sufficiently large applied shear stress. They exhibit a dynamical phase transition between static and flowing states, as for phase transitions of thermodynamic systems, this rigidity transition exhibits a diverging length scales quantifying the degree of cooperatively. Several experiments have shown that the rheology of granular materials and emulsion is non-local, namely that the stress at a given location does not depend only on the shear rate at this location but also on the degree of mobility in the surrounding region. Several constitutive relations have recently been proposed and tested successfully against numerical and experimental results. Here we use discrete elements simulation of 2D shear flows to shed light on the dynamical mechanism underlying non-locality in dense granular flows

    Reproductive aging-associated common genetic variants and the risk of breast cancer

    Introduction: A younger age at menarche and an older age at menopause are well established risk factors for breast cancer. Recent genome-wide association studies have identified several novel genetic loci associated with these two traits. However, the association between these loci and breast cancer risk is unknown.Methods: In this study, we investigated 19 and 17 newly identified single nucleotide polymorphisms (SNPs) from the ReproGen Consortium that have been associated with age at menarche and age at natural menopause, respectively, and assessed their associations with breast cancer risk in 6 population-based studies among up to 3,683 breast cancer cases and 34,174 controls in white women of European ancestry. In addition, we used these SNPs to calculate genetic risk scores (GRSs) based on their associations with each trait.Results: After adjusting for age and potential population stratification, two age at menarche associated SNPs (rs1079866 and rs7821178) and one age at natural menopause associated SNP (rs2517388) were associated with breast cancer risk (p values, 0.003, 0.009 and 0.023, respectively). The odds ratios for breast cancer corresponding to per-risk-allele were 1.14 (95% CI, 1.05 to 1.24), 1.08 (95% CI, 1.02 to 1.15) and 1.10 (95% CI, 1.01 to 1.20), respectively, and were in the direction predicted by their associations with age at menarche or age at natural menopause. These associations did not appear to be attenuated by further controlling for self-reported age at menarche, age at natural menopause, or known breast cancer susceptibility loci. Although we did not observe a statistically significant association between any GRS for reproductive aging and breast cancer risk, the 4 th and 5 th highest quintiles of the younger age at menarche GRS had odds ratios of 1.14 (95% CI, 1.01 to 1.28) and 1.13 (95% CI, 1.00 to 1.27), respectively, compared to the lowest quintile.Conclusions: Our study suggests that three genetic variants, independent of their associations with age at menarche or age at natural menopause, were associated with breast cancer risk and may contribute modestly to breast cancer risk prediction; however, the combination of the 19 age at
