    Meta-analysis of data from animal studies:A practical guide

    AbstractMeta-analyses of data from human studies are invaluable resources in the life sciences and the methods to conduct these are well documented. Similarly there are a number of benefits in conducting meta-analyses on data from animal studies; they can be used to inform clinical trial design, or to try and explain discrepancies between preclinical and clinical trial results. However there are inherit differences between animal and human studies and so applying the same techniques for the meta-analysis of preclinical data is not straightforward. For example preclinical studies are frequently small and there is often substantial heterogeneity between studies. This may have an impact on both the method of calculating an effect size and the method of pooling data. Here we describe a practical guide for the meta-analysis of data from animal studies including methods used to explore sources of heterogeneity

    Understanding the relationship between social determinants of health and maternal mortality

    This paper aims to inform those involved in the care of pregnant women in the UK about the relationship between social determinants of health and the risk of maternal death. Social determinants are the social conditions in which people are born, grow, live, work and age. These conditions can shape a person's health, their exposure to illness and access to care. The worse social and economic positions people are in, the worse their health outcomes are likely to be. Exploring these issues in relation to maternal outcomes is timely because the profile of mothers giving birth in the UK is changing. Increasingly, babies are being born to older or obese mothers, or to women born outside of the UK. The evidence suggests a strong relationship between social determinants and poor maternal outcomes, including an increased risk of maternal death. Despite the provision of free reproductive and maternity services, current models of care are still failing those pregnant women living in adverse social circumstances prior to, during and after pregnancy. The reasons are complex but maternal outcomes are particularly poor for socially disadvantaged women affected by pre- existing physical or mental health problems, those who misuse substances, have a low level of education, are overweight, undernourished or poorly sheltered, and those who are at increased risk because of abusive and unsupportive partners, families and peers. Some traditional, cultural and social norms, and reproductive laws and policies, also appear to influence a pregnant woman's relationships, behaviour and access to healthcare services. These factors can reinforce a web of social disadvantage and poor outcomes for some pregnant women, which can persist for future generations. Public awareness of these issues has increased as a result of the COVID- 19 pandemic which has highlighted the ways in which health inequalities can adversely affect health outcomes beyond an individual's control. However, addressing the relationship between COVID- 19 and social determinants of health is outside the scope of this paper. While social determinants of maternal health also have an impact on offspring health, this is also outside the remit of this paper. Healthcare professionals have limited power on their own to modify social determinants of health, but this paper concludes with recommendations to improve the maternal outcomes of vulnerable pregnant women residing in the UK and proposes improvements to the collection and reporting of maternal outcome data, with further investment in prevention and intervention measures to better support these women. Within this document we use the terms pregnant woman and women's health. However, it is important to acknowledge that it is not only people who identify as women for whom it is necessary to access care. Obstetric and gynaecology services and delivery of care must therefore be appropriate, inclusive and sensitive to the needs of those individuals whose gender identity does not align with the sex they were assigned at birth

    Adaptor and Clathrin Exchange at the Plasma Membrane and trans-Golgi Network

    We previously demonstrated, using fluorescence recovery after photobleaching, that clathrin in clathrin-coated pits at the plasma membrane exchanges with free clathrin in the cytosol, suggesting that clathrin-coated pits are dynamic structures. We now investigated whether clathrin at the trans-Golgi network as well as the clathrin adaptors AP2 and AP1 in clathrin-coated pits at the plasma membrane and trans-Golgi network, respectively, also exchange with free proteins in the cytosol. We found that when the budding of clathrin-coated vesicle is blocked without significantly affecting the structure of clathrin-coated pits, both clathrin and AP2 at the plasma membrane and clathrin and AP1 at the trans-Golgi network exchange rapidly with free proteins in the cytosol. In contrast, when budding of clathrin-coated vesicles was blocked at the plasma membrane or trans-Golgi network by hypertonic sucrose or K(+) depletion, conditions that markedly affect the structure of clathrin-coated pits, clathrin exchange was blocked but AP2 at the plasma membrane and both AP1 and the GGA1 adaptor at the trans-Golgi network continue to rapidly exchange. We conclude that clathrin-coated pits are dynamic structures with rapid exchange of both clathrin and adaptors and that adaptors are able to exchange independently of clathrin when clathrin exchange is blocked

    From plankton to top predators: bottom-up control of a marine food web across four trophic levels

    Abundant mid-trophic pelagic fish often play a central role in marine ecosystems, both as links between zooplankton and top predators and as important fishery targets. In the North Sea, the lesser sandeel occupies this position, being the main prey of many bird, mammal and fish predators and the target of a major industrial fishery. However, since 2003, sandeel landings have decreased by > 50%, and many sandeel-dependent seabirds experienced breeding failures in 2004. Despite the major economic implications, current understanding of the regulation of key constituents of this ecosystem is poor. Sandeel abundance may be regulated 'bottom-up' by food abundance, often thought to be under climatic control, or 'top-down' by natural or fishery predation. We tested predictions from these two hypotheses by combining unique long-term data sets (1973–2003) on seabird breeding productivity from the Isle of May, SE Scotland, and plankton and fish larvae from the Continuous Plankton Recorder survey. We also tested whether seabird breeding productivity was more tightly linked to sandeel biomass or quality (size) of individual fish. The biomass of larval sandeels increased two- to threefold over the study period and was positively associated with proxies of the abundance of their plankton prey. Breeding productivity of four seabirds bringing multiple prey items to their offspring was positively related to sandeel larval biomass with a 1-year lag, indicating dependence on 1-year-old fish, but in one species bringing individual fish it was strongly associated with the size of adult sandeels. These links are consistent with bottom-up ecosystem regulation and, with evidence from previous studies, indicate how climate-driven changes in plankton communities can affect top predators and potentially human fisheries through the dynamics of key mid-trophic fish. However, the failing recruitment to adult sandeel stocks and the exceptionally low seabird breeding productivity in 2004 were not associated with low sandeel larval biomass in 2003, so other mechanisms (e.g. predation, lack of suitable food after metamorphosis) must have been important in this case.
