Pluronic F68 block polymer, a very potent suppressor of carcinogenesis in the colon of rats and mice
Polyethylene-glycol (PEG) is a strong inhibitor of colon cancer in rats, and the most potent suppressor of aberrant crypt foci. 9 PEG-like block copolymers were tested in rodents, after an azoxymethane injection. Dietary pluronic F68 led to a 98.6% reduction in the number of aberrant crypt foci in a first rat study (P< 0.0001). Next 3 studies confirmed this pluronic efficacy in rats and mice. This non-toxic laxative seems roughly 5 times more potent than PEG for chemoprevention. © 2001 Cancer Research Campaign http://www.bjcancer.co
Processes governing development of ecotoxicity in clayey and silty soils incubated with olive mill wast water under different temperature and humidity conditions
Olive oil production generates olive mill wastewater (OMW) with a high content in nutrients and phenolic substances. Its application to soil could be a cost-effective solution for recycling. However, the degree of toxic effects of OMW on soil biota is widely unknown and has to be considered when searching for adisposal strategy.
The objective of this study was to understand the degradation process of OMW organic matter and its influence on toxic effects as well as soil properties. We hypothesized that OMW toxicity decreases with degradation of its phenolic components. A higher soil biological activity was expected to increase degradation.
We incubated a clayey soil and a silty soil with OMW for 60 days under conditions typical for this region in order to simulate the application during various seasons (winter, spring, summer dry, summer wet). Soil respiration, pH, electrical conductivity, total phenolic content as well as anion and cation content, specific ultraviolet absorbance at 254 nm and dissolved organic carbon were measured at ten points of time during incubation. Soils and methanolic soil extracts were tested for ecotoxicity using Lepidium sativum germination and Folsomia candida egg hatching rate.
The degradation and transformation of OMW-organic matter was stronger under warm and humid conditions than under cold and dry conditions. It was furthermore enhanced in the clayey soil compared to the silty soil. Most severe ecotoxicological effects were found under summer dry conditions while spring as well as summer wet conditions led to a fast recovery of both germination and hatching. However, the silty soil did not recover to preapplication levels. In the clayey soil, germination parameter were higher than in control after around 30 days suggesting a fertilizing effect. Effects in methanol extracts were higher in all soils and climatic scenarios. Therefore, remobilization of OMW derived toxic compounds has to be considered on a long-term scale. Egg hatching as most sensitive life-cycle parameter of Folsomia candida showed also the same relation to climatic conditions and soil type but was more robust to OMW compared to Lepidium sativum.
Environmental conditions as well as soil type are key factors determining degradation of OMW organic matter and OMW derived ecotoxicity. Therefore, spring application (warm and wet) of OMW seems to be a compromise with regard to OMW recycling, OMW occurrence in winter and farmer considerations
Pneumothorax spontané et emphysème pulmonaire chez les consommateurs de cannabis
INTRODUCTION: If pulmonary complications of tobacco smoking are well documented, those associated with cannabis use are less known.
OBJECTIVES: Systematic literature review of data on pneumothorax and lung emphysema in cannabis users.
DOCUMENTARY SOURCES: Medline, on the period 1980-2018 with the following keywords cannabis or marijuana and pneumothorax or emphysema, limits "title/abstract". Among 97 articles, 42 abstracts have given use to a dual reading to select 20 studies.
RESULTS: Eighteen case reports (8 with SP) showed bullae in the upper lobes in combined cannabis and tobacco smokers (CS) and in the 2 cannabis only smokers (COS). The risk of SP was increased in CS, but not in COS. In patients less than 35-years old presenting with SP, the incidence of bullae on thoracic computed tomography (CT) was higher in CS than in tobacco only smokers (TOS). CT in patients with SP showed no significant difference as regards of the prevalence, location and type of emphysema between CS and TOS. Proportion of low lung density areas was higher in CS than in non-smokers (NS), but was similar in TOS and NS.
CONCLUSION: These results suggest a cumulative toxic effect of tobacco and cannabis on the risk of SP and lung emphysema

Exponential Random Graph Modeling for Complex Brain Networks
Exponential random graph models (ERGMs), also known as p* models, have been
utilized extensively in the social science literature to study complex networks
and how their global structure depends on underlying structural components.
However, the literature on their use in biological networks (especially brain
networks) has remained sparse. Descriptive models based on a specific feature
of the graph (clustering coefficient, degree distribution, etc.) have dominated
connectivity research in neuroscience. Corresponding generative models have
been developed to reproduce one of these features. However, the complexity
inherent in whole-brain network data necessitates the development and use of
tools that allow the systematic exploration of several features simultaneously
and how they interact to form the global network architecture. ERGMs provide a
statistically principled approach to the assessment of how a set of interacting
local brain network features gives rise to the global structure. We illustrate
the utility of ERGMs for modeling, analyzing, and simulating complex
whole-brain networks with network data from normal subjects. We also provide a
foundation for the selection of important local features through the
implementation and assessment of three selection approaches: a traditional
p-value based backward selection approach, an information criterion approach
(AIC), and a graphical goodness of fit (GOF) approach. The graphical GOF
approach serves as the best method given the scientific interest in being able
to capture and reproduce the structure of fitted brain networks
Aims and challenges of building national trainee networks in clinical microbiology and infectious disease disciplines
Trainees represent the medical practice of tomorrow. Interactions and collaborations at the early stage in career will strengthen the future of our specialties, clinical microbiology and infectious diseases. Trainee networks at the national level help access the best education and career opportunities. The aim of this collaborative white paper between the Trainee Association of European Society of Clinical Microbiology and Infectious Diseases (ESCMID) and four national trainee networks is to discuss the motivation for building such networks and offer guidance for their creation and sustainability even during a health crisis.Publisher PDFPeer reviewe
Obesity-induced insulin resistance in human skeletal muscle is characterised by defective activation of p42/p44 MAP kinase
Insulin resistance (IR), an impaired cellular, tissue and whole body response to insulin, is a major pathophysiological defect of type 2 diabetes mellitus. Although IR is closely associated with obesity, the identity of the molecular defect(s) underlying obesity-induced IR in skeletal muscle remains controversial; reduced post-receptor signalling of the insulin receptor substrate 1 (IRS1) adaptor protein and downstream effectors such as protein kinase B (PKB) have previously been implicated. We examined expression and/or activation of a number of components of the insulin-signalling cascade in skeletal muscle of 22 healthy young men (with body mass index (BMI) range, 20–37 kg/m2). Whole body insulin sensitivity (M value) and body composition was determined by the hyperinsulinaemic (40 mU. min−1.m−2.), euglycaemic clamp and by dual energy X-ray absorptiometry (DEXA) respectively. Skeletal muscle (vastus lateralis) biopsies were taken before and after one hour of hyperinsulinaemia and the muscle insulin signalling proteins examined by western blot and immunoprecipitation assay. There was a strong inverse relationship between M-value and BMI. The most striking abnormality was significantly reduced insulin-induced activation of p42/44 MAP kinase, measured by specific assay, in the volunteers with poor insulin sensitivity. However, there was no relationship between individuals' BMI or M-value and protein expression/phosphorylation of IRS1, PKB, or p42/44 MAP kinase protein, under basal or hyperinsulinaemic conditions. In the few individuals with poor insulin sensitivity but preserved p42/44 MAP kinase activation, other signalling defects were evident. These findings implicate defective p42/44 MAP kinase signalling as a potential contributor to obesity-related IR in a non-diabetic population, although clearly multiple signalling defects underlie obesity associated IR
Using the SWAT model to improve process descriptions and define hydrologic partitioning in South Korea
Watershed-scale modeling can be a valuable tool to aid in quantification of
water quality and yield; however, several challenges remain. In many
watersheds, it is difficult to adequately quantify hydrologic partitioning.
Data scarcity is prevalent, accuracy of spatially distributed meteorology is
difficult to quantify, forest encroachment and land use issues are common,
and surface water and groundwater abstractions substantially modify
watershed-based processes. Our objective is to assess the capability of the
Soil and Water Assessment Tool (SWAT) model to capture event-based and long-term monsoonal rainfall–runoff
processes in complex mountainous terrain. To accomplish this, we developed a
unique quality-control, gap-filling algorithm for interpolation of high-frequency meteorological data. We used a novel multi-location,
multi-optimization calibration technique to improve estimations of
catchment-wide hydrologic partitioning. The interdisciplinary model was
calibrated to a unique combination of statistical, hydrologic, and plant
growth metrics. Our results indicate scale-dependent sensitivity of
hydrologic partitioning and substantial influence of engineered features.
The addition of hydrologic and plant growth objective functions identified
the importance of culverts in catchment-wide flow distribution. While this
study shows the challenges of applying the SWAT model to complex terrain and
extreme environments; by incorporating anthropogenic features into modeling
scenarios, we can enhance our understanding of the hydroecological impact
