    Environmental history of an urban wetland: from degraded colonial resource to nature conservation area

    This paper provides an account of the environmental history of a landlocked coastal wetland and the surrounding nature space (Lake Claremont) located within a large Australian city (Perth, Western Australia). We document the processes and behaviour that led to significant negative change over time and report on recent work to restore the lake and its surrounding ecosystem. The reported community-initiated actions reflect changing human values and add to the accumulating evidence that the increasing trend of restoring greenspace to native ecosystems (nature space) brings about social and conservation benefits within the urban fabric. Moreover, this narrative also indicates that significant progress can be made in recovering and expanding ecological components and natural values within a relatively short period. Implications for city planners and land managers are self-evident with regard to what can be achieved and the values that communities now place on naturally functioning urban wetlands. Furthermore, the presence of wetlands such as Lake Claremont in the urban fabric is seen to enhance the liveability of the surrounding city by offering a convenient way for residents and tourists to connect with nature and the outdoors and realise the health benefits that the lake can provide. This case study adds to the body of work that identifies and describes the growing importance of urban wetlands in urban landscapes and provides a clear example of what can be achieved at the local community level and with local government support

    Scoping recreational disturbance of shorebirds to inform the agenda for research and management in Tropical Asia

    In addition to scoping the impacts of the four most reported sources of recreational disturbance on shorebirds, this study also advances the concept of Tropical Asia (TA) to collectively describe tourist destinations in the ecologically and geopolitically diverse part of the planet that incorporates the tourism megaregion of South and Southeast Asia. At a time of growing global concern about the rapid decline of shorebird populations, many governments in TA are embracing and capitalising on the exponential growth in demand for coastal recreation and tourism across the region. This political response is partly driven by efforts to deliver economic development, aligned to the United Nations Sustainable Development Goals, in order to secure the livelihoods of people living in less developed coastal areas. However, the rapid increase in visitor numbers and the development of infrastructure to support the booming demand for coastal tourism destinations in TA are further exacerbating the pressures on shorebird populations across the region. Despite these growing pressures and the wealth of research reporting on shorebird populations across the Asian flyways, this scoping study identified surprisingly little research that reports on the recreational disturbance of shorebirds in TA. While undertaken to inform future research, this study also provides a synthesis of management strategies reported in the global literature into a set of management recommendations for coastal destinations in TA

    Novel instrumentation and method development for a quadrupole ion trap mass spectrometer

    Part of the work described in this dissertation has involved development of instrumentation for the analysis of aerosol compounds. A quadrupole ion trap mass spectrometer (QITMS) is modified to sample particles via an aerodynamic lens inlet. The instrument has a unique capability for volatilization and chemical ionization within the heated ion trap that reduces fragmentation compared to other methods. Compounds in particles are detected in real time at low concentrations, over a broad concentration range, and in complex mixtures. Unlike most mass spectrometry systems used for aerosol detection, the aerosol QITMS is able to perform tandem mass spectrometry (MS/MS) to identify analyte ions. The performance of the instrument is demonstrated by monitoring the ozonolysis reaction of a volatile organic compound producing aerosol particles. The aerosol QITMS is presented as an effective design for detection and analysis of aerosol compounds. This work also describes the development of more efficient and informative MS/MS methods using infrared multiphoton photodissociation (IRMPD). IRMPD experiments are made practical for the QITMS by focusing the laser irradiation to increase photon flux on trapped ions. Efficient performance is used to increase sensitivity to product ions. Trapped ion clouds are profiled with higher resolution than previously obtained to characterize the effects of trapping parameters. IRMPD is also made selective by controlling the spatial distribution of ions relative to the laser pathway. More informative MS/MS spectra can be acquired without empirical determination of irradiation time, as is necessary for conventional IRMPD. The methods described use the fundamentals of ion trapping to design more effective photodissociation MS/MS experiments

    Effects of recreational camping on the environmental values of national parks in Sri Lanka

    Camping is a popular activity in the contemporary nature-based tourism domain and rapidly gaining momentum as a key recreational activity in Sri Lanka’s national parks (NPs). Recreational uses such as camping in natural areas can induce significant and often localised resource impacts that can affect soil, vegetation, wildlife and water, with the severity of such impacts varying according to the intensity of use. Hence, monitoring of the biophysical conditions of campsites has become an important component in the reserve management agenda in many places, especially in developed countries. To the best of our knowledge, this is the first study to examine the biophysical impacts associated with the recreation ecology of camping in Sri Lanka. Ten campsites from three dry zone NPs were selected to assess biophysical impacts of camping activities. Field measurements were based on the fixed radial transect method. Gathered data included the total area of the campsite, erosion potential measured as the area of exposed soil (devoid of vegetation or organic litter), number of exposed roots and human damage to trees, number of fireplaces/ fire scars on the ground, visual counts of litter, soil compaction measured by penetrometer, loss of woody debris. This study reports significant levels of environmental degradation related to all the indictors of biophysical impacts at both high and low use campsites. There was no evidence for any difference in the level of environmental degradation associated with high and low use campsites. The loss of natural values associated with campsites negatively impacted visitors’ nature-based experience. These findings highlight the importance of managing biophysical impacts in campsites to provide a high-quality visitor experience, while sustainably managing tourism activities in NPs

    Development of an invasively monitored porcine model of acetaminophen-induced acute liver failure

    Background: The development of effective therapies for acute liver failure (ALF) is limited by our knowledge of the pathophysiology of this condition, and the lack of suitable large animal models of acetaminophen toxicity. Our aim was to develop a reproducible invasively-monitored porcine model of acetaminophen-induced ALF. Method: 35kg pigs were maintained under general anaesthesia and invasively monitored. Control pigs received a saline infusion, whereas ALF pigs received acetaminophen intravenously for 12 hours to maintain blood concentrations between 200-300 mg/l. Animals surviving 28 hours were euthanased. Results: Cytochrome p450 levels in phenobarbital pre-treated animals were significantly higher than non pre-treated animals (300 vs 100 pmol/mg protein). Control pigs (n=4) survived 28-hour anaesthesia without incident. Of nine pigs that received acetaminophen, four survived 20 hours and two survived 28 hours. Injured animals developed hypotension (mean arterial pressure; 40.8+/-5.9 vs 59+/-2.0 mmHg), increased cardiac output (7.26+/-1.86 vs 3.30+/-0.40 l/min) and decreased systemic vascular resistance (8.48+/-2.75 vs 16.2+/-1.76 mPa/s/m3). Dyspnoea developed as liver injury progressed and the increased pulmonary vascular resistance (636+/-95 vs 301+/-26.9 mPa/s/m3) observed may reflect the development of respiratory distress syndrome. Liver damage was confirmed by deterioration in pH (7.23+/-0.05 vs 7.45+/-0.02) and prothrombin time (36+/-2 vs 8.9+/-0.3 seconds) compared with controls. Factor V and VII levels were reduced to 9.3 and 15.5% of starting values in injured animals. A marked increase in serum AST (471.5+/-210 vs 42+/-8.14) coincided with a marked reduction in serum albumin (11.5+/-1.71 vs 25+/-1 g/dL) in injured animals. Animals displayed evidence of renal impairment; mean creatinine levels 280.2+/-36.5 vs 131.6+/-9.33 mumol/l. Liver histology revealed evidence of severe centrilobular necrosis with coagulative necrosis. Marked renal tubular necrosis was also seen. Methaemoglobin levels did not rise >5%. Intracranial hypertension was not seen (ICP monitoring), but there was biochemical evidence of encephalopathy by the reduction of Fischer's ratio from 5.6 +/- 1.1 to 0.45 +/- 0.06. Conclusion: We have developed a reproducible large animal model of acetaminophen-induced liver failure, which allows in-depth investigation of the pathophysiological basis of this condition. Furthermore, this represents an important large animal model for testing artificial liver support systems

    Effect of claw horn lesion type and severity at the time of treatment on outcome of lameness in dairy cows

    Prompt diagnosis and treatment of claw horn lesions in cattle affects the likelihood of recovery; however, it is unknown if the type of lesion influences the likelihood of recovery. The aim of this study was to investigate whether the type, severity and frequency of claw horn lesions in newly lame cows (lame for no more than 2 weeks) at the time of corrective foot trimming affects the probability of recovery from lameness after treatment. The images of 112 feet (224 claws) from newly lame cows (n = 112; lame in only one hind foot), which were treated with a standardised therapeutic hoof trim only, were used to score claw horn lesions (sole ulcer, sole haemorrhage, white line haemorrhage or white line separation). Most cows (n = 107/112; 95.5%) were classified as mildly lame at the time of treatment. The proportion of cows that recovered 2 weeks after therapeutic hoof trimming was 88/112 (78.6%). Results of a multilevel logistic regression model indicated that severely lame cows were less likely to recover than those that were mildly lame (odds ratio, OR, 0.16; P = 0.04). White line haemorrhage had a significant negative impact on the likelihood of recovery from lameness (OR 0.14; P > 0.01); however, recovery of cows with white line haemorrhage was positively associated with the length of the lesion (OR 1.05; P = 0.03). This latter finding may be associated with the severity of the lesion, since mild claw horn lesions affected a significantly larger area of the claw than more severe lesions.
