    Metabolomic characterization of strawberry cultivars during postharvest

    The cultivated strawberry (Fragaria x ananassa) is the berry most consumed worldwide and is well-known for its delicate flavour and nutritional characteristics. However, strawberries possess a very short postharvest shelf-life due to their high respiration rate and their susceptibility to water loss, mechanical damage and fungi deterioration. Extension of fruit shelf-life is a major economic goal, and measures are commercially taken to delay senescence. These procedures include low temperature, controlled atmosphere and/or chemical treatments, being the first one the most commonly applied. To improve our understanding of the molecular and biochemical mechanisms underlying the deterioration of fruit quality attributes during senescence, we monitored the metabolomic profiles of five commercial strawberry cultivars under different postharvest treatments. Ripe fruits of F x ananassa cv. ‘Amiga’, ‘Camarosa’, ‘Candonga’, ‘Fortuna’ and ‘Santa Clara’ were harvested and kept at 4ÂșC during three, six and ten days in normal, CO2-enriched and O3-enriched atmospheres. We used a combination of GC-TOF-MS, LC-MS and GC-SPME-MS to identify and semi-quantify 49 primary metabolites (sugars, amino and organic acids), 132 polar secondary metabolites and 70 volatile compounds in all different treatments along postharvest stages. Multivariate statistical approaches, including hierarchical cluster analysis, partial least squares discriminant analyses and k-means clustering, were used to characterize the variation in metabolite content during the strawberry fruit postharvest life and to identify the biochemical pathways which are most affected in the senescence processes. Here, we present the main changes in volatile compounds, primary and secondary metabolites as a consequence of postharvest storage, highlighting the differences between cultivars and treatments.     Use of cystatin C to inform metformin eligibility among adult veterans with diabetes.

    AimsRecommendations for metformin use are dependent on eGFR category: eGFR >45 ml/min/1.73 m2 - "first-line agent"; eGFR 30-44 - "use with caution"; eGFR<30 - "do not use". Misclassification of metformin eligibility by creatinine-based MDRD GFR estimates (eGFRcr) may contribute to its misuse. We investigated the impact of cystatin c estimates of GFR (eGFRcys) on metformin eligibility.MethodsIn a consecutive cohort of 550 Veterans with diabetes, metformin use and eligibility were assessed by eGFR category, using eGFRcr and eGFRcys. Discrepancy in eligibility was defined as cases where eGFRcr and eGFRcys categories (<30, 30-44, 45-60, and >60 ml/min/1.73 m2) differed with an absolute difference in eGFR of >5 ml/min/1.73 m2. We modeled predictors of metformin use and eGFR category discrepancy with multivariable relative risk regression and multinomial logistic regression.ResultsSubjects were 95% male, median age 68, and racially diverse (45% White, 22% Black, 11% Asian, 22% unknown). Metformin use decreased with severity of eGFRcr category, from 63% in eGFRcr >60 to 3% in eGFRcr <30. eGFRcys reclassified 20% of Veterans into different eGFR categories. Factors associated with a more severe eGFRcys category compared to eGFRcr were older age (aOR = 2.21 per decade, 1.44-1.82), higher BMI (aOR = 1.04 per kg/m2, 1.01-1.08) and albuminuria >30 mg/g (aOR = 1.81, 1.20-2.73).ConclusionsMetformin use is low among Veterans with CKD. eGFRcys may serve as a confirmatory estimate of kidney function to allow safe use of metformin among patients with CKD, particularly among older individuals and those with albuminuria

    The balance between fumarate and malate plays an important role in plant development and postharvest quality in tomato fruit

    Organic acids, produced as intermediates of the tricarboxylic cycle, play a crucial role in the plant primary metabolism and are considered as being ones of the most important quality traits in edible fruits. Even if they are key metabolites in a multitude of cellular functions, little is known about their physiological relevance and regulation. Transgenic tomato (Solanum lycopersicum) plants expressing constitutively a bacterial maleate isomerase, which converts reversibly maleate to fumarate, were generated in order to improve our knowledge about the role of organic acids in the crop and fruit metabolism. Growth and reproduction were affected by the unbalance of tricarboxylic cycle intermediates, as a dwarf phenotype and a flowering delay were observed in the transgenic plants. In addition, a delay in chlorophyll synthesis, a decrease in the numbers of stomata and significant changes in some photosynthetic parameters indicated alterations in central primary metabolism. Postharvest was also impaired, as transgenic fruits showed increased water lost and deterioration, indicating a possible role of the organic acids in cell wall metabolism. Finally, preliminary metabolomics analysis pointed out important changes during fruit ripening in flavor-related metabolites, such as acids and sugars, revealing the importance of organic acids in fruit metabolism. Taken together, these data indicate a pivotal role of tricarboxylic cycle intermediates, such as malate or fumarate, as regulatory metabolites. Here we present the metabolomics profiling for the content of primary and secondary metabolites of a 95 F1 individuals strawberry population derived from genotype “1392”, selected for its superior flavour, and “232” (Zorrilla-Fontanesi et al., 2011; Zorrilla-Fontanesi et al., 2012).     Bacterial pathogenesis of plants: Future challenges from a microbial perspective

    Plant infection is a complicated process. Upon encountering a plant, pathogenic microorganisms must first adapt to life on the epiphytic surface, and survive long enough to initiate an infection. Responsiveness to the environment is critical throughout infection, with intracellular and community-level signal transduction pathways integrating environmental signals and triggering appropriate responses in the bacterial population. Ultimately, phytopathogens must migrate from the epiphytic surface into the plant tissue using motility and chemotaxis pathways. This migration is coupled to overcoming the physical and chemical barriers to entry into the plant apoplast. Once inside the plant, bacteria use an array of secretion systems to release phytotoxins and protein effectors that fulfil diverse pathogenic functions (Fig. 1)(Phan Tran et al., 2011, Melotto & Kunkel, 2013)

    Cardiovascular and metabolic responses to the ingestion of caffeinated herbal tea: drink it hot or cold?

    Tea is usually consumed at two temperatures (as hot tea or as iced tea). However, the importance of drink temperature on the cardiovascular system and on metabolism has not been thoroughly investigated. The purpose of this study was to compare the cardiovascular, metabolic and cutaneous responses to the ingestion of caffeinated herbal tea (Yerba Mate) at cold or hot temperature in healthy young subjects. We hypothesized that ingestion of cold tea induces a higher increase in energy expenditure than hot tea without eliciting any negative effects on the cardiovascular system.Methods: Cardiovascular, metabolic and cutaneous responses were analyzed in 23 healthy subjects (12 men and 11 women) sitting comfortably during a 30-min baseline and 90 min following the ingestion of 500 mL of an unsweetened Yerba Mate tea ingested over 5 min either at cold (~3°C) or hot (~55°C) temperature, according to a randomized cross-over design.Results: Averaged over the 90 min post-drink ingestion and compared to hot tea, cold tea induced (1) a decrease in heart rate (cold tea: −5 ± 1 beats.min−1; hot tea: −1 ± 1 beats.min−1, p < 0.05), double product, skin blood flow and hand temperature and (2) an increase in baroreflex sensitivity, fat oxidation and energy expenditure (cold tea: +8.3%; hot tea: +3.7%, p < 0.05). Averaged over the 90 min post-drink ingestion, we observed no differences of tea temperature on cardiac output work and mean blood pressure responses.Conclusion: Ingestion of an unsweetened caffeinated herbal tea at cold temperature induced a greater stimulation of thermogenesis and fat oxidation than hot tea while decreasing cardiac load as suggested by the decrease in the double product. Further experiments are needed to evaluate the clinical impact of unsweetened caffeinated herbal tea at a cold temperature for weight control

    Spitzer Mid-to-Far-Infrared Flux Densities of Distant Galaxies

    We study the infrared (IR) properties of high-redshift galaxies using deep Spitzer 24, 70, and 160 micron data. Our primary interest is to improve the constraints on the total IR luminosities, L(IR), of these galaxies. We combine the Spitzer data in the southern Extended Chandra Deep Field with a K-band-selected galaxy sample and photometric redshifts from the Multiwavelength Survey by Yale-Chile. We used a stacking analysis to measure the average 70 and 160 micron flux densities of 1.5 < z < 2.5 galaxies as a function of 24 micron flux density, X-ray activity, and rest-frame near-IR color. Galaxies with 1.5 < z < 2.5 and S(24)=53-250 micro-Jy have L(IR) derived from their average 24-160 micron flux densities within factors of 2-3 of those derived from the 24 micron flux densities only. However, L(IR) derived from the average 24-160 micron flux densities for galaxies with S(24) > 250 micro-Jy and 1.5 < z < 2.5 are lower than those derived using only the 24 micron flux density by factors of 2-10. Galaxies with S(24) > 250 micro-Jy have S(70)/S(24) flux ratios comparable to sources with X-ray detections or red rest-frame IR colors, suggesting that warm dust possibly heated by AGN may contribute to the high 24 micron emission. Based on the average 24-160 micron flux densities, nearly all 24 micron-selected galaxies at 1.5 < z < 2.5 have L(IR) < 6 x 10^12 solar luminosities, which if attributed to star formation corresponds to < 1000 solar masses per year. This suggests that high redshift galaxies may have similar star formation efficiencies and feedback processes as local analogs. Objects with L(IR) > 6 x 10^12 solar luminosities are quite rare, with a surface density ~ 30 +/- 10 per sq. deg, corresponding to ~ 2 +/- 1 x 10^-6 Mpc^-3 over 1.5 < z < 2.5.Comment: Accepted for Publication in ApJ.     Evaluating diverse electronic consultation programs with a common framework.

    BackgroundElectronic consultation is an emerging mode of specialty care delivery that allows primary care providers and their patients to obtain specialist expertise without an in-person visit. While studies of individual programs have demonstrated benefits related to timely access to specialty care, electronic consultation programs have not achieved widespread use in the United States. The lack of common evaluation metrics across health systems and concerns related to the generalizability of existing evaluation efforts may be hampering further growth. We sought to identify gaps in knowledge related to the implementation of electronic consultation programs and develop a set of shared evaluation measures to promote further diffusion.MethodsUsing a case study approach, we apply the Reach, Effectiveness, Adoption, Implementation and Maintenance (RE-AIM) and the Quadruple Aim frameworks of evaluation to examine electronic consultation implementation across diverse delivery systems. Data are from 4 early adopter healthcare delivery systems (San Francisco Health Network, Mayo Clinic, Veterans Administration, Champlain Local Health Integration Network) that represent varied organizational structures, care for different patient populations, and have well-established multi-specialty electronic consultation programs. Data sources include published and unpublished quantitative data from each electronic consultation database and qualitative data from systems' end-users.ResultsOrganizational drivers of electronic consultation implementation were similar across the systems (challenges with timely and/or efficient access to specialty care), though unique system-level facilitators and barriers influenced reach, adoption and design. Effectiveness of implementation was consistent, with improved patient access to timely, perceived high-quality specialty expertise with few negative consequences, garnering high satisfaction among end-users. Data about patient-specific clinical outcomes are lacking, as are policies that provide guidance on the legal implications of electronic consultation and ideal remuneration strategies.ConclusionA core set of effectiveness and implementation metrics rooted in the Quadruple Aim may promote data-driven improvements and further diffusion of successful electronic consultation programs
