Fogak morfológiai és metrikus jellegzetességeinek vizsgálata marosvásárhelyi középkori leleteken és a jelenkori helyi populáció körében = Dental morphological and metric characteristics in contemporary and historical population of Târgu Mureș
Absztrakt:
Bevezetés: A fogazat a szervezet egységes részeként nem képez
kivételt az embernek a fejlődéstani okokból és életformából következő fizikai
antropológiai változásai alól. A különböző káros stresszhatások mellett a
természetes kiválasztódás is hozzájárul bizonyos fogmorfológiai jellemzők
hangsúlyosabbá válásához. Célkitűzés: Középkori embertani
leletek és mai páciensek gipszmintáin végzett odontometriai elemzés
eredményeinek, valamint a járulékos csücskök jelenlétének összehasonlítása.
Anyag és módszer: A 19 embertani leletből származó 132,
valamint 30 páciens modelljén meglévő 694 fog morfológiai jellegzetességeit
figyeltük meg. A koronák mesiodistalis, incisivogingivalis és vestibulooralis
átmérőinek kétdimenziós képanalízise során nyert értékeit összehasonlítottuk, és
megvizsgáltuk az antropológiai szempontból kiemelt fontosságú Carabelli- és
taloncsücsköket. Eredmények: A XIV. századi és a jelenkori
fogak méretei között statisztikai szempontból jelentős változást találtunk
néhány fogcsoport esetében. A középkori leleteken az oldalsó fogak kevésbé
széles koronával rendelkeztek, mint a mai populációban mért fogak. A történeti
leleteken talált Carabelli-csücskök a Dahlberg-skála szerinti 1. és 3.
fokozathoz tartoztak (23,07%-os előfordulás), míg a mai eseteken megfigyelt
Carabelli-csücskök a 3., 4., 5., 6. és 7. fokozathoz tartoztak (50,90%-os
előfordulás). Taloncsücsökkel a jelenkori csoportban találkoztunk, mindössze egy
esetben. Következtetés: A vizsgált jelenkori fogakon a
Carabelli-csücskök hangsúlyosabb morfológiai formában és nagyobb gyakorisággal
jelentek meg, ami a mai populáció europid jellegét hangsúlyozza. Orv Hetil.

Abstract:
Introduction: Teeth, as an integral part of human organism, are
not exceptions from the physical anthropology changes caused by biological
evolution and the way of living. Different stress factors and the natural
selection are contributing to the emphasis of certain morphological
characteristics. Aim: Comparison of dental metric
characteristics and the presence of accessory tooth cusps between human remains
from the Middle Ages and dental study models of today’s patients.
Material and method: Morphological characteristics of 132
teeth from 19 skulls and 694 teeth on dental models of 30 patients were
examined. The mesiodistal, incisivocervical and buccolingual diameters of crowns
were measured by a validated 2D image analysis method. Carabelli and talon cusps
were also examined using a magnifying glass. Results:
Statistically significant differences between the size of the 14th-century and
the present-day teeth were found in some of the teeth groups. In medieval
artifacts, lateral teeth had smaller crown width than teeth measured in the
contemporary population. The Carabelli cusps found in the archeological human
remains belonged to grades 1 and 3 on Dahlberg scale (23.07%), while the
Carabelli cusps observed in the contemporary group belonged to grades 3, 4, 5, 6
and 7 (50.90%). Talon cusp occurred only once in the contemporary group.
Conclusion: In case of the contemporary teeth, the
Carabelli cusps appeared in a more pronounced morphological form and with a
higher frequency, which emphasizes the European origin of today’s population.

Fehérjék konformációs dinamikája mint a biomolekuláris felismerés és jelátvitel meghatározó eleme = Protein conformational dynamics as a key determinant in biomolecular recognition and signal transmission
A térszerkezet alapján, a konformációs dinamika figyelembevételével kíséreltük meg az intramolekuláris és molekulák közötti jelátvitel megértését atomi felbontással. Kísérleti objektumok: a komplement rendszer, azon belül is a nemrég felfedezett lektin út fehérjekomplexei, a flagelláris exportrendszer valamint moduláris monomer, dimer és oligomer felépítésű enzimek álltak. Megállapítottuk, hogy FliI ATPáz, amely képes az exportálandó fehérjéje kitekerésére, a FliJ, FliH és FliS komponensekkel együtt képez olyan szupramolekuláris komplexet, amely képes az export szubsztrátumok felismerésére. Leírtuk a foszfoglicerát kináz enzim alloszterikus működési mechanizmusát, atomi felbontással. Feltártuk az izopropilmalát dehidrogenáz molekuláris csuklóinak működését és szerepét az alegységek kölcsönhatásaiban. Szelektív inhibitorokkal a tankönyvi tézissel ellentétes felismerésre jutottunk, miszerint a komplement rendszer lektin útjának meghatározó aktivátora a MASP-1 szerin proteáz. Így a komplement aktiválással összefüggő betegségek új gyógyszercélpont molekuláját azonosítottuk. Felfedeztük, hogy a MASP-1 képes a kininogén hasítása útján, bradikinint felszabadítva, komplement függő gyulladást keltésére. Felfedeztük, hogy a trombinhoz hasonlóan a MASP-1, PAR-4 receptoron keresztül endotél sejteket aktivál. Bizonyítékot találtunk arra, hogy a fehérjék konformációs dinamikája meghatározza a szerkezet evolúciójának lehetséges irányait, több milliárd éves időskálán is. | The CUB2 domain of C1r without calcium has disordered structure. This flexibility, necessary for autocativation of C1r inside the C1 complex, is regulated by calcium. Using MASP-selective inhibitors we proved that, in contrast to the previous textbook picture, MASP-1 is the exclusive activator of MASP-2. Blocking the proteolytic activity of MASP-1 prevents activation of the lectin pathway, therefore MASP-1 is a new target in treating complement related diseases. We solved the structure of the catalytic region of MASP-1. The structure explains the special enzymatic characteristics of this complement protease. We discovered a new, inflammation related function of the complement system: MASP-1 is able to directly activate endothelial cells through cleaving protease activated receptor-4. We discovered that MASP-1 is able to cleave kininogen and liberates bradykinin. In this way MASP-1 can contribute to the local inflammatory reaction triggered by complement activation. The allosteric mechanismnof human PGK has been explored at atomic details. In the dimeric enzyme IPMDH structural and site-directed mutagenesis studies revealed the operation of the two main molecular hinges and their relationship with the subunit interactions. We have shown that conformational motions are linked to protein evolution by producing structural variants that can be evolutionarily stabilized. Identification of ?-lactamases in human and bovine isolates of Staphylococcus aureus strains having borderline resistance to penicillinase-resistant penicillins (PRPs) with proteomic methods
Methicillin and oxacillin hydrolyzing enzymes of 6 borderline methicillin-resistant and 1 methicillin-resistant Staphylococcus aureus strains isolated from human clinical samples and 4 borderline methicillin-resistant S. aureus strains isolated from bovine mastitis were investigated. As previous studies suggested the involvement of an additional enzyme besides the penicillinase BlaZ in the determination of borderline resistance, we analyzed the expressed extracellular and membrane-bound ß-lactamases with 2-D gel electrophoresis and mass spectrometry. Our analysis showed that the penicillin-hydrolyzing BlaZ alone was responsible for the hydrolysis of both methicillin and oxacillin. All supernatant and membrane fractions contained the same enzyme with slight sequence variations. The size and pI of the proteins were also variable, probably due to spontaneous hydrolysis and/or post-translational modifications. Interestingly, we found also cytotoxins and other virulence factors in some nitrocefin hydrolyzing dots, suggesting that those proteins might have a role in the reduction of local antibiotic concentratio
Y-chromosome haplogroups from Hun, Avar and conquering Hungarian period nomadic people of the Carpathian Basin
Hun, Avar and conquering Hungarian nomadic groups arrived to the Carpathian Basin from the Eurasian Steppes and significantly influenced its political and ethnical landscape, however their origin remains largely unknown. In order to shed light on the genetic affinity of above groups we have determined Y chromosomal haplogroups and autosomal loci, suitable to predict biogeographic ancestry, from 49 individuals, supposed to represent the power/military elit. Haplogroups from the Hun-age are consistent with Xiongnu ancestry of European Huns. Most of the Avar-age individuals carry east Eurasian Y haplogroups typical for modern north-eastern Siberian and Buryat populations and their autosomal loci indicate mostly un-admixed Asian characteristics. In contrast the conquering Hungarians seem to be a recently assembled population incorporating un-admixed European, Asian as well as admixed components. Their heterogeneous paternal and maternal lineages indicate similar supposed phylogeographic origin of males and females, derived from Central-Inner Asian and European Pontic Steppe sources
