Studies on Regioselective Binding Mode of Steroid Molecules in Homology Modeled Cytochrome P450-2C11
In this study, we investigated the regioselective binding mode of steroid molecules and structure requirements for steroid molecules for 16[alpha]-hydroxylation by Cytochrome P450-2C11. Docking study by using the homology Cytochrome P450-2C11 indicated that 16[alpha]-hydroxylation is favored with steroidal molecules possessing the following components, 1) a bent A-B ring configuration (5[beta]-reduced), 2) C-3[alpha]-hydroxyl group, 3) C-17[beta]-acetyl group, and 4) methyl group at both the C-18 and C-19. These respective steroid components requirements such as A-B ring configuration and functional groups at C-3 and C-17 were defined as the inhibitory contribution factor. Overall results by rat CYP2C11 revealed that steroidal structure requirements resulted in causing an effective inhibition of [^3^H]progesterone 16[alpha]-hydroxylation by the adult male rat liver microsome. As far as docking of homology modeled CYP2C11 against investigated steroids is concerned, they are docked at the active site superimposed with flurbiprofen. It was also found that the distance between heme iron and C16[alpha]-H was between 4 to 6 Å and that the related angle was in the range of 180±45°
Delta Effects in Pion-Nucleon Scattering and the Strength of the Two-Pion-Exchange Three-Nucleon Interaction
We consider the relationship between P-wave pi-N scattering and the strength
of the P-wave two-pion-exchange three-nucleon interaction (TPE3NI). We explain
why effective theories that do not contain the delta resonance as an explicit
degree of freedom tend to overestimate the strength of the TPE3NI. The
overestimation can be remedied by higher-order terms in these ``delta-less''
theories, but such terms are not yet included in state-of-the-art chiral EFT
calculations of the nuclear force. This suggests that these calculations can
only predict the strength of the TPE3NI to an accuracy of +/-25%.Comment: 13 pages, 2 figures, uses eps
Transport properties and electronic states of the thermoelectric oxide NaCo_2O_4
Physical properties of the thermoelectric oxide NaCo_2O_4 are briefly
reviewed. The high thermoelectric properties of this material are attributed to
the substantially enhanced effective mass, which comes from the large entropy
of Co in the low spin state. The large entropy confined in the CoO_2
block causes a spin-density-wave transition at 22 K upon Cu substitution for

Finite size corrections in massive Thirring model
We calculate for the first time the finite size corrections in the massive
Thirring model. This is done by numerically solving the equations of periodic
boundary conditions of the Bethe ansatz solution. It is found that the
corresponding central charge extracted from the term is around 0.4 for
the coupling constant of and decreases down to zero when
. This is quite different from the predicted central

Robustness evaluation of seismic pile response considering uncertainty mechanism of soil properties
Robustness analysis of the seismic pile response of a structureâpileâsoil system with uncertain soil properties and concrete Youngâs modulus is presented in this paper. The bounds of the bending moment of a pile are investigated by means of the previously proposed uncertainty analysis method (Updated Reference-Point method) and the newly developed revised method (NURP method). An efficient finite-element model of a structureâpileâsoil system with smart displacement functions for connecting elements is adopted and a response spectrum method is applied in the evaluation of the seismic pile responses of the system. Two cases of soil uncertainties resulting from different uncertainty mechanisms are considered. It is shown that the worst combination of uncertain soil parameters can be determined by the NURP method in an accurate manner
A possible involvement of ion transporter in tumor necrosis factor alpha and cycloheximide-induced apoptosis of endothelial cells.
We examined the tumor necrosis factor alpha (TNFalpha)-induced apoptosis of vascular endothelial cells from the standpoint of ion channels. Cultured vascular endothelial cells from bovine carotid artery were used. Apoptosis was determined by a propidium iodide assay. Treatment of the endothelial cells with TNFalpha and cycloheximide for 6 h induced nuclear fragmentation in a TNFalpha dose-dependent manner (1-10 ng/ml). Concomitant treatment of endothelial cells with TNFalpha at a dose of 10 ng/ml and cycloheximide at a dose of 10 microg/ml elicited endothelial cell apoptosis as high as 23.4+/-4.1% at 6 h after administration. However, 10 ng/ml TNFalpha alone elicited a little apoptosis at 6 h after its administration (% apoptosis=4.1+/-0.8%). Cycloheximide (10 microg/ml) did not induce apoptosis at all. Concomitant treatment of endothelial cells with 1 mmol/l of 4,4-diisothiocyanatostilbene-2,2-disulfonic acid, which is a chloride bicarbonate exchanger blocker, partially inhibited the TNFalpha and cycloheximide-induced endothelial cell apoptosis. On the other hand, endothelial cell apoptosis due to TNFalpha and cycloheximide was completely inhibited by benzyloxycarbonyl-Asp-CH2OC(O)-2,6-dichlorobenzene (50 micromol/l), an inhibitor of caspase. Moreover, pyrrolidine dithiocarbanate, an inhibitor of nuclear factor kappa B (NF-kappaB), also suppressed endothelial cell apoptosis induced by TNFalpha and cycloheximide completely. These findings suggest that the endothelial cell apoptosis induced by TNFalpha and cycloheximide is closely related to not only chloride ions, but also both NF-kappaB and caspase activation. That is to say, there is a possibility that chloride ions or bicarbonate (pH) may play an important role in signal transduction such as NF-kappaB and caspase activation in the apoptosis induced by TNFalpha and cycloheximide
